歴史的に、科学者は創薬のために前臨床動物実験に依存してきましたが、人間の転帰との相関性が低いため、これらの方法の多くが疑問視されてきました1。動物実験を置き換え、削減し、改良するための「3R」原則の実施は、科学者に前臨床薬物および化学毒物学のリスク評価をサポートするための新しいin vitro代替方法を見つけることを促します2。しかし、これまでに開発された多くのin vitroモデルは、人間の生体器官の動的な性質を再現するために必要な生物学的構造、細胞の複雑さ、および機械的環境を欠いています3,4。
従来のin vitro前臨床システムでは、通常、硬質プラスチック表面上で増殖したヒト細胞の2D単培養が使用されます。これらの方法は、簡単な機構研究を行うためのツールを提供し、医薬品候補の迅速なスクリーニングを可能にします。2Dモデルは比較的低コストで堅牢性が高いため、自動ハイスループットシステムと組み合わされることが多く、医薬品開発プロセスの初期段階で潜在的な医薬品候補を迅速に特定するために使用されます5,6。しかし、このような2Dモデルは、治療薬候補に対する組織レベル、臓器レベル、または全身反応をモデル化するためのトランスレーショナルアプローチを提供しておらず、開発の前臨床段階での医薬品の安全性と有効性を正確に予測する必要があります。フラットセル培養では、複雑な多細胞相互作用、生体力学的特性、ヒト組織の3次元(3D)構造など、天然の組織の微小環境を再現することはできません7。平坦な表面上で増殖する細胞は、成熟した表現型を獲得しないことが多く、したがって、天然組織の場合のように薬理学的刺激に応答することができません。例えば、in vitroで増殖した初代ヒト肺胞上皮細胞は、扁平上皮表現型を示し、サーファクタントタンパク質CおよびB(SP-CおよびSP-B)8を含む重要な表現型マーカーを失います。不十分な分化に加えて、初代細胞は、組織の炎症に関連する特定の生化学的経路が機能しなくなるため、in vitroで生物学的ストレッサーに対して鈍感になることがよくあります9。このような細胞機能の喪失は、主に硬い基質の使用と、肺線維芽細胞や平滑筋細胞などの組織特異的間質細胞によって自然に放出される可溶性因子の欠如に関連しているようです10,11。
化学的物理的および生物学的複雑さの欠如がin vitroでの細胞の生理学的挙動を制限することを理解することで、より洗練された多細胞モデルの開発が促進され、体外のヒト組織の複雑さをよりよく捉えることが証明されています12,13。1970年代初頭に最初の共培養モデルが作成されて以来14、合成および天然のヒドロゲルの導入により、天然の組織微小環境を模倣する能力が大幅に向上し、細胞分化を促進し、細胞の自己組織化を組織様構造に導き、天然の組織機能を回復するための貴重なツールになりました15,16.例えば、適切な3D足場で増殖させると、ヒト細胞はスフェロイドやオルガノイドなどの機能的な構造に自己配列し、幹細胞マーカーを発現し、自己複製することができます17。対照的に、ヒト細胞(幹細胞を含む)は、従来の2D基質上で増殖すると、急速に老化し、数回継代した後に老化を受ける18。さらに、ヒドロゲルは、多孔性、孔径、繊維の太さ、粘弾性、トポグラフィー、剛性などの特定の組織特性に合わせて「調整」することも、組織由来の細胞成分および/または生理活性分子でさらに操作して、生理学的または病理学的状態のエミュレーションを可能にすることもできます19,20.医薬品研究で使用される3Dヒドロゲルベースのモデルは、薬物試験の大きな可能性を秘めているにもかかわらず、in vivo組織の複雑な細胞構造を完全に再現しておらず、静水圧、周期的伸張、流体せん断など、人体に通常存在する重要な血行動態および機械的刺激を欠いています21。
臓器チップ(OOC)などの微小生理学的システム(MPS)は、インビトロで複雑な生理学的応答をキャプチャできるツールとして最近登場しました22,23。これらのモデルは、多くの場合、生体の動的な微小環境のモデリングを可能にするマイクロ流体プラットフォームの使用を採用しています。
3D組織バイオエンジニアリングとメカノバイオロジーの原理を組み合わせて、複雑なヒト上皮組織のオープントップチップモデルを作成しました。これにより、上皮組織の多細胞で動的な微小環境を詳細に再現することができました。これには、生体器官に自然に存在するが、従来のin vitroモデルでは無視されることが多い組織特異的な生化学的および生体力学的手がかりが含まれます24。Open-Top Chipには、多孔質膜で区切られた血管コンパートメント(図1A)と間質コンパートメント(図1B)の2つのコンパートメントが組み込まれており、2つのチャンバー間で栄養素を拡散させることができます(図1C)。血管コンパートメントは、生理学的せん断応力を再現するために連続的な流体の流れにさらされ、間質チャンバーの伸縮性のある設計により、呼吸運動または腸の蠕動運動に関連する機械的ひずみのモデリングが可能になります。間質コンパートメントには、組織特異的線維芽細胞の生理学的成長をサポートするように設計された調整可能な3Dヒドロゲル足場が収容されています。これは、空気 - 液体界面の確立を容易にする取り外し可能な蓋を有し、粘膜組織のヒト生理機能のより大きなエミュレーションを可能にする条件、ならびに上皮層に直接薬物を投与するための組織への直接アクセスを可能にする。補足図1は、寸法や生物学的コンパートメント(補足図1A-D)を含むオープントップチップ設計の主要コンポーネントのいくつかと、このプロトコルで説明されている主な技術的ステップ(補足図1E)を示しています。
オープントップチップの灌流は、プログラム可能な蠕動ポンプで実現されます(図1D)。ペリスタルティックポンプのセットアップにより、12個のオープントップチップを同時に灌流できます。ほとんどのインキュベーターは、インキュベーターあたり最大24チップの培養を可能にする2つのセットアップを収容できます。機械的延伸は、カスタムメイドのプログラム可能な真空圧力調整器を使用して実現されます(図1E)。これは、デジタル-アナログコンバータによって電子的に制御される電空真空レギュレータで構成されています。言い換えれば、電空真空レギュレータは、ユーザーが決定する振幅と周波数を持つ正弦波真空プロファイルを生成します。0%から15%の範囲の周期ひずみは、0〜-90kPaの範囲の振幅と0.2Hzの周波数でオープントップチップの真空チャネルに負圧を加えることによって生成されます。これは、以前に採用され、他の論文25に記載されている市販のFlexcellひずみユニットと同等のカスタムメイドのシステムです。例えば、肺の呼吸運動や腸の蠕動運動に関連する機械的組織変形を模倣するために、空気圧アクチュエータは正弦波真空/ひずみ波を適用し、その大きさと振幅は、ヒト細胞が本来の組織で経験する歪みと周波数の生理学的レベルに一致するように調整できます。

Reconstituting Cytoarchitecture and Function of Human Epithelial Tissues on an Open-Top Organ-Chip

従来の拡散培養法では、生体臓器の複雑な環境を再現できないことが多く、組織特異的なマーカーや機能が失われていました。私たちは、拡散培養とマイクロ流体原理を組み合わせて、上皮組織の微小環境を厳密に模倣するオープントップチップを開発しました。生体臓器に自然に存在するが、従来のin vitroモデルでは無視されがちな生化学的および生体力学的手がかりを含めることにより、オープントップチップはクローズドシステム間のより良い代替手段となります。手順を実演するのは、今日、私の研究室の研究生であるアディア・パンチャルです。はじめに、必要な試薬を氷上のバイオセーフティキャビネットの下に置きます。組織特異的間葉系細胞を培養して80%〜90%のコンフルエント性を得た後、メーカーの推奨に従ってトリプシンまたは他の方法を使用して細胞を解離します。細胞を250gで摂氏24度で5分間ペレット化します。細胞ペレットを氷冷した10X EMEMおよび再構成バッファーのそれぞれ225マイクロリットルに再懸濁します。ピペッティングによって細胞を混合し、1, 800マイクロリットルの氷冷コラーゲン1溶液を加える。次に、プレゲル溶液を氷上で5〜6回ピペッティングして混合します。プレゲル溶液を18マイクロリットルの通常の水酸化ナトリウム1個で中和し、ピペッティングで穏やかに混合した後、気泡を避けながら150マイクロリットルの細胞含有ヒドロゲルをオープントップチップの中央チャンバーにピペットで入れます。各ペトリ皿に2ミリリットルの滅菌二重蒸留水で満たされた遠心分離管キャップを含む、チップを別々のペトリ皿にグループ化し、摂氏37度と5%二酸化炭素でペトリ皿をインキュベートします。ストローマヒドロゲルの表面マイクロパターニングを行うには、滅菌3Dプリントスタンプのパターン化された表面に中和コラーゲン1プレゲル溶液20マイクロリットルをピペットで移し、ストローマヒドロゲルがまだ液体の形である間にオープントップチャンバー内にスタンプを挿入します。アスピレーターを使用して、オープントップチャンバーの上部からこぼれる可能性のあるヒドロゲルの残留物を取り除きます。前に示したように、すべてのチップを水で満たされた円錐形のチューブキャップを備えた別々のペトリ皿にグループ化します。そして、すべてのペトリ皿を摂氏37度と5%二酸化炭素で90分間インキュベートします。インキュベーションの最後に、チップをバイオセーフティキャビネットの下に戻し、精密ピンセットを使用してスタンプをそっと取り除き、ヒドロゲルを損傷するリスクを減らします。培養細胞が80%から90%のコンフルエントに達したら、細胞提供者が推奨するタンパク質分解酵素手順を使用して細胞を解離します。解離後、遠心分離によって細胞をペレット化し、原稿に記載されているように適切な細胞断片密度に上皮細胞を再懸濁する。上皮細胞を播種するには、チップをインキュベーターからバイオセーフティキャビネットに移し、間質表面を100マイクロリットルの新鮮な上皮細胞培養培地で3回すすぎ、余分なコーティング溶液を取り除きます。リンス媒体が吸引されたら、適切な細胞密度を使用して50マイクロリットルの上皮細胞懸濁液をヒドロゲル表面に播種し、チップをインキュベーターに2時間戻します。細胞の破片を除去するには、細胞培養培地でヒドロゲル表面を2回穏やかにすすぎ、密閉されたオートクレーブ可能な容器でオートクレーブして培地をリフレッシュし、チップを蠕動ポンプに接続します。ペリスタルティックポンプを一時停止し、チップをポンプから慎重に外し、チップハウジングキャリアを引き出します。チップをインキュベーターからバイオセーフティキャビネットに移した後、上部のリザーバーに残っている培地の量を取り除きます。次に、上部のマイクロ流体チャネルからすべての媒体を穏やかに吸引し、バインダークリップを使用して上部の入口に接続された短いマイクロ流体チューブをクランプして、媒体の蒸発と気液界面(ALI)の維持を減らします。オープントップチップをハウジングキャリアに置き、インキュベーターに戻します。チップをペリスタルティックポンプに再接続し、ペリスタルティックポンプを始動して流れを再開します。血管室を内皮細胞培養培地ですすぎ、次いで25マイクロリットルの内皮細胞懸濁液を播種する。チップを逆さまにして、内皮細胞がマイクロ流体チャンバーの上面に付着できるようにします。チップをペトリ皿にグループ化します。前に示したようにそれらをインキュベーターに戻し、内皮細胞を1時間付着させます。インキュベーションの最後に、チップをインキュベーターからバイオセーフティキャビネットに移し、血管チャネルを内皮細胞培養培地で2回すすぎ、細胞の破片を除去します。血管チャネルの底面への内皮細胞の接着を容易にするために、チップを平らに置く。バイオセーフティキャビネットの下の脱気された血管細胞培養培地で血管培地リザーバーを満たします。チップをチップハウジングキャリア内に戻し、チップを一方の端の媒体リザーバーともう一方の端のペリスタルティックポンプに再接続します。マイクロ流体接続を検査して、すべてのチップが正しく接続され、媒体の滴りの目に見える液滴がないことを確認します。次に、蠕動ポンプを始動して流体の流れを再開します。細胞の有無にかかわらず、マイクロパターン化されたゲル表面が視覚化されます。組織学的分析により、チップ上で分化した成熟した多層層状表皮が明らかになりました。皮膚チップの上面図は、真皮層の内側の線維芽細胞の存在を示した。PECAM-1、VE-cadherin、およびvon Willebrandは、オープントップスキンチップで共培養したヒト微小血管内皮細胞の分化を示しました。MUC5AC、αおよびβチューブリン、クロロ細胞タンパク質16、p63、およびZO-1蛍光染色は、成熟気道上皮を示した。拍動繊毛と分化した成熟擬似重層上皮がオープントップ気道チップで観察されました。I型、II型、およびE-カドヘリン蛍光染色では、チップ上に成熟肺細胞の存在が示されました。電子顕微鏡検査は、成熟した肺胞表現型の証拠である微絨毛およびリソソーム小胞の存在を明らかにした。組織学的解析では、I型表現型と一致する扁平扁平上皮細胞、II型表現型と一致する直方体玉石様細胞、および真皮層内の線維芽細胞の存在が示されました。腸細胞および成熟杯の存在は、ムチン2およびE-カドヘリン染色によって確認された。真皮層内部の線維芽細胞は単純な円柱状上皮の存在を確認した。すべての試薬は冷たくなければならず、ゲルの表面は十分にすすがれて均一な表面を得る必要があります。接着していない細胞や破片を取り除き、細胞接着を均一にし、コンパクトな汚れた単層と血管壁を得ることが重要です。同様のアプローチを使用して、腸や肺などの他のタイプの上皮器官モデルを生成し、特定の薬物または環境要因がヒト組織の恒常性にどのように影響するかを評価し、特に上皮および血管バリア機能に重点を置くことができます。間質コンパートメントの有効性段階に簡単にアクセスできるため、研究は特定の顔パターンを生成し、他のデバイスでは達成が困難なcryptviveoliなどの機能的な組織構造を再現できるようになりました。
