현재까지 LT-HSC와 ST-HSC를 구별하는 자가 재생 능력의 분자 메커니즘은 잘 알려져 있지 않습니다. 자가 재생 능력에 따라 두 세포 집단을 분리하고 기본 분자 메커니즘을 발견하는 것이 중요합니다. LT-HSC13,14,15를 정제하기 위해 여러 리포터 시스템도 도입되었습니다. 그러나 각 리포터 시스템에서 정의한 LT-HSC 순도는 가변적이며 현재까지 독점적인 LT-HSC 정제가 달성되지 않았습니다.
따라서 LT-HSC 및 ST-HSC에 대한 격리 시스템을 개발하면 pHSC 부분의 자가 재생 용량에 관한 연구가 가속화될 것입니다. LT-HSC와 ST-HSC의 분리에서 다단계 편향되지 않은 스크리닝을 사용한 연구에서는 pHSC 분획16에서 이질적으로 발현되는 단일 유전자 Hoxb5를 확인했습니다. 또한, Hoxb5 리포터 마우스의 골수 분석은 pHSC 분획의 약 20%-25%가 Hoxb5 pos 세포로 구성되어 있음을 밝혀냈다. Hoxb5pos pHSC 및 Hoxb5 neg pHSC를 사용한 경쟁적 이식 분석에서 Hoxb5pos pHSC만이 장기 자가 재생 능력을 갖는 반면 Hoxb5neg pHSC는 단기간 내에 자가 재생 능력을 상실하는 것으로 나타났으며, 이는 Hoxb5가 pHSC 분획16에서 LT-HSC를 식별함을 나타냅니다.
