ナノ粒子の抗菌活性はここ数十年で広く研究され報告されてきましたが、研究間の直接比較を可能にするために使用される材料と研究方法の基準はありません。このため、直接共培養法(A法)と直接暴露法(B法)の2つの方法を提示し、材料と方法の一貫性を保ちながらナノ粒子の抗菌活性を特徴付け、比較します。
ナノ粒子に加えて、ナノ構造表面も抗菌活性について調べられています。これらには、単層カーボンナノシート、カーボンナノチューブ、グラファイト29などの炭素ベースの材料、ならびに純粋なMgおよびMg合金が含まれる。これらの各材料は、炭素系材料によって細胞膜に課せられる物理的損傷や、Mgが分解する際の活性酸素種(ROS)の放出による代謝過程またはDNAの損傷など、少なくとも1つの抗菌メカニズムを示しています。さらに、亜鉛(Zn)とカルシウム(Ca)を組み合わせてMg合金を形成すると、Mgマトリックスの粒径の微細化が促進され、Mgのみのサンプル14と比較して、基板表面への細菌接着が減少します。抗菌活性を実証するために、直接的および間接的な表面接触を伴う細菌コロニー形成単位(CFU)の定量を通じて、ナノ構造材料上およびナノ構造材料周辺の細菌接着を経時的に決定する直接培養法(方法C)を紹介します。
サイズ、形状、配向など、表面上のナノ構造の形状は、材料の殺菌活性に影響を与える可能性があります。例えば、Linら16 は、陽極酸化および電気泳動堆積(EPD)によって、Mg基板の表面に異なるナノ構造MgO層を作製した。 in vitroでナノ構造表面への一定期間の曝露後、 黄色ブドウ球菌 の成長は、未処理のMgと比較して大幅に減少した。これは、未処理の金属Mg表面に対する細菌接着に対するナノ構造表面のより大きな効力を示した。本稿では、様々なナノ構造表面の抗菌特性のさまざまなメカニズムを明らかにするために、対象領域内の細胞表面相互作用を決定する集束接触露光法(方法D)について説明します。
この記事の目的は、さまざまなナノ粒子、ナノ構造表面、および微生物種に適用できる4つのin vitroメソッドを提示することです。比較可能性のために一貫性のある再現性のあるデータを生成するための各方法の主な考慮事項について説明します。具体的には、ナノ粒子の抗菌性を調べるために、直接共培養法17および直接露光法が用いられる。直接共培養法により、最小発育阻止濃度と最小殺菌濃度(それぞれMICとMBC90-99.99)を個々の種について決定でき、最も強力な濃度(MPC)を複数の種について決定できます。直接暴露法により、最小阻害濃度でのナノ粒子の静菌または殺菌効果を、経時的なリアルタイムの光学濃度測定値によって特徴付けることができます。直接培養14法は、ナノ構造表面に直接的および間接的に接触している細菌を検査するのに適している。最後に、細菌の直接適用と細胞-ナノ構造界面での細菌増殖の特性評価を通じて、ナノ構造表面上の特定の領域の抗菌活性を調べるために、集束接触曝露16法を提示します。この方法は、日本工業規格JIS Z 2801:200016から修正されたもので、微生物と表面の相互作用に着目し、微生物培養におけるバルク試料分解が抗菌活性に及ぼす影響を排除することを目的としています。
