Microfluïdische poreuze media-analogen in combinatie met druksensoren bieden een geschikt hulpmiddel om de ontwikkeling van biofilms in poreuze media te bestuderen. De veelzijdigheid in het ontwerp van het microfluïdische poreuze medium, met name de opstelling van de pilaren, inclusief diameter, onregelmatige vormen en poriegrootte, maakt het mogelijk om vele geometrieën te onderzoeken. Biofilms - bacteriekolonies ingebed in een zelfuitgescheiden matrix van extrapolymere stoffen (EPS) - zijn alomtegenwoordig in natuurlijke poreuze media, zoals bodems en watervoerende lagen1, en technische en medische toepassingen, zoals bioremediatie2, waterfiltratie3 en medische hulpmiddelen4. De biofilmmatrix bestaat uit polysacchariden, eiwitvezels en extracellulair DNA5,6 en is sterk afhankelijk van de micro-organismen, de beschikbaarheid van voedingsstoffen en de omgevingscondities7. Toch zijn de functies van de matrix universeel; Het vormt de steiger van de biofilmstructuur, beschermt de microbiële gemeenschap tegen mechanische en chemische spanningen en is grotendeels verantwoordelijk voor de reologische eigenschappen van de biofilms5.
In poreuze media kan de groei van biofilms de poriën verstoppen, waardoor de zogenaamde bioclogging ontstaat. De ontwikkeling van biofilms wordt geregeld door de vloeistofstroom en poriegrootte, gedefinieerd als de afstand tussen twee pilaren, van het poreuze medium 8,9,10. Zowel de poriegrootte als de vloeistofstroom regelen het transport van voedingsstoffen en lokale schuifkrachten. Op zijn beurt verstopt de groeiende biofilm de poriën, wat de snelheidsverdeling van de vloeistof 11,12,13, het massatransport en de hydraulische geleidbaarheid van het poreuze medium beïnvloedt 14,15. De veranderingen in hydraulische geleidbaarheid worden weerspiegeld door verhoogde druk in gesloten systemen16,17,18,19. Huidige microfluïdische studies in biofilmontwikkeling en bioclogging richten zich op het bestuderen van de impact van stroomsnelheden in homogene geometrieën16,20 (d.w.z. met een enkelvoudige poriegrootte) of heterogene poreuze media12,21,22. Om echter de effecten van stroomsnelheden en poriegrootte op de ontwikkeling van biofilms en de daaruit voortvloeiende drukveranderingen in het bioclogeerde poreuze medium te ontwarren, is een zeer controleerbaar en veelzijdig experimenteel platform nodig dat de studie van verschillende poreuze mediageometrieën en omgevingscondities parallel mogelijk maakt.
De huidige studie introduceert een microfluïdisch platform dat drukmetingen combineert met gelijktijdige beeldvorming van de evoluerende biofilm in het poreuze medium. Vanwege de gasdoorlaatbaarheid, biocompatibiliteit en flexibiliteit in het kanaalgeometrieontwerp, is een microfluïdisch apparaat gemaakt van polydimethylsiloxaan (PDMS) een geschikt hulpmiddel voor het bestuderen van biofilmontwikkeling in poreuze media. Microfluïdica maken het mogelijk om de fysische en chemische omstandigheden (bijv. vloeistofstroom en nutriëntenconcentratie) met hoge precisie te beheersen om de omgeving van microbiële habitats na te bootsen23. Verder kunnen microfluïdische apparaten eenvoudig worden afgebeeld met micrometrische resolutie met behulp van een optische microscoop en gekoppeld aan online metingen (bijvoorbeeld de lokale druk).
In dit werk richten de experimenten zich op het bestuderen van de impact van poriegrootte in een homogeen poreus medium analoog onder gecontroleerde opgelegde stromingsomstandigheden. De stroom van een kweekmedium wordt opgelegd met behulp van een spuitpomp en het drukverschil door het microfluïdische kanaal wordt gelijktijdig gemeten met druksensoren. Biofilmontwikkeling wordt geïnitieerd door het zaaien van een planktoncultuur van Bacillus subtilis in het microfluïdische kanaal. Regelmatige beeldvorming van de evoluerende biofilm en beeldanalyse maakt het mogelijk om porieschaal opgeloste informatie over de oppervlaktedekking onder verschillende experimentele omstandigheden te verkrijgen. De gecorreleerde informatie over drukverandering en de mate van bioclogging levert cruciale input voor permeabiliteitsschattingen van bioclogged poreuze media.

a microfluidic platform to study bioclogging in porous media

Bacteriële biofilms worden aangetroffen in verschillende milieu- en industriële poreuze media, waaronder bodems en filtratiemembranen. Biofilms groeien onder bepaalde stromingsomstandigheden en kunnen de poriën verstoppen, waardoor de lokale vloeistofstroom wordt omgeleid. Het vermogen van biofilms om poriën te verstoppen, de zogenaamde bioclogging, kan een enorm effect hebben op de lokale permeabiliteit van het poreuze medium, waardoor een drukopbouw in het systeem ontstaat en de massastroom erdoorheen wordt beïnvloed. Om de wisselwerking tussen biofilmgroei en vloeistofstroom onder verschillende fysieke omstandigheden (bijvoorbeeld bij verschillende stroomsnelheden en poriegroottes) te begrijpen, wordt in deze studie een microfluïdisch platform ontwikkeld om de ontwikkeling van biofilms te visualiseren met behulp van een microscoop onder extern opgelegde, gecontroleerde fysieke omstandigheden. De door de biofilm geïnduceerde drukopbouw in het poreuze medium kan gelijktijdig worden gemeten met behulp van druksensoren en later worden gecorreleerd met de oppervlaktedekking van de biofilm. Het gepresenteerde platform biedt een basis voor een systematische aanpak om bioclogging veroorzaakt door biofilms in poreuze media onder stromingsomstandigheden te onderzoeken en kan worden aangepast aan het bestuderen van omgevingsisolaten of biofilms met meerdere soorten.

Voor deze studie werd een microfluïdisch apparaat met drie parallelle microfluïdische kanalen met verschillende poriegroottes gebruikt (figuur 1) om biofilmvorming in poreuze media systematisch te bestuderen. A Microfluidic Platform to Study Bioclogging in Porous Media

Deze methode maakt het mogelijk om systematisch de invloed van poriegrootte en stroomsnelheid op de ontwikkeling van biofilms in zowel natuurlijke als kunstmatige poreuze media te bestuderen, waardoor het fundamentele mechanisme van poreuze mediabioclogging kan worden ontrafeld. De belangrijkste voordelen van de technieken zijn de hoogste ruimtelijke en temporele resolutie van de wijziging en het aanpassingsvermogen van het platform aan een reeks experimentele omstandigheden en vereisten. Begin met het inschakelen van de boxincubator van de microscoop drie uur voor het experiment om een stabiele temperatuur van 25 graden Celsius te garanderen.Sluit de inlaat- en uitlaatslang aan op het microfluïdische apparaat. Bevestig de verbinding tussen de slang en de spuit door een naald met een buitendiameter van 0,6 millimeter in de inlaatslang te steken. Plaats het microfluïdische apparaat, 30 milliliter gedeïoniseerd water en 30 milliliter kweekmedium in een vacuümexsiccator en ontgast ze gedurende ten minste een uur.Als u klaar bent, trekt u het kweekmedium en het gedeïoniseerde water langzaam in twee afzonderlijke spuiten van 30 milliliter. Monteer vervolgens het microfluïdische apparaat op de microscoop en plaats de uitlaatslang in een afvalcontainer. Sluit de spuit gevuld met gedeïoniseerd water aan op het microfluïdische kanaal via de microfluïdische slang en injecteer het water langzaam totdat het uit de uitlaat van de druksensor komt.Vul de druksensor met water en spoel alle bellen uit de slang die het microfluïdische kanaal en de druksensor verbindt. Sluit de uitlaat van de druksensor met de schroeven die aan de druksensor zijn gewijd. Vul de rest van het microfluïdische kanaal met gedeïoniseerd water.Plaats vervolgens een filter van 1,2 micrometer op de kweekmediaspuit. Verwijder de waterspuit en sluit de spuit voorzichtig aan op de microfluïdische slang van de inlaat. Nadat u de spuit op de spuitpomp hebt gemonteerd, spoelt u het kanaal gedurende één uur met het kweekmedium met een stroomsnelheid van twee milliliter per uur.Stel daarna de spuitpomp tijdens het experiment in op het gewenste debiet en stel de drukmeting van de druksensoren in op nul. Vervolgens pipetteer je één milliliter van de Bacillus subtilis NCIB 3610-cultuur met een optische dichtheid van 0,1 bij een golflengte van 600 nanometer in een injectieflacon met centrifuges van 1,5 milliliter. Om de bacteriecultuur in het microfluïdische kanaal te laden, plaatst u de uitlaatbuis in de injectieflacon met centrifuge en wacht u vijf minuten om eventuele luchtbellen uit de uitlaat van de buis te verwijderen.Als u klaar bent, trekt u 150 microliter bacteriële oplossing op met een stroomsnelheid van één milliliter per uur totdat het microfluïdische kanaal is gevuld met de bacteriecultuur. Verwijder vervolgens voorzichtig het filter van de kweekmediaspuit en plaats de uitlaat in de afvalcontainer. Laat de bacteriële cellen drie uur lang op nulstroomcondities in het microfluïdische kanaal om hun oppervlakteaanhechting in het poreuze medium mogelijk te maken.Om het experiment te starten, start u de stroom door de spuitpomp in te stellen op het gewenste debiet en start u de drukmeting op één hertz voordat u de beelden van de groeiende biofilms met het gewenste tijdsinterval, optische configuratie en vergroting verkrijgt. Het biofilmvormingsproces werd gevisualiseerd met behulp van bright-field microscopie, waarbij de bacteriële cellen en de biofilm als donkere pixels in de beelden verschenen. Tijdens een 24-uurs experiment koloniseerde de aanvankelijk willekeurig groeiende biofilm bijna het hele poreuze medium.De oppervlaktedekking van de biofilm vond 10% sneller plaats bij de kleinste poriegrootte dan bij de grootste poriegrootte na 20 uur. Correlatie van de dekking van het biofilmoppervlak met de drukopbouw toonde aan dat de verstopping in het microfluïdische kanaal met kleinere poriegrootte leidde tot een hoger drukverschil tussen de inlaat en de uitlaat dan in de grotere poriegrootte. De belangrijkste aspecten om te onthouden zijn om het experiment uit te voeren in een temperatuurstabiele omgeving en om het microfluïdische apparaat en de oplossingen goed te ontgassen om bubbelvorming te voorkomen.Deze methode maakt systematische studies mogelijk om fundamentele mechanismen van biofilmgroei in zowel industriële als natuurlijke poreuze media te ontrafelen met betrekking tot de invloed van stroomsnelheid en poriegrootte.
