Klinische studies geven aan dat hersenletsel het risico op psychiatrische stoornissen kan verhogen26,27. Bovendien beïnvloedt TBI de ontwikkeling van sociaal gedrag28,29. Intraspecies concurrentie treedt op wanneer leden van dezelfde soort tegelijkertijd strijden om een beperkte hulpbron1. Daarentegen vindt interspecies competitie plaats tussen leden van twee verschillende soorten2. Intraspecies concurrentie is verdeeld in twee soorten, waaronder interferentie (aangepast) en exploitatie (wedstrijd), en ontstaat afhankelijk van het type hulpbron in geschil, zoals voedsel en territorium3.
Het bestaan van sociale hiërarchieën is onmogelijk zonder dominant-onderdanige relaties (DSR's). Dominantie presenteert zich als "winnen" en ondergeschiktheid als "verliezen" binnen paren van dieren4. DSR's verschijnen echter niet alleen in paren, maar ook in groepen van drie of meer. In 1922 beschreef Thorleif Schjelderup-Ebbe de dominantiehiërarchie bij gedomesticeerde kippen. De belangrijkste onderscheidende tekenen tussen de dominante en ondergeschikte dieren waren tijd doorgebracht bij de feeder en agressief gedrag. De dominantiehiërarchie is verdeeld in twee vormen: lineair en niet-lineair5. Lineaire dominantie omvat twee groepen, A en B. In dit paradigma van transitieve relaties6 domineert groep A groep B, of groep B domineert groep A. Niet-lineaire dominantie treedt op wanneer er ten minste één cirkelvormige relatie is: A domineert B, B domineert C en C domineert A7.
Er bestaan modellen voor het beoordelen van dominant-onderdanig gedrag voor verschillende soorten, waaronder knaagdieren, vogels8, niet-menselijke primaten 9,10,11 en mensen 12. De dominant-onderdanige methode is goed vertegenwoordigd in de literatuur en is toegepast als een model om manie en depressie13 te beoordelen, evenals antidepressiva activiteit14. Dit model is gebruikt om stress in het vroege leven na moederscheiding bij volwassen ratten te onderzoeken15. De DSR-paradigma's kunnen worden onderverdeeld in drie modellen: de reductie van dominant gedrag model 13,16, de reductie van onderdanig gedrag model14 en de clonidine-omkering van dominantie model17.
Deze studie toont een onderzoek naar DSR door middel van taken op basis van voedselconcurrentie. De voordelen van deze methode zijn de eenvoudige reproduceerbaarheid en het vermogen om dominant-onderdanig gedrag te observeren en nauwkeurig te analyseren. Bovendien is de dominant-onderdanige gedragstaak afhankelijk van voedsel in plaats van territorium, in tegenstelling tot vergelijkbare gedragstaken, waardoor deze gedragstaak goedkoper en eenvoudiger is en onderzoekers geen ingewikkelde training hoeven te ondergaan om de taak uit te voeren en de gegevens te verwerken.
Het algemene doel van de huidige studie is om de ontwikkeling van DSR na traumatisch hersenletsel (TBI) aan te tonen. TBI wordt geassocieerd met sociale beperkingen, depressie en angst. Het model van het induceren van TBI is een eenvoudig en effectief standaardmodel waarbij traumatisch hersenletsel wordt geïnduceerd met een vloeistofpercussieapparaat18,19.

Concurrentie over hulpbronnen zoals voedsel, territorium en partners beïnvloedt relaties binnen diersoorten aanzienlijk en wordt gemedieerd door sociale hiërarchieën die vaak gebaseerd zijn op dominant-onderdanige relaties. De dominant-onderdanige relatie is een normaal gedragspatroon tussen de individuen van een soort. Traumatisch hersenletsel is een veel voorkomende oorzaak van sociale interactiestoornissen en de reorganisatie van dominant-onderdanige relaties in dierenparen. Dit protocol beschrijft onderdanig gedrag bij volwassen mannelijke Sprague-Dawley-ratten na de inductie van traumatisch hersenletsel met behulp van een vloeistofpercussiemodel in vergelijking met naïeve ratten door middel van een reeks dominant-onderdanige tests uitgevoerd tussen 29 dagen en 33 dagen na inductie. De dominant-onderdanige gedragstest laat zien hoe hersenletsel onderdanig gedrag kan veroorzaken bij dieren die concurreren om voedsel. Na traumatisch hersenletsel waren de knaagdieren meer onderdanig, zoals blijkt uit het feit dat ze minder tijd bij de feeder doorbrachten en minder snel als eerste bij de trog aankwamen in vergelijking met de controledieren. Volgens dit protocol ontwikkelt onderdanig gedrag zich na traumatisch hersenletsel bij volwassen mannelijke ratten.

Assessing Dominant-Submissive Behavior in Adult Rats Following Traumatic Brain Injury

Het huidige protocol beschrijft een rattenmodel van door vloeistofpercussie geïnduceerd traumatisch hersenletsel, gevolgd door een reeks gedragstests om de ontwikkeling van dominant en onderdanig gedrag te begrijpen. Het gebruik van dit model van traumatisch hersenletsel in combinatie met specifieke gedragstests maakt de studie van sociale stoornissen na hersenletsel mogelijk.

In deze studie willen we een onderzoek naar dominant-onderdanige relaties aantonen door middel van tests op basis van voedselconcurrentie. Het algemene doel van de huidige studie is om de ontwikkeling van dominant-onderdanige relatie na traumatisch hersenletsel aan te tonen. De voordelen van deze methode zijn de eenvoudige reproduceerbaarheid en het vermogen om dominant-onderdanig gedrag te observeren en nauwkeurig te analyseren.De dominant-onderdanige methode is goed vertegenwoordigd in de literatuur en is toegepast als model om manie en depressie te beoordelen. Dit model is ook gebruikt om stress in het vroege leven na moederscheiding bij volwassen ratten te onderzoeken. Selecteer om te beginnen willekeurig 30 volwassen mannetjes.Verdeel ze in traumatisch hersenletsel en schijngroepen. Zorg voor chow en water ad libitum. Voer vóór het letsel basislijnbeoordelingen uit van de neurologische ernstscore in beide groepen ratten zoals beschreven in het manuscript.Controleer na het verdoven van de ratten op de immobilisatie van de rat door te testen op gebrek aan beweging of pedaalreflex als reactie op een stimulerend middel. Voer craniotomie vier millimeter posterieur en vier millimeter lateraal van bregma uit. Met behulp van een vloeibaar percussieapparaat traumatisch hersenletsel veroorzaken gedurende 21 tot 23 milliseconden door de driewegstopkraan.Voer craniotomie uit op de groep schijnratten en infiltreer 0,1% bupivacaïne voordat de wond wordt gesloten. Verdeel de ratten een week voor de test willekeurig in kooien. Gebruik een apparaat gemaakt van twee transparante acrylglasdozen verbonden door een slanke tunnel.Vul vervolgens een feeder met gezoete melk en plaats deze in het tunnelmidden. Plaats elke rat gedurende 15 minuten in het apparaat voor bewoning op de eerste twee dagen. Selecteer willekeurig één rat uit de controlegroep en één uit de traumatische hersenletselgroep en zet ze op gelijke afstand van de feeder, zodat ze vijf minuten kunnen verkennen.Geef de ratten ad libitum toegang tot water. Plaats een camera. Installeer de aanbevolen computersoftware om de gegevens vast te leggen, op te slaan en te verwerken en neem de video op terwijl de ratten in de arena zijn.De neurologische ernstscore toonde aan dat 48 uur na de operatie de neurologische tekorten voldoende groter waren voor de traumatische hersenletselratten in vergelijking met de schijnratten. 28 dagen na de operatie waren de verschillen tussen de traumatisch hersenletsel en de schijngeopereerde groepen echter onbeduidend. De tijd doorgebracht bij de feeder was significant lager voor de traumatische hersenletselratten in vergelijking met de schijnratten.Minder traumatisch hersenletsel dan schijnratten kwamen als eerste naar de feeder. De belangrijkste dingen zijn het reinigen van het apparaat voor elke sessie en het uitvoeren van de test in een ruimte met een goede luchtcirculatie. Deze techniek maakt de weg vrij voor onderzoekers om dominant-onderdanig gedrag bij ratten na hersenletsel te onderzoeken.
