Denne protokol bruger ultralyd med høj opløsning til at analysere ortopiske ATC-tumorer i en genetisk manipuleret musemodel. Selvom anaplastisk skjoldbruskkirtelkarcinom (ATC) tegner sig for færre end 2% af kræft i skjoldbruskkirtlen, forårsager det mere end 50% af skjoldbruskkirtelkræftrelaterede dødsfald årligt. Den mediane overlevelsestid efter diagnose med ATC er kun ca. 6 måneder, og der findes ingen behandlinger, der signifikant forbedrer overlevelsen 1,2.
Sjældenheden af ATC har hæmmet forskningen, der studerer, hvordan sygdommen begynder og aggressivt skrider frem. Gensplejsede musemodeller, der efterligner sygdommen, er for nylig blevet tilgængelige, som giver indsigt i sygdommen og dens reaktioner på mulige behandlinger 3,4,5. Sådanne undersøgelser kræver nøjagtig tumorbilleddannelse til målinger og overvågning, som typisk udføres ved hjælp af magnetisk resonansbilleddannelse, computertomografi eller ultralydsscanning med høj opløsning 6,7. Ultralyd har været meget udbredt i museorganer. Det har fordele i forhold til magnetisk resonansbilleddannelse og computertomografi, da det kan udføres i realtid og ikke udsætter motivet for stråling, og det nødvendige udstyr er lille nok til at være bærbart 8,9. Imidlertid er undersøgelser af kontinuerlig overvågning af ATC-vækst ved hjælp af ultralyd sjældne; Derfor udforsker dette arbejde nytten af ultralyd i denne sammenhæng.
Her præsenteres en protokol til brug af ultralyd med høj opløsning til nøjagtigt at scanne, overvåge og måle tumorer i en musemodel af ATC.

high-resolution ultrasonography for the analysis of orthotopic atc tumors in a genetically engineered mouse model

Anaplastisk thyreoideakarcinom (ATC) er forbundet med en dårlig prognose og kort median overlevelsestid, men ingen effektiv behandling forbedrer resultaterne betydeligt. Gensplejsede murinmodeller, der efterligner ATC's progression, kan hjælpe forskere med at studere behandlinger for denne sygdom. Krydsning af tre forskellige genotyper af mus, en TPO-cre / ERT2; BrafCA/wt; Trp53 Δex2-10/Δex2-10 transgen ATC-model blev udviklet. ATC-murinmodellen blev induceret ved en intraperitoneal injektion af tamoxifen med overekspression af BrafV600E og deletion af Trp53, og tumorerne blev genereret inden for ca. 1 måned. Ultralyd med høj opløsning blev anvendt til at undersøge tumorinitiering og progression, og den dynamiske vækstkurve blev opnået ved at måle tumorstørrelserne. Sammenlignet med magnetisk resonansbilleddannelse (MRI) og computertomografiscanning har ultralyd fordele ved at observere ATC-murinmodellen, såsom at være ikke-invasiv, bærbar, i realtid og uden strålingseksponering. Ultralyd med høj opløsning er velegnet til dynamiske og flere målinger. Imidlertid kræver ultralydsundersøgelse af skjoldbruskkirtlen hos mus relevant anatomisk viden og erfaring. Denne artikel indeholder en detaljeret procedure til brug af ultralyd med høj opløsning til at scanne tumorer i den transgene ATC-model. Denne protokol hjælper brugerne med at udnytte ultralyd med høj opløsning til at scanne, overvåge og måle tumorer i musemodellen af anaplastisk skjoldbruskkirtelkarcinom. Ultralyd har vist sig pålidelig hos mus til billeddannelse væv, der optager lignende kropssteder som humane væv, herunder leveren, skjoldbruskkirtlen, og fosteret. For at begynde skal du tænde ultralydssystemet og oprette en ny mappe til optagelse af billederne og indsamling af data.Vælg derefter 40-megahertz-linjesonden, klik på overfladisk vævsmønster for at aktivere den overfladiske vævstransducer, og brug den resulterende B-tilstand til skjoldbruskkirtelbilleddannelse. Rengør dyrebilledplatformen, og tænd for varmeknappen, så platformens temperatur når 38 til 40 grader Celsius, før du placerer dyret på den. Bedøv musen, og læg den på den opvarmede platform.Dæk snuten med en næsekegle, der er forbundet til anæstesiudløbet. Brug derefter tape med musen i liggende stilling til at fastgøre lemmerne til den opvarmede platform. Til tumorbilleddannelse skal du justere billeddannelsessystemet for at optimere opløsningen.Indstil den todimensionelle forstærkning til 25 til 30 decibel, billeddybden til 10 millimeter og centrer til tre til seks millimeter. Hold sonden, læg den i kontakt med ultralydgelen på brystet.Scan derefter fra brystet mod halsen for at finde skjoldbruskkirtlen. Identificer anaplastiske skjoldbruskkirtelkarcinomtumorer fra luftrøret og remmusklerne afhængigt af den anatomiske placering og ultralydekko. Baseret på indtrykket af den samlede tumor skal du bekræfte billedafsnittet med den største venstre mod højre tumordiameter.Tryk derefter på fryseknappen, og mål anteroposterior og venstre mod højre tumordiametre ved hjælp af ultralydstykkelsen. Når scanningerne er afsluttet, skal du tørre ultralydgelen og fjerne fastholdelsesbåndet fra dyrets lemmer. ATC-tumorstørrelsen af højre og venstre lobes blev målt over fire tidsintervaller.Ved den fjerde måling var den gennemsnitlige højre ATC-størrelse vokset til 11.844 kvadratmillimeter, mens tumorstørrelsen på venstre lap var vokset til 9.280 kvadratmillimeter, hvilket gjorde den samlede gennemsnitlige størrelsesforøgelse til 15.843 kvadratmillimeter. Interessant nok var tumorstørrelsen i musen mærket P92 vokset til næsten fire gange større end størrelsen på den oprindelige måling. Den største fordel ved denne teknik er, at ultralyd er fleksibel og kan udføres i realtid uden stråling.
