甲状腺がんは、甲状腺上皮に由来する最も一般的な内分泌悪性腫瘍1の1つです。近年、甲状腺がんの発生率は世界中で急速に増加しています2。甲状腺がんは、腫瘍細胞の分化の程度に応じて異なるタイプに分類できます。甲状腺がんは、臨床行動と組織学に基づいて、甲状腺乳頭がん(PTC)や濾胞性甲状腺がん(FTC)、低分化型がん(PDTC)、甲状腺の未分化がん(ATC)などの高分化型がんに分類されます3。軽度の行動と予後が良好な一般的なタイプであるPTCとは対照的に4、ATCはまれで非常に侵攻性の高い悪性腫瘍であり、すべての甲状腺腫瘍の2%から3%を占めています5。ATCはまれですが、甲状腺がん関連の死亡の約50%の原因であり、生存期間は悲惨です(6〜8か月)6,7。ATC症例の50%以上が肺転移と診断されています8。ATCの攻撃的な性質に加えて、限られた効果的な治療法が診療所で開発されています。したがって、ATC患者は暗い予後を有する9、10、11。このことは、ATCの開発と治療の根底にある分子メカニズムについて、さらなる詳細な研究が緊急に必要であることを示唆しています。
ATCの腫瘍形成は動的な脱分化過程である。臨床研究の各段階でヒト腫瘍サンプルを収集することの難しさは、高分化型癌から未分化癌への発生メカニズムの理解を妨げてきました。対照的に、マウスATCモデル(mATC)の使用は、腫瘍形成過程全体におけるmATCサンプルの収集に有利である。したがって、動的脱分化過程を解析することで、腫瘍形成のメカニズムをよりよく理解することができます。さらに、臨床ATCサンプルの不均一性も、分子メカニズムの理解の難しさの一因となっています。それにもかかわらず、マウスは同じ遺伝的背景を共有し、同様の生活環境で維持され、各腫瘍の一貫性を確保しました。これにより、ATC開発の一般化された役割の調査が容易になります12、13、14。さらに、mATCは、解剖学的位置と組織特異的微小環境の影響を回復できるin situ腫瘍モデルです。このように、一般的に使用される免疫不全マウスと比較して、mATCは無傷の免疫系および免疫微小環境を有する自発的なマウスモデルである。
そこで、脱分化型甲状腺癌の病理学的特徴を再現できるマウスモデルであるC57BL/6株を用いて条件付き誘導mATCを構築した。このモデルに基づいて、mATCの分子基盤、構築のアイデア、病理学的特徴、およびアプリケーションの概要を簡単に説明しました。また,mATCの腫瘍増殖,生存期間,転移,病理学的特徴を観察・報告した.これは、他の研究者がこのモデルを簡単に使用できるようにするための有益な概要になると考えています。
McFadden15によって最初に報告された条件付き誘導性mATCモデルを構築しました。最初に、TPO-cre/ERT2、Braf flox/wt、およびTrp53flox/wtのマウスを作製しました。 具体的には、TPO-cre/ERT2マウスには、ヒト甲状腺ペルオキシダーゼ(TPO)プロモーター(甲状腺特異的プロモーター)が含まれ、cre-ERT2融合遺伝子(ヒトエストロゲン受容体リガンド結合ドメインに融合したcreリコンビナーゼ)の発現を駆動しました。Cre-ERT2は通常、細胞質に限局し、タモキシフェンにさらされた場合にのみ核に入り、creが組換え酵素活性を発揮するように誘導します。マウスをloxP隣接配列を有するマウスと交配すると、タモキシフェン誘導後、cre媒介組換えにより甲状腺細胞内のfloxed配列が除去され、特定の遺伝子をノックアウトまたはノックする目的が達成されます。
さらに、Brafflox/wtマウスは、cre-loxPシステムに基づくヒトBrafのノックイン対立遺伝子です。Brafflox/wtマウス転写産物は、内因性エクソン1〜14およびloxP隣接ヒトエクソン15〜18によってコードされています。FLOXED領域のcreを介した切除後、変異エクソン15(ヒト癌において構成的に活性なBraf V600Eと結合したV600Eアミノ酸置換で修飾)および内因性エクソン16〜18を使用して転写物を生成します。さらに、Trp53 flox/wtマウスはヒトTrp53のノックアウト対立遺伝子であり、Trp53のエクソン2〜10に隣接するloxP部位を有する。creリコンビナーゼを有するマウスと交配すると、cre媒介組換えは、Trp53をノックアウトするためにフロックス配列を欠失させる。次に、TPO-cre/ERT2、Braf flox/w、およびTrp53flox/wtマウスを交配してTB(TPO-cre/ERT2;ブラフフロックス/重量)マウスおよびTBP(TPO-cre/ERT2;ブラフフロックス/重量;Trp53flox/wt)マウスは、PTCおよびATCを生成するために使用することができる。約8週間後、マウスは、コーン油に溶解した150 mg / kgのタモキシフェンを2回の投与で腹腔内(i.p.)投与することにより誘導されました。腫瘍の成長は高周波超音波検査によってモニターすることができた(超音波検査の最初の時点は0日目として記録された)。最初の超音波検査は、タモキシフェン導入の40日後に実施した。

spontaneous murine model of anaplastic thyroid cancer

未分化甲状腺がん(ATC)はまれですが致命的な悪性腫瘍であり、予後は悲惨です。ATC患者では標準治療が本質的に枯渇しているため、ATCの発がんと発症、および治療法に関するより詳細な研究が緊急に必要です。しかし、有病率が低いため、徹底的な臨床研究や組織サンプルの採取が妨げられており、効果的な治療法の作成はほとんど進んでいません。遺伝子工学を用いて、C57BL/6バックグラウンドで条件付き誘導性ATCマウスモデル(mATC)を作成しました。ATCマウスモデルはTPO-cre/ERT2によって遺伝子型決定されました。ブラフCA / wt;Trp53ex2-10 / ex2-10およびタモキシフェンによる腹腔内注射によって誘導された。マウスモデルを用いて、腫瘍動態(導入後4ヶ月後の腫瘍サイズは12.4 mm 2から32.5 mm2の範囲)、生存期間(生存期間の中央値は130日)、転移(肺転移はマウスの91.6%で発生した)曲線および病理学的特徴(Cd8、Foxp3、F4/80、Cd206、Ki67、およびCaspase-3免疫組織化学染色によって特徴付けられる)を調査した。その結果、自然発生型mATCはヒトATC腫瘍と高度に類似した腫瘍動態と免疫学的微小環境を有することが示された。結論として、mATCモデルは、病態生理学的特徴と統一された遺伝子型の高い類似性により、臨床ATC組織とサンプルの不均一性の不足をある程度解決しました。したがって、ATCのメカニズムとトランスレーショナル研究を促進し、ATCに対する低分子医薬品と免疫療法剤の治療可能性を調査するためのアプローチを提供します。
