많은 연구에도 불구하고 생체 외 HSC를 안정적으로 확장하는 것은 여전히 어려운 과제입니다 7. 우리는 최근 마우스 HSC를 위한 최초의 장기 생체 외 확장 배양 시스템을 개발했습니다8. 이 접근법은 4주간의 배양 기간 동안 이식 가능한 HSC를 234-899배까지 확장할 수 있습니다. 대체 접근법과 비교하여 프로토콜의 주요 변경 사항은 혈청 알부민을 제거하고 합성 고분자로 대체하는 것이었습니다. 폴리비닐알코올(Polyvinyl alcohol, PVA)은 마우스 HSC 배양을 위한 최적의 고분자로 확인되었다8, 현재는 다른 조혈 세포 유형9 배양에도 사용되고 있다. 그러나, Soluplus(폴리비닐 카프로락탐-아세테이트-폴리에틸렌 글리콜 그라프트 공중합체)라고 불리는 또 다른 중합체도 최근에 확인되었는데, 이는 클론 HSC 확장을 개선하는 것으로 보인다10. 중합체를 사용하기 전에는 소 태아 혈청, 소 혈청 알부민 분획 V 또는 재조합 혈청 알부민 형태의 혈청 알부민이 사용되었으나, 이들은 HSC 확장에 대한 지원이 제한적이었고 단기간(~1주) 생체 외 배양만 지원했다7. 그러나, HSC를 정지 상태로 유지하는 HSC 배양 프로토콜은 더 긴 생체외 배양 시간을 지원할 수 있다는 점에 유의해야 한다(11,12).
