Черепно-мозговая травма (ЧМТ) вызвана внешней силой на мозг, часто связанной с падениями, спортивными травмами, физическим насилием или дорожно-транспортными происшествиями. В 2014 году Центры по контролю и профилактике заболеваний определили, что 2,53 миллиона американцев обратились в отделение неотложной помощи, чтобы обратиться за медицинской помощью в связи с несчастными случаями, связанными с ЧМТ1. Поскольку легкая ЧМТ (мЧМТ) составляет большинство случаев ЧМТ, за последние несколько десятилетий было принято несколько моделей ЧМТ, которые включают снижение веса, закрытую черепно-мозговую травму, управляемую поршнем, и контролируемое воздействие коры, ротационную травму, легкую жидкостную ударную травму и модели взрывной травмы 2,3. Из широко используемых моделей закрытой черепно-мозговой травмы одной из первых и наиболее широко используемых моделей является метод сброса веса, при котором на голову животного (обезболиваемый или бодрствующий) падает предмет с определенной высоты)2,4. В методе снижения веса тяжесть травмы зависит от нескольких параметров, в том числе от трепанации черепа, неподвижной или свободной головы, а также расстояния и веса падающего предмета 2,4. Одним из недостатков этой модели является высокая вариабельность тяжести травмы и высокий уровень смертности, связанный с угнетением дыхания 5,6. Распространенной альтернативой является нанесение удара с помощью пневматического или электромагнитного устройства, которое может быть выполнено непосредственно на открытой твердой мозговой оболочке (контролируемое корковое воздействие: CCI) или закрытом черепе (закрытая черепно-мозговая травма: CHI). Одной из сильных сторон поршневой травмы является ее высокая воспроизводимость и низкая смертность. Однако CCI требует трепанации черепа 7,8, а сама трепанациячерепа вызывает воспаление9. Вместо этого в модели ОМС нет необходимости в трепанации черепа. Как уже говорилось, каждая модель имеет ограничения. Одним из ограничений модели ОМС, описанной в данной статье, является то, что операция проводится с использованием стереотаксического каркаса, а голова животного обездвиживается. В то время как полная иммобилизация головы обеспечивает воспроизводимость, она не учитывает движение после удара, которое может способствовать травме, связанной с мЧМТ.
В этом протоколе описывается базовый способ выполнения воздействия ОМС с помощью коммерчески доступного электромагнитного ударного устройства10 в мыши. В этом протоколе подробно описаны точные параметры, необходимые для достижения высоковоспроизводимой травмы. В частности, исследователь имеет точный контроль над параметрами (глубина травмы, время выдержки и скорость удара) для точного определения тяжести травмы. Как описано, эта модель ОМС вызывает травму, которая приводит к двусторонней патологии, как диффузной, так и микроскопической (т.е. хронической активации глии, аксонального и сосудистого повреждения), а также поведенческим фенотипам 11,12,13,14,15. electromagnetic controlled closed-head model of mild traumatic brain injury in mice

Высоковоспроизводимые модели черепно-мозговой травмы (ЧМТ) на животных с четко определенными патологиями необходимы для тестирования терапевтических вмешательств и понимания механизмов того, как ЧМТ изменяет функцию мозга. Наличие нескольких моделей ЧМТ на животных необходимо для моделирования различных аспектов и тяжести ЧМТ, наблюдаемых у людей. В этой рукописи описывается использование средней закрытой черепно-мозговой травмы (ОМС) для разработки мышиной модели легкой ЧМТ. Модель считается мягкой, потому что она не вызывает структурных поражений головного мозга, основанных на нейровизуализации или грубой потере нейронов. Тем не менее, один удар создает достаточно патологии, чтобы когнитивные нарушения можно было измерить по крайней мере через 1 месяц после травмы.
