Mens neuroner kan studeres i cellekultur og eksplanter, fjerner det dem fra deres normale fysiologiske kontekst. Primære sensoriske neuroner innerverer huden direkte og bærer somatosensorisk information tilbage til centralnervesystemet 1,2. Dorsalrodsganglier (DRG'er) er cellekropsklynger på 10.000-15.000 primære sensoriske neuroner 3,4. DRG-neuroner præsenterer forskellig størrelse, myelineringsniveauer og gen- og receptorekspressionsmønstre. Neuroner med mindre diameter inkluderer smertefølende neuroner, og neuroner med større diameter reagerer typisk på ikke-smertefulde mekaniske stimuli 5,6. Forstyrrelser i de primære sensoriske neuroner såsom skade, kronisk inflammation og perifere neuropatier kan sensibilisere disse neuroner til forskellige stimuli og bidrage til kronisk smerte, allodyni og smerteoverfølsomhed 7,8. Derfor er undersøgelsen af DRG-neuroner vigtig for at forstå både somatosensation generelt og mange smerte- og kløeforstyrrelser.
Neuroner, der affyres in vivo, er afgørende for somatosensation, men indtil for nylig har værktøjer til at studere intakte ganglier in vivo været begrænset til et relativt lille antal celler 9. Her beskriver vi en kraftfuld metode til at studere neuronernes handlingspotentialer eller aktiviteter på populationsniveau in vivo som et ensemble. Metoden anvender billeddannelse baseret på cytoplasmatisk Ca2+ dynamik. Ca 2+ følsomme fluorescerende indikatorer er gode proxyer til måling af cellulær aktivitet på grund af den normalt lave koncentration af cytoplasmatisk Ca2+. Disse indikatorer har muliggjort samtidig overvågning af hundreder til flere tusinde primære sensoriske neuroner i mus 9,10,11,12,13,14,15,16 og rotter 17. Metoden til in vivo Ca2+ billeddannelse beskrevet i denne undersøgelse kan bruges til direkte at observere reaktioner på populationsniveau på mekaniske, kolde, termiske og kemiske stimuli.
Det phosphoinositidbindende membranprotein, Pirt, udtrykkes i høje niveauer i næsten alle (>95%) primære sensoriske neuroner18,19 og kan bruges til at drive ekspressionen af Ca 2+ sensoren, GCaMP3, til at overvåge neuronaktivitet in vivo20. in vivo calcium imaging of neuronal ensembles in networks of primary sensory neurons in intact dorsal root ganglia

Ca 2+ billeddannelse kan bruges som en proxy for cellulær aktivitet, herunder handlingspotentialer og forskellige signalmekanismer, der involverer Ca 2+ indgang i cytoplasmaet eller frigivelse af intracellulære Ca2+ lagre. Pirt-GCaMP3-baseret Ca2+ billeddannelse af primære sensoriske neuroner i dorsalrodsganglion (DRG) hos mus giver fordelen ved samtidig måling af et stort antal celler. Op til 1.800 neuroner kan overvåges, hvilket gør det muligt at studere neuronale netværk og somatosensoriske processer som et ensemble i deres normale fysiologiske kontekst på populationsniveau in vivo. Det store antal neuroner, der overvåges, gør det muligt at detektere aktivitetsmønstre, der ville være udfordrende at opdage ved hjælp af andre metoder. Antallet af neuroner, der overvåges ved hjælp af denne metode, tillader indsamling af neuronnetværksdata, der ville være umulige ved hjælp af ældre eller andre metoder. Den største fordel ved denne teknik er evnen til at overvåge næsten 2.000 primære sensoriske neuroner på én gang og reagere direkte på stimuli påført bagpoten. Brug derefter otte millimeter saks til at skære musklen og bindevævet væk for at rense den tværgående proces på højre side L5 DRG. Prøv at minimere blødning ved at bruge bomuld eller gelfoam til at absorbere blodet.Skær højre side L5 tværgående proces op ved hjælp af Friedman-Pearson rongeurs eller stærke fine tang, pas på ikke at røre DRG. Flyt derefter musen og varmepuden til den brugerdefinerede scene. Brug scenetape til at fastgøre dyret og varmepuden på plads.Placer dyrets næse i næsekeglen til kontinuerlig isofluranbedøvelse. Fastgør den højre bagpote, der stikker ud til en position uden for scenen for nem anvendelse af stimuli. Fastgør rygsøjlen på plads med sceneklemmerne over huden på ryghvirvlerne eller bækkenbenet lige rostral og kaudale til L5 DRG.Juster klemmerne og scenen for at gøre DRG'ens overflade så plan som muligt. Placer derefter scenen under mikroskopet, så målet er direkte otte millimeter over DRG, når det sænkes. Indsæt det rektale termometer.Tilslut kraftledningerne til varmepuden og rektaltermometeret. Brug et opretstående konfokalmikroskop med et 10x 0,4 DIC-mål og den tilhørende software til billeddannelse.Brug de grønne FITC-filterindstillinger for excitation ved 495 nanometer, emission ved 519 nanometer og detektionsbølgelængde mellem 500 og 580 nanometer. Under mikroskopet finder du overfladen af DRG. Juster klemmerne på scenen, så DRG-overfladen er så plan som muligt, og det maksimale overfladeareal visualiseres i brændplanet.Dyret skal overvåges under hele proceduren for at opretholde isofluranæstesi uden overdosering. For at indlæse mikroskopets hurtige scanningsprotokol skal du bruge de typiske indstillinger for voxelstørrelse 2.496 ved 2.496 ved 16 mikron, 512 x 512 pixels, 10 optisk skive Z-stack, en Airy-enhed til 32 mikrometer, 1% lasereffekt på 488 nanometer og fem milliwatt, pixeltid 1.52 mikrosekunder, linjetid 0.91 millisekunder, billedtid 465 millisekunder, LSM-scanningshastighed på otte, tovejsscanning, PMT-detektorforstærkning 650 volt og digital forstærkning på en. Tag en kort, otte-cyklus scanning af DRG ved at klikke på Start eksperiment under fanen Erhvervelse.Opret en film ved at lave en ortogonal projektion af scanninger med én scanning pr. billede over tid. Kontroller manuelt for billedklarhed og billedartefakter, såsom bølger af lysstyrke, der krydser DRG. Juster klemmeposition og optisk sektionstykkelse, og gentag dette trin, indtil der opnås en klar film i høj kvalitet.Indlæs derefter mikroskopets scanningsprotokol med høj opløsning ved hjælp af de typiske indstillinger for voxelstørrelse 1.248 x 1.248 x 14 mikron, 1024 x 1024 pixels, seks optiske skiver Z-stack, 1.2 Airy-enhed eller 39 mikrometer, 5% lasereffekt på 488 nanometer og 25 milliwatt, pixeltid 2.06 mikrosekunder, linjetid 4.95 millisekunder, billedtid 5.06 sekunder, LSM-scanningshastighed på seks, tovejsscanning, PMT-detektorforstærkning ved 650 volt og digital forstærkning på en. Klik på knappen Start eksperiment under fanen Erhvervelse for at lave et billede i høj opløsning af DRG. Indlæs mikroskopets hurtigscanningsprotokol, og registrer spontan aktivitet i DRG i 80 cyklusser.Generer en ortogonal projektionsfilm, og kontroller, at billedet er af tilstrækkelig kvalitet til analyse. For at anvende stimuli skal du indstille mikroskopet til at udføre 15 til 20 scanninger. Vent på, at scanninger et til fem er fuldført for at producere grundlinjen.Anvend stimulus under scanninger seks til 10. Vent mindst fem minutter efter hver stimulus, før du anvender den næste for at forhindre desensibilisering. For en mekanisk presse skal du holde algometerets pincher med poten mellem padlerne uden at røre poten.Klem poten med start umiddelbart efter afslutningen af scanning fem og stop umiddelbart efter scanning 10. Overvåg pressekraften med et algometer, og sørg for, at den ikke overstiger 10 g over den ønskede kraft. Til termiske stimuli opvarmes et bægerglas vand til lige over den ønskede temperatur.Når vandet har den korrekte temperatur, skal du anvende stimulus umiddelbart efter scanning fem ved at nedsænke poten i vandet. Træk bægerglasset væk umiddelbart efter scanning 10. Kirurgisk L5 DRG-eksponering efterfulgt af konfokal mikroskopi tillod op til 1.800 neuroner at blive afbildet på én gang ved hjælp af Pirt-GCaMP3-mus.Primære sensoriske neuroner blev observeret i et ensemble på populationsniveau for spontane calciumtransienter i fravær af stimuli og som reaktion på stimuli i deres normale fysiologiske sammenhæng. Stærke stimuli eller skadelig varme øgede calciumresponserne. Sammenlignet med presning med 100 g øgede presning med 300 g antallet af neuroner, der producerede calciumtransienter og delta F ved F0-området under kurven.Varm varme i et ikke-skadeligt temperaturområde på 21 og 45 grader Celsius øgede antallet af neuroner, der producerede calciumtransienter og delta F ved F0-området under kurven. Ikke-skadelige temperaturer aktiverede et mindre antal neuroner, der producerede transienter sammenlignet med en skadelig varmestimulus ved 57 grader Celsius. Endvidere producerede neuroner med mindre og mellemstor diameter calcium forbigående spontant og under alle stimuli, men neuroner med større diameter producerede kun calciumtransienter som reaktion på 300 g pressestimulus.DRG skal være plan og optimal optisk skivetykkelse. Det skal være sådan, at du kan registrere det maksimale antal celler, og der er ingen bølgegang i filmen. Denne teknik er blevet brugt til at analysere sensoriske modaliteter på befolkningsniveau for somatosensation og smag og til at studere tilstødende neuroner, der aktiveres parvis eller i synkroniserede klynger, der bidrager til kronisk og neuropatisk smerte.
