外傷性脳損傷(TBI)は、スポーツ傷害、車両衝突、軍事戦闘に起因する可能性があり、世界的な健康上の懸念事項です。TBIは、生理学的、認知的、行動的障害、および生涯にわたる障害または死亡率につながる可能性があります1,2。TBIの重症度は、軽度、中等度、重度に分類でき、大多数は軽度のTBI(75%-90%)です3。TBI、特にTBIの反復発生は、神経変性を促進し、アルツハイマー病(AD)、筋萎縮性側索硬化症(ALS)、前頭側頭型認知症(FTD)、慢性外傷性脳症(CTE)などのいくつかの神経変性疾患の危険因子として機能する可能性があることがますます認識されています4,5,6.しかし、TBI誘発性神経変性の根底にある分子メカニズムは依然として不明であり、したがって活発な研究分野を表しています。ニューロンがTBIに応答し、TBIから回復する方法についての洞察を得るために、TBI後のマウスにおける縦断的生体内イメージングによって、特にニューロン内で目的の蛍光タグ付きタンパク質をモニタリングする方法を本明細書に記載する。

intravital imaging of fluorescent protein expression in mice with a closed-skull traumatic brain injury and cranial window using a two-photon microscope

私たちのプロトコルは、生きたマウスの機械的ストレスに対するニューロンの応答を視覚化する方法を提供し、外傷性脳損傷が脳にどのように影響するかの直接的な証拠を提供することができます。この技術は、外傷性脳損傷後の急性期と慢性期の両方について、同じ動物の同じ脳位置にある標的タンパク質をモニターすることができます。目的の遺伝子とウイルスプロモーターを適宜変更して、脳内の特定の細胞タイプの他のタンパク質を研究することができます。まず、麻酔をかけたマウスの目に眼科用軟膏を塗ります。通常のハサミでトリミングして、頭のてっぺんから髪の毛を取り除きます。長さ12〜15ミリメートルの正中線切開を確立した後、湾曲した先端のスプリングハサミを使用して頭蓋骨の左右の半球の皮膚を切除します。頭蓋骨が露出したら、滅菌綿の先端アプリケーターでそっとこすり、滅菌生理食塩水で洗い流して骨膜を取り除きます。頭蓋骨領域が乾燥したら、ブレグマの後方2.5ミリメートル、矢状縫合糸から右側の2ミリメートルの座標に外傷性脳損傷(TBI)の衝撃部位をマークします。マウスを脳定位固定装置フレームからすばやく取り外し、TBIデバイスの下のバッファークッションに頭を置きます。インパクターの先端をマークされた衝撃部位に合わせます。繋がれたナイロン製の紐をマウスの頭から15センチ上に引っ張って金属柱を持ち上げてから放し、TBI部位の頭蓋骨の上部に接触しているトランスデューサーロッドに重りが自由に落ちるようにします。麻酔をかけたマウスの頭を、手術用顕微鏡下に保管されている脳定位固定装置フレームに置きます。FG4カーバイドビットを使用して頭蓋骨の表面をゆっくりとゆっくりとローター速度でドリルで穴を開け、残りの骨膜を取り除き、歯科用セメントが頭蓋骨としっかりと結合するように粗い頭蓋骨表面を作成します。生理食塩水を使用して、頭蓋骨の表面から骨のほこりを洗い流して取り除きます。頭頂骨と側頭蓋骨をつなぐ縫合糸がある眼の後約5ミリメートルで、外側の筋肉を頭蓋骨から分離するには、細かい閉じた先端をそっと挿入し、閉じた先端をラムドイド縫合糸まで後方向に静かに動かします。頭蓋窓の移植が行われる側の外側の筋肉を分離します。生理食塩水を使用して手術部位の破片を洗い流し、ガーゼでその部分を乾かします。ヘッドポストを埋め込むには、マーカーペンと外科用ノギスを使用して、ブレグマの後方2.5ミリメートル、右半球の矢状縫合糸から1.5ミリメートル離れた中心点に印を付けます。次に、清潔で乾燥した頭蓋骨の開頭術の周囲をトレースします。イヤーバーを緩め、頭を回転させて、開頭面が完全に水平になるようにしてから、イヤーバーを再度締めます。先端が綿のアプリケーターの木製スティックを使用して、ヘッドポストの前端と後端に瞬間接着剤を2滴加えます。チタン製のヘッドポストを開頭術の中心に配置し、頭蓋窓が埋め込まれるのと同じ平面内に収まるようにすばやく調整します。瞬間接着剤が乾くまで軽く圧力をかけますが、通常は約30秒かかります。300ミリグラムのセメント粉末、6滴のQuickBase液体、および1滴の触媒を完全に混合することにより、予冷したセラミック皿で歯科用セメントを調製します。トレースされた円周の外周にたっぷりの量の歯科用セメント混合物をすばやく塗布し、露出した骨表面を覆います。ただし、開頭部位は覆わないでください。イヤーバーを外し、ヘッドポストを金属フレームに固定して、頭が安定し、マークされた開頭術の円周に沿って正確に穴を開けることができるようにします。開頭手術を行うには、外科用ノギスを使用して、前に示したようにマークされた円の直径を確認し、必要に応じて調整して、頭蓋窓が開頭術の内側にぴったりと収まるようにします。電動歯科用ドリルを使用して、最初にFG4カーバイドビットを使用して、マークされた円の外側に沿って頭蓋骨をエッチングして薄くし、頭蓋骨を薄くするためのトラックを作成します。その部分を生理食塩水で灌漑して、骨の粉を洗い流します。頭蓋骨が紙のように薄く透明になるまで、FG 1/4カーバイドビットを使用して頭蓋骨を薄くし続けます。定期的に掘削を中止し、ドリルからの加熱を減らし、骨粉を洗い流すために、滅菌生理食塩水で領域全体を再度灌漑します。EF4カーバイドビットを使用して頭蓋骨の薄化を完了し、トラックに沿って頭蓋骨の残りの部分の薄肉化と穴あけを続けます。ひびの入った場所に0.5ミリの細い先端の鉗子を挿入し、下にある脳をへこませることなく、骨皮弁をゆっくりと上に持ち上げます。骨皮弁を除去した後、開頭部位を生理食塩水で洗浄します。湾曲した先端の外科用鉗子を使用して、目に見えるくも膜をそっと取り除きます。先端がまっすぐな手術用鉗子を使用して、3ミリメートルのガラスカバースリップを下に向けて滅菌ガラス窓を持ち上げます。開頭部位の上にガラス窓を配置して調整し、窓が開頭部の端にぴったりと収まるようにします。5ミリガラスのカバースリップが上部にあります。前に示したように歯科用セメントを準備し、セメントがペースト状で厚くなるまで約6分間待ちます。セメントがペースト状で厚くなるのを待っている間に、脳定位固定装置マニピュレーターを介して窓に十分な量の圧力をかけ、頭蓋骨がガラス窓にしっかりとしっかりと接触することを確認します。調整可能な精密アプリケーターブラシを使用して、窓の端に沿って少量のセメントを追加し、ガラス窓を頭蓋骨で密閉します。約10分待ってセメントを完全に乾かしてから、静かに解放し、窓の上のマニピュレーターを取り外します。余分なセメントが窓を覆っている場合は、FG4カーバイドビット付きの歯科用ドリルを使用して歯科用セメントをトリミングして完了します。術後約4時間で2光子イメージングを行い、表在性では血管系が黒く見え、約400マイクロメートルの深部ではEGFPタンパク質の発現が細胞体全体にびまん性に分布していました。TBIの1週間後と4か月後に、0日目の時点で観察された血管系パターンを基準として使用して、同じ画像領域を特定しました。血管系パターンは0日目のパターンと似ています。同様に、EGFP発現は細胞体全体で検出されました。0日目と4か月の蛍光強度は、表層レベルと深部レベルの両方で類似していました。しかし、1週間後の蛍光強度は、0日目および4か月の蛍光強度よりも低く、おそらく他のTBIモデルで報告された翻訳抑制が原因でした。開頭手術を行う際には、脳組織を傷つけないように細心の注意を払うことが重要です。ドリルの先端を脳に挿入しないでください。この技術を用いることで、研究者は、TBI後の異なる段階において、同じ細胞内の異なるタンパク質を縦断的に可視化し、外傷後の哺乳類脳におけるそのタンパク質の局在、発現、溶解性に関する情報を提供することができます。
