אפיתל פיגמנט ברשתית או RPE הוא קריטי לבריאות הרשתית. וחקר RPE המבודד הוא חיוני למחלת הרשתית. שיטה זו של בידוד RPE מהשרקן, מודל קוצר ראייה פופולרי, מסייעת בפתרון הבעיה המאתגרת.היתרון העיקרי של טכניקה זו הוא שהיא פשוטה יחסית ומניבה דגימת RPE באיכות גבוהה המתאימה למחקרים המולקולריים הביולוגיים כולל ניתוח RPE. על ידי ביצוע מושלם של הטכניקה שהודגמה והתבוננות כיצד לתפעל את כלי הניתוח, ניתן לשלוט בהליך זה במהירות עם קצת תרגול. לאחר המתת חסד אנושית של השרקן וחניכת עיני החיה, שטפו מיד את העיניים על ידי העברתן לצלחת פטרי בגודל 10 ס"מ המכילה מלח חוצץ פוספט סטרילי או PBS.לאחר שטיפה, להעביר את העיניים לתמיסת PBS טרייה. כעת עובדים תחת מיקרוסקופ מנתח, השתמשו במחט של 18 מד כדי ליצור פתח קטן ראשוני בלובן העין של עין אחת, כמילימטר אחד מאחורי הגבול הגפיים בין הקרנית לסקלרה. בעזרת מספריים, הסר את החלק הקדמי, כולל הקרנית, הקשתית, הגוף הריסי והעדשה הגבישית.לאחר מכן עבודה עם מקטע עיני אחורי שנותר, השתמש במלקחיים כדי לתפוס ולמשוך בעדינות על הזונולה של, ולאחר מכן לנתק את הרשתית מקומפלקס RPE/choroid/sclera, ולקלף את הרשתית ללא פיצול. לאחר הסרת הרשתית לחלוטין, טבלו את העין האחורית הנותרת, הכוללת את RPE, כורויד וסקלרה באחת הבארות של צלחת בת 12 בארות המכילה שני מיליליטר של מגיב אחסון רקמות והשאירו אותה שקועה במשך חמש דקות. כדי לשטוף את מגיב האחסון, העבירו את העין לבאר אחרת מלאה בארבעה מיליליטר PBS למשך 10 שניות לפני העברתה לבאר שלישית מלאה בשני מיליליטר PBS.לפני שתמשיך לשלב הבידוד הסופי של RPE, הכינו מזרק של מיליליטר מלא PBS וחברו מחט של 30 מד. לחץ בעדינות על בוכנה מזרק כדי ליצור זרם סילון של PBS. בעבודה תחת מיקרוסקופ, ראשית, לכוון את הזרם הזה של PBS על RPE כדי ליצור קרע קטן או חור בו.לאחר מכן כוון את זרם PBS לתוך הפתח שנוצר כדי לנתק את RPE כמו יריעה מן choroid. לאחר ניתוק ה-RPE מהכורואיד, אספו את תאי ה-RPE במזרק של מיליליטר אחד ללא מחט והעבירו את הדגימה שנאספה לצינור של 1.5 מיליליטר. צנטריפוגה את הצינור עם RPE שנאסף ב 8, 000 x גרם במשך דקה אחת כדי לקבל גלולת RPE.כדי שדגימות ישמשו בניתוחי RNA, השליכו את הסופרנטנט, כלומר תמיסת PBS, והחליפו אותו ב-350 מיליליטר של חיץ ליזיס כפי שנכלל בערכות בידוד RNA. בצע פיטום של התוכן למעלה ולמטה 20 פעמים כדי לערבב היטב ולשמור על איכות הדגימה. לאחסון ושימור לטווח ארוך, מעבירים את הדגימות למקפיא מינוס 80 מעלות צלזיוס.השתמש בערכת בידוד RNA ופעל לפי הוראות היצרן כדי לבודד ולאסוף את ה- RNA מדגימות RPE לפני הערכת איכות הדגימה באמצעות אלקטרופורזה. במחקר שלנו, דגימות RPE שנאספו הראו RNA שמור היטב בעל מספר שלמות RNA או RIN גדול משמונה. ספירת RNA כוללת של כ-240 ננוגרם לעין.רמת הביטוי של הגן הספציפי RPE, Rpe65, נמצאה גבוהה משמעותית בדגימות RPE מאשר בכורואיד וסקלרה. לעומת זאת, דגימות RPE הראו ביטוי מינימלי של הגן הספציפי לובן העין הכורואידי שנבחר, קולגן מסוג אלפא-אחד, מה שמצביע על היעדר מזהמים כורואידיים וסקלרליים בדגימות RPE המבודדות. השלב החשוב ביותר של ההליך הוא הסרה חלקה של הרשתית מבלי להשאיר מאחור כל קטע רשתית.הליך בידוד RPE זה מצדיק הסבר מחקרי תרבית תאים חוץ גופית עם כמה הבדלים חשובים, כולל בחירת המדיה לטבילה של תאי RPE לאחר הבידוד.
