동결된 스톡 뉴런의 사용에 관한 제한은 없다. 그러나 저밀도 배양 방법에는 몇 가지 한계가 있습니다. 저밀도 배양이 뉴런의 형태학적 관찰, 시냅스 기능 평가 및 GFP 형질감염에 적용될 수 있음을 확인했습니다. 그러나 우리는 살아있는 세포 이미징을 조사하지 않았습니다. 또한, 위에서 언급했듯이, 전기 생리학을 수행하기 위해서는 고밀도 배양이 필요합니다.
시냅스 성숙은 일반적으로3주 7주가 소요되며, 배양된 뉴런이 끝까지 적절한 시냅스를 가지고 있는지 확인할 수 없습니다. 3 주 후에 시냅스 성숙이 좋지 않으면 다시 배양해야합니다. 실험을 시작하기 전에 뉴런의 품질을 알면이 3 주를 절약 할 수 있습니다. 따라서 실험을 효율적으로 수행하기 위해서는 사전에 뉴런의 품질을 확인하는 것이 가장 좋다. 냉동 된 주식은 사전에 뉴런의 품질을 확인하는 것을 가능하게합니다. 냉동 재고의 각 배치는 한 깔짚의 쥐에서 생성되며 품질 검사를 위해 각 배치의 재고 중 하나를 사용할 수 있습니다. easy and reproducible low-density primary culture using frozen stock of embryonic hippocampal neurons
