치주염(PD)은 전 세계적으로 6번째로 널리 퍼진 공중 보건 질환으로, 전체 인구의 약 11%에 영향을 미치며, 진행성, 비가역적, 파괴적 형태의 치주 질환이다 1,2. PD는 치은 및 치주 조직에 영향을 미치는 염증 과정으로, 치은 후퇴, 포켓 발달을 동반한 접합 상피의 정점 이동, 치조골 손실을 초래합니다3. 또한, 파킨슨병은 심혈관 질환, 비만, 당뇨병 및 류마티스 관절염을 포함한 여러 전신 질환과 관련이 있으며, 환경 및 숙주 특이적 요인이 중요한 역할을 한다 4,5.
따라서, PD는 주로 미생물 군집의 dysbiosis로 인한 미생물 플라크의 축적과 치주 병원체에 대한 과장된 숙주 면역 반응에 의해 시작되어 치주 조직의 파괴를 초래하는 다인성 질환입니다 4,6. 여러 치주 박테리아 중에서 그람 음성 혐기성 박테리아인 Porphyromonas gingivalis는 PD4의 주요 병원체 중 하나입니다. P. gingivalis는 염증이 있는 치주 조직에서 다형핵 백혈구 침윤 및 혈관 확장을 유도하는 것으로 알려진 분자인 복합 지질다당류(LPS)를 벽에 함유하고 있다7. 그 결과 인터루킨 1(IL-1), IL-6, IL-8과 같은 염증 매개체, 종양 괴사 인자(TNF) 또는 프로스타글란딘이 생성되어 파골세포가 활성화되고 뼈 흡수가 일어나 조직 파괴와 궁극적인 치아 손실이 발생합니다3.
동물 모델의 다양한 장점 중에는 인간에서와 같이 세포 복잡성을 모방할 수 있는 능력, 또는 제한된 세포 유형의 플라스틱 표면에서 수행되는 시험관 내 연구보다 더 정확하다는 점이 있다8. 생체 내에서 PD를 실험적으로 모델링하기 위해 인간이 아닌 영장류, 개, 돼지, 흰 족제비, 토끼, 생쥐 및 쥐와 같은 다양한 동물 종이 사용되었습니다9. 그러나, 래트는 파킨슨병의 발병기전에 대해 가장 광범위하게 연구된 동물 모델인데, 그 이유는 이들이 저렴하고 다루기 쉽기 때문이다10. 그들의 치은 조직은 인간의 치은 조직과 유사한 구조적 특징을 가지고 있으며, 얕은 치은 고랑과 접합 상피가 치아 표면에 부착되어 있습니다. 또한, 인간과 마찬가지로 접합 상피는 염증 세포에서 박테리아, 이물질 및 삼출물의 통과를 촉진한다 9.
쥐의 PD 유도에 대해 가장 많이 보고된 실험 모델 중 하나는 치아 주위에 합자를 배치하는 것인데, 이는 기술적으로 어렵지만 신뢰할 수 있습니다10. 합자 배치는 치태와 박테리아 축적을 촉진하여 치주 조직 염증 및 파괴를 유발하는 치은 고랑에 dysbiosis를 생성합니다11. 치주 부착의 상실 및 치조골의 재흡수는 이 쥐 모델8에서 7일 이내에 발생할 수 있다.
PD에 대한 또 다른 동물 모델은 치은 조직에 LPS를 주입하는 것으로 구성됩니다. 결과적으로 파골 세포 형성과 뼈 손실이 자극됩니다. 이 모델의 조직병리학적 특징은 더 높은 수준의 전염증성 사이토카인, 콜라겐 분해 및 치조골 흡수를 특징으로 하는 인간이 확립한 PD와 유사합니다 6,8.
따라서 본 연구의 목적은 제1 상악 어금니(M1) 주위의 합자 배치와 결합된 P. gingivalis-LPS(Pg-LPS) 주사 기술을 기반으로 한 실험 PD의 간단한 쥐 모델을 설명하는 것이었습니다. 이것은 인간 PD 질환에서 관찰되는 것과 유사한 특성을 가진 모델로, 질병 진행 메커니즘 및 향후 가능한 치료법 연구에 사용될 수 있습니다.
