ジカウイルス(ZIKV)感染症は、蚊が媒介する新しいウイルス性疾患です。ZIKVの最初の孤立は1947年にウガンダで行われた1,2;1947年から2007年までは、臨床症状は無症候性であり、自己制限的な熱性疾患を特徴とするため、無視されたままでした。2007年、ジカ熱の流行はヤップ諸島で始まり3,4、続いて2013年から2014年にかけて太平洋地域(フランス領ポリネシア、イースター島、クック諸島、ニューカレドニア)でより大きな流行が続き5,6,7,8、重度の神経学的合併症であるギランバレー症候群(GBS)が初めて成人で報告されました9.2015年から2016年にかけて、早くも2013年にブラジルでZIKVのアジア遺伝子型が出現した後、最初の広範囲にわたるZIKVの流行が南北アメリカ大陸を席巻しました10。このアウトブレイクでは、新生児で44万〜130万件の小頭症やその他の神経障害が報告されました11。現在、ZIKV感染症の特定の治療法はありません。したがって、特に妊娠中の感染を予防できるZIKVワクチンの緊急の医学的必要性があります。
ウイルス定量は、サンプル中に存在するウイルスの数を決定するプロセスです。研究において重要な役割を担っており、学術研究所には医学や生命科学など多くの分野が関わっています。このプロセスは、ウイルスワクチン、組換えタンパク質、ウイルス抗原、抗ウイルス剤の製造などの商業分野でも重要です。ウイルスの定量には、多くの方法やアッセイを用いることができる12。方法やアッセイの選択は、通常、ウイルスの特性、望ましい精度のレベル、および実験や研究の性質によって異なります。一般に、ウイルスを定量する方法は、ウイルス核酸(DNAまたはRNA)の存在を検出する分子アッセイと、in vitroでウイルス感染性を測定するアッセイの2つのカテゴリーに分けることができます12。定量的ポリメラーゼ連鎖反応(qPCR、DNA用)または定量的逆転写ポリメラーゼ連鎖反応(qRT-PCR、RNA用)13およびデジタルドロップレットPCR14は、所与のサンプル中のウイルス核酸を定量するために使用される一般的な分子技術の例です15。しかし、これらの高感度の分子技術では、生存可能なウイルスと非生存可能なウイルスを区別することはできません15。そのため、細胞に対するウイルス感染力などの生物学的特徴に関する情報を必要とする研究は、上記の分子技術では完結できません。生存可能なウイルスの存在を測定および決定できるアッセイが必要です。ウイルスの感染性を測定するアッセイには、プラーク形成アッセイ(PFA)、50%組織培養感染線量(TCID50)、蛍光フォーカスアッセイ、透過型電子顕微鏡(TEM)などがあります12。
1952年にレナート・ダルベッコによって開発されたPFAは、ZIKV16を含むウイルス滴定に最も一般的に使用される方法の1つです。感染性溶解性ビリオンのウイルス濃度を直接測定するために使用されます。この方法は、溶解性ウイルスがウイルス感染後に接種された細胞単層に細胞変性効果(CPE、細胞死またはプラーク、感染していない細胞に囲まれた感染領域)を産生する能力に基づいています。しかし、このアッセイには、その有用性に影響を与えるいくつかの欠点があります。アッセイは時間がかかり(ウイルスにもよりますが、約7〜10日かかります)、CPEに依存し、エラーが発生しやすいです。本研究では、24ウェルプレートおよび96ウェルプレートフォーマットでZIKVを検出および定量するための免疫比色技術である焦点形成アッセイ(FFA)を報告します。
