1. 板材涂层

神经元可以在其他培养容器中培养：24孔板，8孔室，60 mm培养皿或MED探针。然而，在这些情况下，需要调整最终浓度和添加Ara-C的时间。此外，神经元的密度需要在不同类型的实验中进行优化。例如，电生理实验需要高密度培养，在这种情况下，每周需要两次培养基更换（图6）。因此，低密度培养比高密度培养需要的步骤更少。
低密度神经元培养通常需要先进的技术;但是，使用即用型冷冻库存可以解决此问题。所描述的方法不依赖于实验者的技能。冷冻浆料质量稳定，只要储存在液氮中，避免温度变化长达4年，就可以稳定养殖。
在没有培养基交换的情况下培养细胞3周提出了是否存在显着渗透压变化或培养基蒸发的问题。但是，我们已经确认培养基蒸发量很小（还原率为3.6%）。突触蛋白的定位和神经元的形态在3周后看起来正常。因此，没有培养基交换的3周培养不会引起影响培养神经元条件的较大渗透压变化。在Ara-C处理后将板保持在培养箱中也是最大限度地减少蒸发的重要一点。
冷冻存量神经元的使用没有限制。然而，低密度培养方法存在一些局限性。我们确认低密度培养物可用于神经元的形态学观察、突触功能的评估和GFP转染。然而，我们还没有检查活细胞成像。此外，如上所述，需要进行电生理学的高密度培养。
突触成熟通常需要3周7，我们无法确认培养的神经元是否具有适当的突触，直到最后。如果突触在3周后发育不良，我们将不得不再次培养。通过在开始实验之前了解神经元的质量，我们可以节省这 3 周的时间。因此，为了有效地进行实验，最好提前检查神经元的质量。冷冻库存可以事先检查神经元的质量。每批冷冻库存由一窝老鼠产生，我们可以使用每批中的一种库存进行质量检查。Drebrin是神经元质量检查的良好标志物。如上所述，drebrin积聚在成熟神经元的脊柱头中，并对突触刺激做出反应。因此，我们可以通过使用drebrin作为标记来检查冷冻库存中神经元的质量。
该方法可用于评估药物对突触状态的影响。树突棘的drebrin外流发生在突触可塑性的初始阶段22。因此，通过药物治疗引起的drebrin簇减少的检测表明，该药物刺激突触并引起突触可塑性。此外，为了确定还原是否依赖于NMDAR，使用2-氨基-5-磷酸新钙酸（APV，一种NMDAR拮抗剂）的实验是有用的。使用drebrin作为标记，即使是NMDAR依赖性也可以明确确定10，15。所描述的方法可用于药物筛选、安全药理学研究和突触功能评估。

开发一种体外实验系统，可以评估参与学习和记忆的突触功能是很重要的。神经元培养是体外评估突触功能的宝贵系统。神经元培养技术于 1980 年代首次使用，在 1990 年代，开发了原代海马神经元的低密度培养1，2，3 用于研究单个神经元的蛋白质成分亚细胞定位、蛋白质运输、神经元极性、脊柱形态、突触发育和可塑性4，5，6，7，8.然而，这种技术涉及许多步骤：交配动物、解剖胚胎、制备培养容器以及每周更换一次培养基培养细胞 3 周。此外，它需要先进的技术3.
我们已经从大鼠胚胎9，10中开发了分离的海马神经元的冷冻库存。冷冻的神经元储备是即用型的，培养细胞不需要先进的技术11，12。换句话说，从冷冻的原种培养神经元并不取决于实验者的技术。它消除了对动物实验的需要（例如，允许动物实验，安排定时怀孕的动物和解剖大鼠胚胎），从而减少了使用的动物数量。最近，高质量、即用型冷冻神经元库存已经商业化。在这里，我们使用了胚胎日（E）18大鼠海马体13，14，15的市售冷冻储备。从冷冻原液中培养神经元不需要胶质条件培养基或与神经胶质细胞共培养。无需额外血清的普通原代培养基可用于培养细胞;因此，我们可以获取可重复的数据。此外，细胞接种后3周内不需要更换培养基，因为神经胶质细胞的生长减少（图1）。
