급성 골수성 백혈병(AML)은 예후가 좋지 않은 다양한 병인의 혈액학적 악성 종양의 일종이다1. AML 동물 모델의 생성은 새로운 치료법을 발견하기 위한 노력의 일환으로 복잡한 변이와 병리학을 이해하기 위한 토대를 마련한다2. 생쥐의 백혈구 생성은 혼합 혈통 백혈병(mixed lineage leukemia, MLL) 유전자를 포함하는 융합체를 포함하여 융합 종양 단백질을 발현하는 공여 세포를 이식하여 AML을 강력하게 유도하여 인간의 질병을 모방하는 것을 포함한다3. MLL 유전자 관련 AML4의 이식에서 공여자 세포의 다양한 세포 기원이 보고되었으며, 질병 기원을 담당하는 세포에 대해서는 알려진 바가 거의 없습니다.
생쥐 이식을 위해 여러 경로가 개발되었습니다. 돌연변이 공여 세포를골수 내로 직접 도입하는 대퇴골 내 주사 5 대신, 정맥동 신경총, 꼬리 정맥 및 경정맥을 이용하는 정맥 주사가 쥐 AML 모델 6,7,8,9을 생성하는데 널리 사용되어 왔다. retro-orbital (r.o.) 주사의 경우, 부피 제한, 높은 기술적 요구, 반복적인 시도 또는 오류의 가능성 적음, 잠재적인 안구 손상과 같은 다양한 내재적 단점이 제한적이거나 실행 가능한 대안이 없는 주요 걸림돌이 되었습니다7. 꼬리 정맥 주사는 국소 부상 외에도 유사한 문제를 가질 수 있습니다. 시술을 용이하게 하기 위해, 생쥐는 종종 꼬리 정맥을 확장시키기 위해 워밍업을 할 필요가 있다10. 또한 추가 광원 없이는 꼬리 정맥을 찾기가 어려우며, 특히 C57BL/6 균주의 마우스에서 그렇습니다. 경정맥 주사의 경우 연구 인력은 정맥을 찾고 가능한 합병증을 제한하기 위해 충분한 교육이 필요합니다. 또한 정맥동과 경정맥 주사는 모두 마취하에 수행해야하므로 또 다른 수준의 복잡성이 추가됩니다. 따라서 AML 쥐 모델의 확립을 촉진하기 위해 이식을 위한 새로운 경로를 탐색하고 싶은 유혹이 있습니다.
