Het hier beschreven protocol biedt een methode voor het evalueren en monitoren van het dynamische proces van mitofagie in levende cellen, waarbij autofagosomen, lysosomen en mitochondriale splijting betrokken zijn, door co-kleuring met celpermeant mitochondriën en lysosoomkleurstoffen. De methode kan ook worden gebruikt om mitochondriën te identificeren en mitochondriale morfologie te beoordelen. Mitochondriën zijn de krachtpatsers van bijna alle eukaryote cellen 1,2. Naast ATP-productie door oxidatieve fosforylering spelen mitochondriën een vitale rol in andere processen zoals bio-energetica, calciumhomeostase, vrije radicalengeneratie, apoptose en cellulaire homeostase 3,4,5. Omdat mitochondriën reactieve zuurstofsoorten (ROS) genereren uit meerdere complexen in de elektronentransportketen, worden ze voortdurend gestimuleerd door potentiële oxidatieve stress, wat uiteindelijk kan leiden tot structurele schade en disfunctie wanneer het antioxidantafweersysteem instort 6,7. Mitochondriale disfunctie blijkt bij te dragen aan vele ziekten, waaronder metabole stoornissen, neurodegeneratie en hart- en vaatziekten8. Daarom is het cruciaal om gezonde mitochondriale populaties en hun juiste functie te behouden. Mitochondriën zijn zeer plastische en dynamische organellen; hun morfologie en functie worden gecontroleerd door mitochondriale kwaliteitscontrolemechanismen, waaronder posttranslationele modificaties (PTM) van mitochondriale eiwitten, mitochondriale biogenese, fusie, splijting en mitofagie 9,10. Mitochondriale splijting gemedieerd door dynamin-gerelateerd eiwit 1 (DRP1), een GTPase van de dynamin superfamilie van eiwitten, resulteert in kleine en ronde mitochondriën en isoleert de disfunctionele mitochondriën, die kunnen worden opgeruimd en afgebroken door mitofagie11,12.
Mitofagie is een cellulair proces dat selectief mitochondriën afbreekt door autofagie, meestal optredend in beschadigde mitochondriën na letsel, veroudering of stress. Vervolgens worden deze mitochondriën aan lysosomen geleverd voor afbraak10. Mitofagie is dus een katabolisch proces dat helpt de kwantiteit en kwaliteit van mitochondriën in een gezonde toestand te behouden in een breed scala aan celtypen. Het speelt een cruciale rol bij het herstel van cellulaire homeostase onder normale fysiologische en stressomstandigheden13,14. Cellen worden gekenmerkt door een complex mitofagiemechanisme, dat wordt geïnduceerd door verschillende signalen van cellulaire stress en ontwikkelingsveranderingen. Mitofagie regulerende routes worden geclassificeerd als ubiquitine-afhankelijk of receptor-afhankelijk15,16; de ubiquitine-afhankelijke autofagie wordt gemedieerd door het kinase PINK1 en de rekrutering van ubiquitineligase Parkin E3 naar de mitochondriën 17,18, terwijl receptorafhankelijke autofagie de binding van autofagiereceptoren aan de microtubule-geassocieerde eiwitlichtketen LC3 omvat die mitofagie bemiddelt als reactie op mitochondriale schade19.
Transmissie-elektronenmicroscopie (TEM) is de meest gebruikte methode, en nog steeds een van de beste methoden, om mitofagie te observeren en te detecteren20. De morfologische kenmerken van mitofagie zijn autofagosomen of autolysosomen gevormd door de fusie van autofagosomen met lysosomen, die kunnen worden waargenomen op elektronenmicroscopiebeelden21. De zwakte van elektronenmicroscopie (EM) is echter het onvermogen om de dynamische processen van mitofagie, zoals mitochondriale depolarisatie, mitochondriale splijting en fusie van autofagosomen en lysosomen, in de levende celte volgen 20. Het evalueren van mitofagie door middel van beeldvorming van levende organellen is dus een aantrekkelijke alternatieve methode voor mitochondriaal onderzoek. De hier beschreven live cell imaging-techniek maakt gebruik van twee fluorescerende kleurstoffen om mitochondriën en lysosomen te kleuren. Wanneer mitofagie optreedt, worden beschadigde of overbodige mitochondriën die door autofagosomen worden overspoeld, groen gekleurd door de mitochondriale kleurstof, terwijl de rode kleurstof de lysosomen kleurt. De fusie van deze autofagosomen en lysosomen, aangeduid als autolysosomen, zorgt ervoor dat de groene en rode fluorescentie elkaar overlappen en zich manifesteren als gele stippen, wat wijst op het optreden van mitofagie22. De celpermeante mitochondriakleurstof (MitoTracker Green) bevat een mild thiol-reactieve chloromethylgroep om mitochondriën23 te labelen. Om mitochondriën te labelen, worden cellen eenvoudig geïncubeerd met de kleurstof, die passief over het plasmamembraan diffundeert en zich ophoopt in actieve mitochondriën. Deze mitochondriale kleurstof kan gemakkelijk levende cellen kleuren en is minder effectief in het kleuren van aldehyde-vaste of dode cellen. De lysosoomkleurstof (LysoTracker Red) is een fluorescerende acidotrope sonde die wordt gebruikt voor het labelen en volgen van zure organellen in levende cellen. Deze kleurstof vertoont een hoge selectiviteit voor zure organellen en kan levende cellen effectief labelen in nanomolaire concentraties24.
De procedures voor het gebruik van deze fluorescerende kleurstoffen in levende cellen, inclusief het laden van de kleurstoffen en de visualisatie van mitochondriën en lysosomen, worden hier gepresenteerd. Deze methode kan onderzoekers helpen mitofagie te observeren met behulp van live-cell fluorescerende microscopie. Het kan ook worden gebruikt om mitochondriën en lysosomen te kwantificeren en mitochondriale morfologie te beoordelen.

Mitochondriën, die de krachtpatsers van de cel zijn, spelen een belangrijke rol in de bio-energetica, het genereren van vrije radicalen, calciumhomeostase en apoptose. Mitofagie is het primaire mechanisme van mitochondriale kwaliteitscontrole en wordt over het algemeen bestudeerd met behulp van microscopische observatie, maar in vivo mitofagie-assays zijn moeilijk uit te voeren. Het evalueren van mitofagie door levende organellen in beeld te brengen is een alternatieve en noodzakelijke methode voor mitochondriaal onderzoek. Dit protocol beschrijft de procedures voor het gebruik van de celpermeant groen-fluorescerende mitochondriënkleurstof MitoTracker Green en de rood-fluorescerende lysosoomkleurstof LysoTracker Red in levende cellen, inclusief het laden van de kleurstoffen, visualisatie van de mitochondriën en het lysosoom, en verwachte resultaten. Gedetailleerde stappen voor de evaluatie van mitofagie in levende cellen, evenals technische opmerkingen over microscoopsoftware-instellingen, worden ook verstrekt. Deze methode kan onderzoekers helpen mitofagie te observeren met behulp van live-cell fluorescerende microscopie. Bovendien kan het worden gebruikt om mitochondriën en lysosomen te kwantificeren en mitochondriale morfologie te beoordelen.

MitoTracker Green is een groen-fluorescerende mitochondriale vlek die in staat is om nauwkeurig te lokaliseren naar mitochondriën. Deze methode helpt bij het observeren van mitofagie in levende cellen met behulp van een celpermeant groen-fluorescerende mitochondriënkleurstof en een rood-fluorescerende lysosoomkleurstof. Mitofagie speelt een belangrijke rol bij het handhaven van mitochondriale homeostase en verschillende aspecten van de cellulaire functie. Deze techniek zou een nieuwe benadering kunnen bieden voor de behandeling van mitochondriën-gerelateerde ziekten.Deze procedure is niet erg ingewikkeld. Het vastleggen van duidelijke beelden is erg belangrijk voor het tellen van de overlay van mitochondriën en lysosomen. Om te beginnen kweek muis embryonale fibroblast of MEF-cellen in een celkweekschaal van 10 centimeter met 10 milliliter Dulbecco's Modified Eagle Medium of DMEM.Incubeer bij 37 graden Celsius en 5% koolstofdioxide en controleer de cellen onder een microscoop bij 100X vergroting. Wanneer de cellen 80 tot 90% samenvloeiing bereiken, was ze dan met twee milliliter dulbecco's fosfaat gebufferde zoutoplossing. Voeg vervolgens twee milliliter 0,05% trypsine EDTA toe gedurende één minuut om de cellen te dissociëren, gevolgd door twee milliliter DMEM om de reactie te stoppen.Centrifugeer de celsuspensie bij 100 G gedurende drie minuten en resuspenseer de pellet in één milliliter DMEM. Tel de cellen met behulp van een geautomatiseerde celteller en celtelkamerglaasjes en ent 1,5 keer 10 tot de zesde cellen in een nieuwe celkweekschaal van 10 centimeter met 10 milliliter DMEM. Bereid voor de mitofagietest een celsuspensie zoals beschreven en verdun de celsuspensie tot één keer 10 tot de vijfde cellen per milliliter verse DMEM.Voeg twee milliliter van de verdunde celsuspensie toe aan een confocale schaal van 20 millimeter en schud de schaal in een kruis. Incubeer bij 37 graden Celsius en 5% koolstofdioxide gedurende 24 uur. Bereid werkoplossingen van de kleurstoffen voor door de voorraadoplossingen te verdunnen.Voeg twee microliter elk van 0,2 millimolaire mitochondriale kleurstof en 50 micromolaire lysosoomkleurstof toe aan twee milliliter DMEM om een werkconcentratie van 0,2 micromolaire mitochondriale kleurstof en 50 nanomolaire lysosoomkleurstof te verkrijgen. Verwijder het medium uit de confocale kweekschaal en voeg een milliliter van de kleuringsoplossing toe om de cellen te bedekken. Zet de celkweekschaal op 37 graden Celsius en 5% koolstofdioxide gedurende 20 tot 30 minuten.Stel de parameters van de confocale microscopie beeldvormingssoftware in. Gebruik voor dubbele excitatiebeelden sequentiële excitatie op 488 nanometer en 543 nanometer en verzamel emissie op respectievelijk 505 tot 545 nanometer en meer dan 560 nanometer. Verwijder de kweekmediumschaal met de kleurstof uit de broedmachine en voeg een milliliter Krebs-Henseleit of KH-buffer toe aan het gerecht.Om mitofagie te induceren, behandelt u de cellen met FCCP op één micromolair in KH-buffer gedurende 10 minuten bij kamertemperatuur en gaat u onmiddellijk verder met het in beeld brengen van de cellen met behulp van de confocale microscoop. Breng een geschikte hoeveelheid olie aan op de bovenkant van de 63X olielens. Plaats het monster op het monsterstadium van de confocale microscoop en beweeg het direct boven de objectieflens.Gebruik de beeldbewerkingssoftware om het voorbeeld te vinden door op het tabblad Zoeken in de linkerbovenhoek van de software-interface te klikken. Selecteer een groene filterset voor het experiment. Gebruik de grove instelknop om snel scherp te stellen door de objectieflens op en neer te bewegen.Nadat het celmonster duidelijk zichtbaar is door het oculair, zoekt en focust u het gebied van afzonderlijke cellen en verplaatst u het naar het midden van het gezichtsveld. Klik op het tabblad Acquisitie in de linkerbovenhoek van de software-interface om afbeeldingen te verkrijgen. Selecteer alleen het kanaal van 488 nanometer en de frameresolutie 1024 bij 1024 voor een voorbeeld.Klik op het tabblad Live in de linkerbovenhoek om een live scan te starten. Pas het gezichtsveld aan op het scherpst en pas het laservermogen aan door de schuifregelaar naar links of rechts te bewegen. Houd de versterkingsinstelling onder de 600 om overmatige blootstelling te voorkomen.Stel de pinhole-waarde in op 156, de winstwaarde op 545 en de digitale offsetwaarde op nul. Selecteer het beste gezichtsveld, controleer de twee kanalen en kies de frameresolutie 1024 bij 1024. Klik op Uitlijnen om 2D-afbeeldingen te verkrijgen en de verkregen afbeeldingen op te slaan.In deze studie werd FFCP gebruikt om mitofagie in MEF-cellen te activeren voor confocale beeldvorming. Representatieve afbeeldingen van cellen gekleurd met de groen-fluorescerende mitochondriënkleurstof met mitochondriën en gekleurd met de rood-fluorescerende lysosoomkleurstof die lysosoom vertoont, worden hier weergegeven. Wanneer beschadigde groengekleurde mitochondriën worden overspoeld door roodgekleurde lysosomen, overlappen de groene en rode fluorescentie elkaar om gele co-gelokaliseerde mitochondriën lysosomen te onthullen.De gele stippen komen overeen met deze co-gelokaliseerde mitochondriale lysosomen die voortdurende mitofagie vertegenwoordigen en kunnen dus worden geteld om de mate van mitofagie te evalueren. Voorbereiding van een werkoplossing van kleurstoffen voor mitochondriën en lysosoomkleuring en het handhaven van de juiste celconfluentie bij beeldvorming met confocale microscopie zijn belangrijke stappen om te onthouden. Deze procedure kan ook worden gebruikt om mitochondriaal getal en morfologie te analyseren, wat belangrijk is voor het beoordelen van mitochondriale dynamiek.Mitofagie is nauw verbonden met een verscheidenheid aan menselijke ziekten en deze techniek kan worden gebruikt om het verband tussen mitofagie en menselijke ziekten te onderzoeken.
