세포 분류 회로를 개발하는 데 가장 시간이 많이 소요되는 단계 중 하나는 회로 내에서 센서 및 처리 인자와 같은 개별 구성 요소 유전자의 상대적 발현 수준을 최적화하는 것입니다. 회로 최적화를 가속화하고 보다 정교한 회로의 구축을 허용하기 위해, 최근 연구에서는 최적의 조성 및 토폴로지를 예측하기 위해 셀 분류기 회로 및 그 구성 요소의 수학적 모델링을 사용했습니다(6,7). 이것은 지금까지 강력한 결과를 보여주었지만 수학적 분석은 회로에서 구성 요소 유전자의 입출력 거동을 체계적으로 특성화해야 하는 필요성으로 인해 시간이 많이 걸리기 때문에 제한적입니다. 또한, 복잡한 유전 회로에서 무수히 많은 문맥 의존적 문제가 나타날 수 있으며, 이로 인해 전체 회로의 동작이 개별 부분 특성화에 기초한 예측을 무시하게 됩니다(8,9).
세포 상태 분류기와 같은 복잡한 포유류 회로를 보다 신속하게 개발하고 테스트하기 위해 우리 연구실은 플라스미드 공동 형질주입 프로토콜의 진화인 폴리 형질감염10이라는 기술을 개발했습니다. 동시 형질감염에서 여러 플라스미드 DNA 종은 양전하를 띤 지질 또는 고분자 시약과 함께 복합체화된 다음 상관관계가 있는 방식으로 세포에 전달됩니다(그림 1A). 폴리형질감염에서, 플라스미드는 시약과 개별적으로 복합체화되어, 각각의 형질주입 복합체로부터의 DNA가 비상관된 방식으로 세포에 전달된다 (도 1B). 이 방법을 사용하여, 형질주입된 집단 내의 세포는 상이한 회로 성분을 운반하는 2개 이상의 DNA 페이로드 비율의 수많은 조합에 노출된다.

여기서는 유전자 회로를 신속하게 최적화하기 위해 폴리 트랜스펙션을 사용하는 워크플로우를 설명하고 묘사합니다(그림 2). 이 프로토콜은 고품질 poly-transfection 데이터를 생성하고 poly-transfection 프로토콜 및 데이터 분석에서 몇 가지 일반적인 오류를 방지하는 방법을 보여줍니다(그림 3). 그런 다음 poly-transfection을 사용하여 간단한 회로 구성 요소를 특성화하고 그 과정에서 co-transfection에 대한 poly-transfection 결과를 벤치마킹하는 방법을 보여줍니다(그림 4). 마지막으로, 폴리형질감염의 결과는 암 분류자 회로의 최적화를 보여준다(도 5).
