NIR-I fluorescerende beeldvorming kan tot op zekere hoogte worden gebruikt voor tumor- en vasculaire beeldvorming, maar vanwege de beperkte maximale emissiegolflengte van NIR-I-fluoroforen (<900 nm), resulteert dit in slechte weefselpenetratie en tumorsignaalachtergrondverhouding33,34. Fluorescentiebeeldvorming is het veelgebruikte moleculaire beeldvormingsinstrument in fundamenteel onderzoek en wordt ook vaak gebruikt om chirurgische tumorresectie in kliniekente begeleiden 1. Het essentiële principe van fluorescentiebeeldvorming is om een camera te gebruiken om fluorescentie te ontvangen die door een laser wordt uitgezonden na de bestraling van monsters (weefsels, organen, enz.) 2. Het proces is binnen enkele milliseconden voltooid3. De fluorescentiebeeldgolflengten kunnen worden onderverdeeld in ultraviolet (200-400 nm), zichtbaar gebied (400-700 nm), nabij-infrarood I (NIR-I, 700-900 nm) en nabij-infrarood II (NIR-II, 1000-1700 nm)4,5,6. Omdat de endogene moleculen zoals hemoglobine, melanine, deoxyhemoglobine en bilirubine in biologische weefsels een sterke absorptie en een verstrooiend effect op het licht in zichtbare gebieden hebben, worden de penetratie en gevoeligheid van licht sterk verminderd en wordt de fluorescentiebeeldvorming in zichtbare lichtgolflengten nadelig beïnvloed 7,8,9.
NIR-II fluorescentiebeeldvorming heeft een lage fotonenabsorptie en -verstrooiing, een hoge beeldsnelheid en een hoog beeldcontrast (of gevoeligheid)10,11. Naarmate de fluorescentiegolflengte toeneemt, neemt de absorptie en verstrooiing van fluorescentie in biologische weefsels geleidelijk af en is de autofluorescentie in het NIR-II-gebied extreem laag12. Zo verhoogt het NIR-II-venster de penetratiediepte van weefsels aanzienlijk en verkrijgt het een hogere resolutie en signaal-ruisverhouding13,14,15. Het NIR-II venster kan verder worden onderverdeeld in de NIR-IIa (1300-1400 nm) en NIR-lIb (1500-1700 nm) windows16. Tot op heden zijn verschillende mijlpaal NIR-II-materialen gemeld, waaronder anorganisch materiaal enkelwandige koolstofnanobuizen, zeldzame aardnanodeeltjes, quantum dots en organische materiaal halfgeleiderpolymeer nanodeeltjes, kleinmolecuulkleurstoffen, aggregatie-geïnduceerde luminescerende materialen, enz. 1,17,18,19,20,21,22. Anorganische nanomaterialen worden gemakkelijk opgehoopt in de lever, milt, enz., en hebben potentiële biotoxiciteit op lange termijn23. Organische fluorofoor met kleine moleculen heeft de voordelen van een snel metabolisme, lage toxiciteit, eenvoudige modificatie en een duidelijke structuur, wat de meest veelbelovende sonde is voor klinisch gebruik24.
Het NIR-II optische beeldvormingssysteem is ook een cruciaal onderdeel van fluorescentiebioimaging omdat het effectief NIR-II-fluorescentiesignalen van de NIR-II-sonde kan verzamelen, waardoor nauwkeurige functionele, anatomische en moleculaire beelden25,26 worden weergegeven. Het NIR-II beeldvormingssysteem bestaat voornamelijk uit kortegolf infraroodcamera's, long-pass (LP) filters, lasers en computerprocessors. In vivo NIR-II fluorescerende beeldvorming wordt beschouwd als een van de meest haalbare beeldvormingsbenaderingen voor het ophelderen van de mechanismen van ziekten en de aard van het leven27,28,29. NIR-II beeldvormingstechnologie is op grote schaal gebruikt in biomedische gebieden zoals kankerceldetectie, dynamische beeldvorming, in vivo gerichte tracering en gerichte therapie, vooral in oncologisch onderzoek30,31. Gezien de hoge technische vereisten van NIR-II-beeldvormingstechnologie op beeldvormingsondes en -instrumenten, puzzelt en beperkt het echter ook het praktische gebruik van onderzoekers op verschillende gebieden. Daarom worden de voorbereiding van NIR-II imaging probes en de toepassingen van NIR-II imaging in detail geïntroduceerd in dit artikel.

a bright nir-ii fluorescence probe for vascular and tumor imaging

Als een opkomende beeldvormingstechnologie heeft nabij-infrarood II (NIR-II, 1000-1700 nm) fluorescentiebeeldvorming een aanzienlijk potentieel op biomedisch gebied, vanwege de hoge gevoeligheid, diepe weefselpenetratie en superieure beeldvorming met ruimtelijke en temporele resolutie. De methode om de implementatie van NIR-II fluorescentiebeeldvorming voor sommige dringend noodzakelijke gebieden, zoals medische wetenschap en farmacie, te vergemakkelijken, heeft relevante onderzoekers echter verbaasd. Dit protocol beschrijft in detail de constructie en bioimaging toepassingen van een NIR-II fluorescentie moleculaire sonde, HLY1, met een D-A-D (donor-acceptor-donor) skelet. HLY1 vertoonde goede optische eigenschappen en biocompatibiliteit. Bovendien werd NIR-II vasculaire en tumorbeeldvorming bij muizen uitgevoerd met behulp van een NIR-II optisch beeldvormingsapparaat. Real-time hoge resolutie NIR-II fluorescentiebeelden werden verkregen om de detectie van tumoren en vaatziekten te begeleiden. Van sondevoorbereiding tot data-acquisitie, de beeldkwaliteit is sterk verbeterd en de authenticiteit van de NIR-II moleculaire sondes voor gegevensregistratie in intravitale beeldvorming is gewaarborgd.

De fluorescerende intensiteit en helderheid van wateropschortende HLY1-stippen werden bepaald door een NIR-II-beeldvormingsinstrument. De fluorescerende intensiteit van HLY1 in het 90% fwTHF/H2O-mengsel was vijf keer zo hoog als in de THF-oplossing, wat wees op een prominent AIE-kenmerk van HLY1 (figuur 1B). Hoge resolutie vasculaire en tumor beeldvorming werd gerealiseerd door de nabij-infrarode tot fluorescerende nanoprob, die een nauwkeurige en effectieve methode biedt voor het richten van vaatziekten en kanker. In vivo beeldvorming in het NIR-II-venster, dat ons in staat stelt om diep in levende onderwerpen te kijken, biedt kansen voor biomedisch onderzoek en onze klinische toepassingen. Bereid u voor op de nabij-infrarood II- of NIR-II-beeldvorming door in de handel verkrijgbaar zwart karton in het midden van de drager van het beeldvormingsapparaat te plaatsen.Plaats het monster vervolgens bovenop het zwarte karton, zodat het zich in het midden van de drager bevindt. Nadat u het juiste filter hebt geselecteerd, voert u de platformhoogteaanpassing uit. Druk lang op de optie van het platform omhoog op de touchscreen-interface van het bedieningsgebied van de dragerconsole om deze naar boven te verplaatsen.En druk lang op het platform om het naar beneden te bewegen. Na het synthetiseren van de NIR-II-kleurstof HLY1, bereidt u de HLY1-stippen voor met behulp van de nanoprecipitatiemethode met behulp van DSPE-PEG-2K als een inkapselingsmatrix. Plaats hiervoor een bekerglas met een oplossing van 10 milligram DSPE-PEG-2K in negen milliliter water voor ultrasoonapparaat bij 25 graden Celsius.Los vervolgens één milligram HLY1 op in één milliliter tetrahydrofuraan of THF. En voeg de oplossing langzaam toe aan de DSPE-PEG-2K waterige oplossing die sonicatie ondergaat. Verwijder vervolgens THF uit het mengsel door dialyse.Nadat u de bovenstaande oplossing centrifugaal hebt geconcentreerd met ultrafiltratie op 7, 100 G gedurende 10 minuten, plaatst u deze in een koelkast van vier graden Celsius voor toekomstig gebruik. Voor in vivo beeldvorming, injecteer de HLY1-stippen intraveneus in de verdoofde muizen. En drie minuten later, ga verder met het uitvoeren van NIR-II fluorescentie beeldvorming van de bloedvaten van het hele lichaam van de muizen met behulp van een optisch NIR-II beeldvormingssysteem.Focus verder op het hoofd van de muis om vasculaire beeldvorming van de hersenen te verzamelen. Stel de instrumentparameters van het optische NIR-II-beeldvormingssysteem in op 90 milliwatt per vierkante centimeter en 808 nanometer laser. Verzamel de beelden vijf minuten na de injectie van HLY1-stippen in muizen en verwerk de gegevens met behulp van ImageJ-software.De fluorescentie-intensiteit van HLY1 in de 90% THF-oplossing met 90% water was vijf keer zo hoog als in de THF-oplossing, wat wijst op een prominente aggregatie-geïnduceerde emissie of AIE-kenmerk van HLY1. De HLY1-stippen zonden sterke fluorescerende signalen uit onder een 1.500 nanometer laagdoorlaat- of LP-filter om de geschiktheid voor NIR-IIb-beeldvorming vast te stellen. De maximale absorptie- en emissiegolflengte van HLY1-stippen was respectievelijk 740 nanometer en 1.040 nanometer.Bovendien werd de hydrodynamische grootte van HLY1-stippen bepaald op 145 nanometer door dynamische lichtverstrooiing. Bij muizen toegediend met HLY1-stippen werden de hersenvaten en de microvaten in de achterpoot duidelijk geïdentificeerd door NIR-II-beeldvorming onder een LP-filter van 1.500 nanometer. Verder was de vier T1-tumor van de tumordragende muizen ook duidelijk zichtbaar door NIR-II-beeldvorming, wat wijst op de verbeterde permeabiliteit en retentie of EPR-effect van HLY1-stippen.Al deze resultaten suggereerden dat HLY1-stippen een heldere NIR-II fluorescentiesonde zijn die van toepassing is op vasculaire en tumorbeeldvorming. Voorbereiding van de waterzwevende nanoprobe onder de in vivo en NIR-II fluorescentiebeeldvorming is het belangrijkste onderdeel van de procedure.
