Sommige eencellige of koloniale rode algen, zoals Chroothece, groeien langzaam in vitro, maar bevatten meerdere autofluorescente verbindingen die kunnen worden geanalyseerd door spectrale analyse onder een confocale microscoop, waar verschillen in pigmentemissiepieken kunnen worden gedetecteerd. Spectrale confocale fluorescentiemicroscopie heeft ons in staat gesteld om in vivo studies uit te voeren om de aanpassing of acclimatisatie van fotosynthetische organismen te evalueren 8,10,17,18,19,20....

Rode algen, zoals het geslacht Chroothece, kunnen groeien in extreme habitats, waar ze vaak te maken hebben met duidelijke veranderingen in het milieu1. Overstromingen en droogtes komen vaak voor in semi-aride gebieden waar dit geslacht te vinden is, en sommige soorten zijn gemeld in kreken, kliffen, grotten of zelfs thermaal water2. Meestal degraderen biologische variabelen, zoals competitie of begrazing, soorten echter naar niet-optimale omstandigheden voor hun groei. Omdat deze organismen vaak moeilijk te kweken zijn en niet of zeer langzaam groeien onder laboratoriumomstandigheden, is een belangrijke beperking de beschikbare steekproefgrootte. Daarom is het erg belangrijk om niet-destructieve methoden of methoden te volgen die minimale monstermanipulatie inhouden 3,4.
De fysiologische vaardigheden die nodig zijn om te overleven in deze ruwe omgevingen kunnen worden gecontroleerd door veranderingen in hun fotosynthetische systemen te volgen. Metabole mechanismen, fotosynthetische efficiëntie en gevoeligheid voor licht of kweekomstandigheden kunnen worden onthuld door pigmentfluorescentie-emissieprofielen, als gevolg van nauwkeurige veranderingen in hun energieoverdracht of het vangenvan 5,6,7,8.
Autofluorescentie van cellulaire verbindingen kan worden gebruikt als een marker voor cytodiagnostiek of als een natuurlijke indicator van cellulaire toestand of metabolisme in reactie op externe en interne signalen door veranderingen in emissie9. Het kan ook worden gebruikt om taxonomisch verschillende groepen fotosynthetische organismen te onderscheiden10. Afhankelijk van de fylogenetische positie van fototrofe micro-organismen, kan men verschillende in vivo fluorescentiekenmerken vinden. Daarom is bij verschillende gelegenheden geprobeerd een taxonomische identificatie te maken op basis van de in vivo kenmerken van fototrofe fluorescentie (met inbegrip van fluorescentieabsorptie en emissiespectra)11,12. Vanwege de diversiteit in accessoire pigmenten tussen fytoplanktontaxa, kunnen verschillen in de golflengten waarbij chlorofyl a (Chl a) fluorescentie wordt gestimuleerd, of verschillen in emissiespectra, worden gebruikt om taxonomie13 af te leiden. De in vivo fluorescentie-excitatie en emissiespectra van deze exemplaren zijn niet alleen afhankelijk van de phyla van algen, maar ook van fotosysteemaanpassing14. De efficiëntie van energieoverdracht naar Chl a, of de verhouding van Chl a tot accessoire pigmenten, en het cellulaire pigmentgehalte zijn gevoelig voor groeiomstandigheden5.
Rode algen, met name Chroothece, hebben verschillende accessoire fluorescerende pigmenten-phycobiliproteïnen en carotenoïden; Het eerste concentraat in fycobilisomen gehecht aan de thylakoïden van chloroplasten. Phycobiliproteïnen (phycocyanine, phycoerythrin en allophycocyanine) kunnen licht op verschillende golflengten opvangen en doorgeven aan Chl a, wat fotosynthese in zeer verschillende licht- en kweekomstandigheden mogelijk maakt15. Chroothece-soorten kunnen bijvoorbeeld in grotten groeien of bijna opduiken in licht zoute kalkhoudende beken2.
Monochromatische lichten beïnvloeden de groei en pigmentsamenstelling van fotosynthetische organismen en zijn bestudeerd om de groei van fotosynthetische organismen in grotten te voorkomen of te beheersen. Mulec et al. toonden aan dat rood verrijkte verlichting de groei van cyanobacteriën, algen en planten bevordert16. Eerdere studies hebben ook gemeld dat groen licht de pigmentsamenstelling van cyanobacteriënbeïnvloedt 17, terwijl anderen hebben aangetoond dat groen licht de groei van de meeste fotosynthetische organismen voorkomt en sommige cyanobacteriën vertonen een vermindering van thylakoïden en een zwakkere gemiddelde fluorescentie-intensiteit18.
Om het vermogen van Chroothece als modelorganisme om zware omstandigheden te overwinnen te begrijpen, zijn gekweekte cellen blootgesteld aan toenemende lichtintensiteiten en monochroom licht (groen of rood)15, om te zien hoe het omgaat met de schemerige omstandigheden van grotten (waar rood licht overheerst). Het hierin gepresenteerde protocol reproduceert het effect van de bovengenoemde variabelen op Chroothece's phycobiliproteïnen op cellulair niveau met behulp van zijn eigen autofluorescentie.
Tegenwoordig wordt fluorescentie vaak gebruikt als een hulpmiddel om de fysiologische reacties van vaatplanten, microalgen, macroalgen en cyanobacteriën te bestuderen13,14,16. Spectrale confocale fluorescentiemicroscopie is een uitstekend hulpmiddel voor in vivo studies om de fysiologie van fotosynthetische monsters op eencellig niveau 10,17,18,19,20 te evalueren, door problemen in verband met de lage groeisnelheid in het laboratorium en de moeilijkheden bij het verkrijgen van voldoende biomassa voor de bijbehorende extractie en biochemische methodente vermijden 8 . Zodra cellen gedurende 2 weken onder verschillende kweekomstandigheden zijn behandeld, kan het lambdasscanprofiel in vivo worden gemeten. Hoewel er verschillende publicaties zijn waarin verschillende golflengten van excitatie door confocale beeldvorming zijn gebruikt 3,4,10,17, kunnen de meeste fycobiliproteïnen en Chl a worden gedetecteerd met behulp van een excitatielijn met een golflengte van 561 nm en de gedetecteerde emissie varieert van 570 tot 760 nm golflengte. Deze criteria zijn gebaseerd op een analyse die eerder is uitgevoerd 10 met commerciële zuivere pigmenten (tabel 1) door confocale beeldvorming en de verkregen resultaten bij verschillende algensoorten20,21,22.

Deze tabel toont emissiepieken en schouders/fluorescentiebandmaxima van verschillende fluorochromen/pigmenten door confocale beeldvormingsspectrofotometrie voor alle excitatiegolflengten, en het percentage lichtemissie door pigmenten/fluorochromen. De waarden werden berekend met de formule: = MFI*100/255. Elke waarde is het gemiddelde ± SE (gemiddelde ± standaardfout ten opzichte van het gemiddelde). Zuivere pigmenten werden gebruikt voor het kalibreren van de confocale scanning laser microscoop als volgt 1,2,10. Chlorofyl a werd verkregen uit Spinacia oleracea, R-phycoerythrin (R-PE) uit Porphyra tenera en C-phycocyanine (C-PE) uit Spirulina sp. Alle soorten werden opgelost in gefilterd gedestilleerd water. Allophycocyanine-XL (APC-XL) werd verkregen uit Mastigocladus laminosus, dat werd opgelost in ammoniumsulfaat (60%) en kaliumfosfaat (pH = 7) om een concentratie van 38 mM te bereiken. De scans werden uitgevoerd met 400 μL van elke pigmentoplossing (concentratie van 1 mg / ml) met behulp van een 8-goed bedekte glazen bodemkamer.
De studie van een enkele excitatiegolflengte is een vrij nuttige eerste benadering. In dit geval is het echter noodzakelijk om de relatieve bijdrage van de verschillende complexen in het fluorescentiesignaal op te helderen, wat wordt aanbevolen om onder andere een fluorescentieverhouding of spectrumanalyse op verschillende golflengten uit te voeren.

Rode algen (Rhodophyta) bevatten fycobiliproteïnen en koloniseren habitats met weinig licht, maar sommige (bijvoorbeeld sommige Chroothece-soorten ) kunnen zich ook in de volle zon ontwikkelen. De meeste rhodofyten zijn rood, maar sommige kunnen blauwachtig lijken, afhankelijk van de verhouding blauwe en rode biliproteïnen (phycocyanine en phycoerythrin). Verschillende fycobiliproteïnen kunnen licht op verschillende golflengten opvangen en doorgeven aan chlorofyl a, wat fotosynthese onder zeer verschillende lichtomstandigheden mogelijk maakt. Deze pigmenten reageren op habitatveranderingen in licht en hun autofluorescentie kan helpen bij het bestuderen van biologische processen. Met behulp van Chroothece mobilis als modelorganisme en de spectrale lambda-scanmodus in een confocale microscoop, werd de aanpassing van fotosynthetische pigmenten aan verschillende monochromatische lichten op cellulair niveau bestudeerd om de optimale groeiomstandigheden van de soort te raden. De resultaten toonden aan dat, zelfs wanneer de bestudeerde soort uit een grot werd geïsoleerd, deze zich aanpaste aan zowel zwakke als gemiddelde lichtintensiteiten. De gepresenteerde methode is vooral nuttig voor het bestuderen van fotosynthetische organismen die niet of zeer langzaam groeien onder laboratoriumomstandigheden, wat meestal het geval is voor mensen die in extreme habitats leven.

Chlorofyl a absorbeert over het algemeen blauwe en rode golflengten van zichtbaar licht, terwijl fycobiliproteïnen groene, gele en oranje golflengten gebruiken7. De autofluorescentie van deze pigmenten maakt de eerste benadering om fycobiliproteïnen en chlorofylgedrag onder experimentele en veldomstandigheden te bestuderen mogelijk.

Autofluorescence Imaging to Evaluate Red Algae Physiology

Het huidige protocol beschrijft stapsgewijze autofluorescentiebeeldvorming en evaluatie van fycobiliproteïneveranderingen in rode algen op basis van spectrale analyse. Dit is een labelvrije en niet-destructieve methode om cellulaire aanpassing aan extreme habitats te evalueren, wanneer alleen schaars materiaal beschikbaar is en cellen langzaam of helemaal niet groeien onder laboratoriumomstandigheden.

Dit confocale microscopische protocol zal de interpretatie van het ecologische gedrag en de aanpassingen van microalgen uit extreme omgevingen met langzame groei in het laboratorium mogelijk maken met behulp van autofluorescentie van hun pigmenten. Deze techniek is niet invasief en minimaliseert artefacten omdat chemische verbindingen niet worden gebruikt. Het kennen van de prestaties van pigmenten van autotrofen en de bijbehorende groei zal het ontwerpen van de controlemethoden mogelijk maken die het grootste belang hebben voor toeristische grotten en architecturale monumenten.Begin met het bereiden van het entmateriaal van Chroothece mobilis door het over te brengen van de agarcultuur naar het zeewatermedium of swes vloeibaar medium. Houd alle culturen gedurende twee weken met een 16 tot 8 licht donkere fotoperiode bij 20 graden Celsius onder een lage witte lichtintensiteit en zonder schudden totdat de gewenste celdichtheid is verkregen. Gebruik één milliliter van de exponentiële fasecultuur met vijf keer 10 tot de derde cellen per milliliter celdichtheid om te enten in een 24-putplaat voor verschillende experimenten.Om het monochromatische lichteffect te reproduceren, gebruikt u het groene filter dat groen licht doorlaat van 470 tot 570 nanometer met een piek op de golflengte van 506 nanometer en rood licht aangepast met behulp van een filter tussen 590 en 720 nanometer en piekt op 678 nanometer. Stel de celculturen gedurende twee weken bloot aan dit licht. Schakel alle componenten van de omgekeerde confocale laserscanmicroscoop in, inclusief de laser.Monteer de cellen in het SWES-groeimedium van elke experimentele put van de 24-putplaat op een glazen bodemschaal van 35 millimeter voor beeldvorming. Kies het 63X of 1.30 numerieke diafragma of NA glycerol onderdompelingsobjectief en plaats glycerol over de lens. Plaats vervolgens de putplaat op het microscooppodium en zorg ervoor dat het monster niet beweegt tijdens de beeldacquisitie.Centreer het monster in het lichtpad en focus op het midden van de cel door het vlak met de hoogste fluorescentie-intensiteit te selecteren. Als u klaar bent, opent u de beeldacquisitiesoftware en kiest u XY lambda in de vervolgkeuzelijst in de acquisitiemodus. Selecteer de excitatielijn van de laser 561 nanometer, acht bits dynamisch bereik en 1024 bij 1024 pixels.Verzamel de fluorescentie-emissiespectra in de bandbreedte van 10 nanometer en de lambdastapgrootte van vier nanometer binnen het bereik van 570 tot 760 nanometer. Zet het gaatje op één luchtunit en voer de lambdascan uit. Nadat u dit proces in verschillende gezichtsvelden en onder de verschillende omstandigheden van rode en groene lichten hebt herhaald, slaat u alle gegevens op.Zodra de lambdascan is verkregen, klikt u op het kwantificeervenster bovenaan de software om de verzamelde fluorescentie-emissiespectra te evalueren. Ga naar het geopende projectvenster en selecteer één XY lambda-bestand. Selecteer gestapelde profielanalyse in de beeldvormingssoftware en definieer een interessegebied van vier micrometer vierkant in het midden van een cel om de gemiddelde fluorescentie-intensiteit te analyseren.Exporteer de gegevens en het CSV-bestand voordat u het proces herhaalt met verschillende cellen onder verschillende omstandigheden. Open vervolgens de CSV-bestanden om de verschillende fluorescentie-emissiepieken van alle gemeten interessegebieden te selecteren door de maximale fluorescentiegegevens van respectievelijk fycoerythrin phycocyanobilin, C.phycocyanine, allophycocyanine en chlorofyl A in het CSV-bestand te selecteren. Als u klaar bent, maakt u een nieuwe tabel met alle maximale fluorescentiewaarden die zijn verkregen uit elke fycobiliproteïne- en chlorofylpiek en plot u de gegevens in een grafiek.Er werden significante verschillen in gemiddelde fluorescentie-intensiteit waargenomen. De gemiddelde fluorescentie-intensiteit van phycoerythrin phycocyanobilin nam aanzienlijk af wanneer de cellen werden blootgesteld aan monochroom groen licht. Daarentegen werd geen verschil waargenomen in rood licht in vergelijking met de controle.Het groene licht had het tegenovergestelde effect op allophycocyanine en chlorofyl A, waarbij de gemiddelde fluorescentie-intensiteit aanzienlijk toenam als gevolg van de aanpassing van antennecomplexen als gevolg van een verhoogde hoeveelheid of verbeterde connectiviteit van onder andere de antennecomplexen. Het rode licht produceerde een niet-significante toename van allophycocyanine en chlorofylfluorescentie. Om het effect van verschillende groeiomstandigheden op cellen in cultuur te bestuderen, is het cruciaal om de metingen uit te voeren met precies dezelfde microscoopinstellingen.Het is mogelijk om andere omgevingsparameters te analyseren die relevant zijn voor de cultuur van autofluorescentie-organismen, evenals het fluorescentiesignaal binnen het zichtbare spectrum. Deze techniek is een zeer krachtige benadering van een studie van levensorganismen met verschillende autofluorescentiepigmenten.
