1. iPSC 재프로그래밍 및 유지보수

</ol>4. 마이크로필라멘트 준비

</ol>5. 배아체(EB) 형성

	<li>RT에서 290 x g 에서 3분 동안 원심분리기.</li>
	<li>피펫으로 상청액을 흡인하고, 50 μM ROCK 억제제가 보충된 1 mL의 에센셜 6 (E6, 재료 표 참조) 배지에 세포 펠릿을 재현탁시키고, 혈구계19를 사용하여 세포를 계수한다.</li>
	<li>60,000μM의 ROCK 억제제가 보충된 E90,000 배지에서 원하는 파종 밀도에 따라 50-50 cells/mL의 세포 현탁액을 준비합니다( 재료 표 참조).</li>
	<li>150 μL의 세포 현탁액을 96-웰 ULA 플레이트 (또는 제조된 오목 플레이트20)의 각 웰에 첨가한다. 웰 당 9,000-11,000 개의 세포를 시드합니다.</li>
	<li>플레이트를 원심분리하여 세포를 RT에서 1분 동안 290 x g 에서 강제 응집하고, 플레이트를 5%CO2로 가습된 분위기에서 37°C의 인큐베이터에 놓는다.</li>
</ol>6. 신경상피 유도
<ol><li>24시간 후 피펫으로 배지 120μL를 조심스럽게 흡입합니다. 피펫 팁을 웰 안으로 너무 멀리 내려 EB를 흡인하지 않도록 하십시오.</li>
	<li>2 μM의 XAV939 및 SMAD 억제제로 보충된 150 μL의 E6 배지(실온에서)를 추가합니다: 웰당 10 μM의 SB431542 및 500 nM의 LDN 193189 ( 재료 표 참조).</li>

</ol>7. 오가노이드 분화 및 성숙

	<li>빈 P4 상자에 필름 시트(약 200인치 길이)를 놓아 열가소성 밀봉 필름( 재료 표 참조)에서 딤플 임베딩 시트를 준비합니다. 15mL 원뿔형 튜브 또는 500μL 미세 원심분리 튜브를 사용하여 필름 시트를 구멍에 부드럽게 눌러 12개의 딤플을 만듭니다. 필름 시트에 70% 에탄올을 뿌리고 UV 라이트를 켠 상태에서 플로우 후드 내부에서 최소 30분 동안 건조시킵니다.</li>
	<li>충분한 양의 기저막 매트릭스(Matrigel, 재료 표 참조)를 얼음 위에서 해동하고 플로우 후드 안에 넣습니다. 참고: EB당 약 30μL의 희석되지 않은 멤브레인 매트릭스가 필요합니다. 멤브레인 매트릭스가 겔화되는 것을 방지하기 위해 항상 4 °C 미만으로 유지하십시오. 사전 냉각된 피펫 팁은 피펫팅 중에 팁에서 매트릭스가 중합되는 속도를 늦추어 재료 손실을 줄이기 때문에 권장됩니다.</li>
	<li>광구경 P200 팁을 사용하여 각 딤플에 하나의 EB를 옮기고 일반 파이펫 팁으로 가능한 한 많은 배지를 제거합니다. EB가 마르지 않도록 주의하십시오. 일반 P200 팁을 사용하여 각 오가노이드에 ~30μL의 희석되지 않은 멤브레인 매트릭스를 추가하여 EB가 액적의 중심에 오도록 합니다.</li>

	<li>내장된 12개의 오가노이드 세트마다 ULA 6웰 플레이트의 한 웰에 비타민 A가 없는 B27이 포함된 분화 배지 5mL(표 1)를 준비합니다. 인큐베이터에서 플레이트를 37°C로 예열합니다.</li>

robust tissue fabrication for long-term culture of ipsc-derived brain organoids for aging research

초기 신경 유도 기간은 이전에 발표된 절차(11)와 비슷하다. 배아체(EB)는 가장자리가 흰색 또는 투명한 원형 응집체(그림 2B)로 시작합니다. 오가노이드의 치수와 외부 형태는 오가노이드 내부의 복잡한 구조로 보완됩니다. 분화의 첫 번째 단계 후 조직을 DIV7에 고정한 후, EB의 세포는 다분화능 신경 줄기 세포21의 마커 역할을 하는 HMG 상자 전사 인자인 SOX2와 신경 전구 세포를 나타내는 쌍을 이루는 단백질 상자 Pax-6을 발현합니다(그림 3). TuJ1(클래스 III β-튜불린)22 로 표시된 미성숙 뉴런은 이미 조직 전체에 흩어져 있는 것을 볼 수 있습니다.

오가노이드가 노화됨에 따라 발달 중인 뉴런의 조직과 마커가 생리적 조건을 복제하기 시작합니다. DIV30에서는 발달 중인 뇌와 동등한 신경성 영역을 생성하는 많은 로제트를 관찰할 수 있다25. DIV60에 의해 이러한 SOX2+ 신경원성 영역은 존재하지 않으며 성숙한 MAP2 및 NeuN26, 뉴런 분화 마커 및 양성 뉴런으로 대체됩니다(그림 4 및 그림 5).
