Selv om anaplastisk skjoldbruskkarsinom (ATC) står for færre enn 2% av skjoldbruskkreftene, forårsaker det mer enn 50% av skjoldbruskkreftrelaterte dødsfall årlig. Median overlevelsestid etter diagnose med ATC er bare ca. 6 måneder, og det finnes ingen behandlinger som signifikant forbedrer overlevelsen 1,2.
Sjeldenheten av ATC har hemmet forskningen som studerer hvordan sykdommen begynner og aggressivt utvikler seg. Genetisk konstruerte musemodeller som etterligner sykdommen har nylig blitt tilgjengelige, noe som gir innsikt i sykdommen og dens respons på mulige behandlinger 3,4,5. Slike studier krever nøyaktig tumoravbildning for målinger og overvåking, som vanligvis utføres ved hjelp av magnetisk resonansbilder, computertomografi eller høyoppløselig ultralyd 6,7. Ultralyd har vært mye brukt i museorganer. Det har fordeler i forhold til magnetisk resonansavbildning og computertomografi siden det kan utføres i sanntid og ikke utsetter motivet for stråling, og det nødvendige utstyret er lite nok til å være bærbart 8,9. Imidlertid er studier på kontinuerlig overvåking av ATC-vekst ved hjelp av ultralyd sjeldne; Derfor utforsker dette arbeidet nytten av ultralyd i denne sammenhengen.
Her presenteres en protokoll for bruk av høyoppløselig ultralyd for nøyaktig skanning, overvåking og måling av svulster i en musemodell av ATC.

high-resolution ultrasonography for the analysis of orthotopic atc tumors in a genetically engineered mouse model

Anaplastisk skjoldbruskkarsinom (ATC) er assosiert med dårlig prognose og kort median overlevelsestid, men ingen effektiv behandling forbedrer resultatene signifikant. Genetisk utviklede murine modeller som etterligner ATCs progresjon kan hjelpe forskere til å studere behandlinger for denne sykdommen. Krysser tre forskjellige genotyper av mus, en TPO-cre/ERT2; BrafCA / wt; Trp53 Δex2-10/Δex2-10 transgen ATC-modell ble utviklet. Denne protokollen hjelper brukerne til å bruke høyoppløselig ultralyd for å skanne, overvåke og måle svulster i musemodellen av anaplastisk skjoldbruskkarsinom. Ultralyd har vist seg pålitelig hos mus for avbildning av vev som opptar lignende kroppssteder som menneskelig vev, inkludert leveren, skjoldbruskkjertelen og fosteret. For å begynne, slå på ultralydssystemet, og opprett en ny mappe for å ta bildene og samle inn data.Velg deretter 40-megahertz-linjesonden, klikk på overfladisk vevsmønster for å aktivere den overfladiske vevstransduseren, og bruk den resulterende B-modusen for skjoldbruskbilde. Rengjør dyrebildeplattformen, og slå på varmeknappen for at plattformtemperaturen skal nå 38 til 40 grader Celsius før du plasserer dyret på den. Etter å ha fullført skanningene, tørk ultralydgelen og fjern fastholdelsesbåndet fra dyrets lemmer. ATC-tumorstørrelsen på høyre og venstre ble målt over fire tidsintervaller.På den fjerde målingen hadde den gjennomsnittlige høyre ATC-størrelsen vokst til 11.844 kvadratmillimeter, mens tumorstørrelsen på venstre lobe hadde vokst til 9.280 kvadratmillimeter, noe som gjorde totalen til en gjennomsnittlig størrelsesøkning til 15.843 kvadratmillimeter.
