L’imagerie par fluorescence est l’outil d’imagerie moléculaire couramment utilisé en recherche fondamentale, et est également souvent utilisée pour guider la résection chirurgicale de la tumeur dans les cliniques1. Le principe essentiel de l’imagerie par fluorescence est d’utiliser une caméra pour recevoir la fluorescence émise par un laser après l’irradiation d’échantillons (tissus, organes, etc.) 2. Le processus est terminé en quelques millisecondes3. Les longueurs d’onde d’imagerie de fluorescence peuvent être divisées en ultraviolet (200-400 nm), région visible (400-700 nm), proche infrarouge I (NIR-I, 700-900 nm) et proche infrarouge II (NIR-II, 1000-1700 nm)4,5,6. Parce que les molécules endogènes telles que l’hémoglobine, la mélanine, la désoxyhémoglobine et la bilirubine dans les tissus biologiques ont une forte absorption et un effet de diffusion sur la lumière dans les régions visibles, la pénétration et la sensibilité de la lumière sont considérablement réduites, et l’imagerie de fluorescence dans les longueurs d’onde de la lumière visible est affectéenégativement 7,8,9.
L’imagerie par fluorescence NIR-II présente une faible absorption et diffusion des photons, une vitesse d’imagerie élevée et un contraste (ou une sensibilité) d’image élevé10,11. À mesure que la longueur d’onde de fluorescence augmente, l’absorption et la diffusion de la fluorescence dans les tissus biologiques diminuent progressivement, et l’autofluorescence dans la région NIR-II est extrêmement faible12. Ainsi, la fenêtre NIR-II augmente considérablement la profondeur de pénétration des tissus et obtient une résolution et un rapport signal sur bruit plus élevés13,14,15. La fenêtre NIR-II peut être subdivisée en fenêtres NIR-IIa (1300-1400 nm) et NIR-lIb (1500-1700 nm)16. À ce jour, plusieurs matériaux NIR-II importants ont été signalés, notamment des nanotubes de carbone à paroi simple en matériaux inorganiques, des nanoparticules de terres rares, des points quantiques et des nanoparticules de polymères semi-conducteurs de matériaux organiques, des colorants à petites molécules, des matériaux luminescents induits par agrégation, etc. 1,17,18,19,20,21,22. Les nanomatériaux inorganiques s’accumulent facilement dans le foie, la rate, etc., et peuvent être biotoxiques à long terme23. Le fluorophore organique à petites molécules présente les avantages d’un métabolisme rapide, d’une faible toxicité, d’une modification facile et d’une structure claire, ce qui en fait la sonde la plus prometteuse pour un usage clinique24.
Le système d’imagerie optique NIR-II est également un élément essentiel de la bioimagerie par fluorescence, car il peut collecter efficacement les signaux de fluorescence NIR-II de la sonde NIR-II, produisant ainsi des images fonctionnelles, anatomiques et moléculaires précises25,26. Le système d’imagerie NIR-II comprend principalement des caméras infrarouges à ondes courtes, des filtres passe-long (LP), des lasers et des processeurs informatiques. In vivo L’imagerie fluorescente NIR-II est considérée comme l’une des approches d’imagerie les plus réalisables pour élucider les mécanismes des maladies et la nature de la vie27,28,29. La technologie d’imagerie NIR-II a été largement utilisée dans des domaines biomédicaux tels que la détection des cellules cancéreuses, l’imagerie dynamique, le traçage ciblé in vivo et la thérapie ciblée, en particulier dans la recherche en oncologie30,31. Cependant, compte tenu des exigences techniques élevées de la technologie d’imagerie NIR-II sur les sondes et les instruments d’imagerie, elle laisse également perplexe et restreint l’utilisation pratique des chercheurs dans différents domaines. a bright nir-ii fluorescence probe for vascular and tumor imaging

En tant que technologie d’imagerie émergente, l’imagerie par fluorescence proche infrarouge II (NIR-II, 1000-1700 nm) présente un potentiel important dans le domaine biomédical, en raison de sa sensibilité élevée, de sa pénétration des tissus profonds et de son imagerie supérieure avec une résolution spatiale et temporelle. Cependant, la méthode visant à faciliter la mise en œuvre de l’imagerie par fluorescence NIR-II pour certains domaines urgents, tels que les sciences médicales et la pharmacie, a intrigué les chercheurs concernés. Ce protocole décrit en détail les applications de construction et de bioimagerie d’une sonde moléculaire de fluorescence NIR-II, HLY1, avec un squelette D-A-D (donneur-accepteur-donneur). HLY1 a montré de bonnes propriétés optiques et biocompatibilité. De plus, l’imagerie vasculaire et tumorale NIR-II chez la souris a été réalisée à l’aide d’un dispositif d’imagerie optique NIR-II. Des images de fluorescence NIR-II haute résolution en temps réel ont été acquises pour guider la détection des tumeurs et des maladies vasculaires. De la préparation de la sonde à l’acquisition des données, la qualité de l’imagerie est grandement améliorée et l’authenticité des sondes moléculaires NIR-II pour l’enregistrement des données en imagerie intravitale est assurée.

L’intensité fluorescente et la luminosité des points HLY1 suspensibles dans l’eau ont été déterminées par un instrument d’imagerie NIR-II. L’imagerie vasculaire et tumorale à haute résolution a été réalisée par le nanoprob proche infrarouge à fluorescent, fournissant une méthode précise et efficace pour diriger les maladies vasculaires et le cancer. L’imagerie in vivo dans la fenêtre NIR-II, qui nous permet d’examiner en profondeur le sujet vivant, offre des opportunités pour la recherche biomédicale et nos applications cliniques. Préparez-vous à l’imagerie proche infrarouge II ou NIR-II en plaçant du carton noir disponible dans le commerce au centre du support de l’appareil d’imagerie.Ensuite, placez l’échantillon sur le carton noir, de sorte qu’il soit au centre du support. Après avoir sélectionné le filtre approprié, effectuez le réglage de la hauteur de la plate-forme. Appuyez longuement sur l’option de plate-forme vers le haut sur l’interface de l’écran tactile de la zone de contrôle de la console porteuse pour la déplacer vers le haut.Et appuyez longuement sur la plate-forme pour la déplacer vers le bas. Après avoir synthétisé le colorant NIR-II HLY1, préparer les points HLY1 par la méthode de nanoprécipitation en utilisant DSPE-PEG-2K comme matrice d’encapsulation. Pour ce faire, placez un bécher contenant une solution de 10 milligrammes de DSPE-PEG-2K dans neuf millilitres d’eau pour la sonication à 25 degrés Celsius.Ensuite, dissoudre un milligramme de HLY1 dans un millilitre de tétrahydrofurane ou THF. Et ajoutez lentement la solution dans la solution aqueuse DSPE-PEG-2K en cours de sonication. Par la suite, éliminer le THF du mélange par dialyse.Après avoir concentré la solution ci-dessus par centrifugation avec ultrafiltration à 7, 100 g pendant 10 minutes, placez-la dans un réfrigérateur à quatre degrés Celsius pour une utilisation ultérieure. Pour l’imagerie in vivo, injectez les points HLY1 par voie intraveineuse dans les souris anesthésiées. Et trois minutes plus tard, procédez à l’imagerie par fluorescence NIR-II des vaisseaux sanguins de tout le corps des souris à l’aide d’un système d’imagerie optique NIR-II.Concentrez-vous davantage sur la tête de la souris pour recueillir l’imagerie vasculaire cérébrale. Réglez les paramètres de l’instrument du système d’imagerie optique NIR-II à 90 milliwatts par centimètre carré et laser de 808 nanomètres. Collectez les images cinq minutes après l’injection de points HLY1 chez la souris et traitez les données à l’aide du logiciel ImageJ.L’intensité de fluorescence de HLY1 dans la solution à 90% de THF contenant 90% d’eau était cinq fois supérieure à celle de la solution de THF, indiquant une caractéristique importante d’émission induite par agrégation ou d’AIE de HLY1. Les points HLY1 émettaient de forts signaux fluorescents sous un filtre passe-bas ou LP de 1 500 nanomètres, établissant leur aptitude à l’imagerie NIR-IIb. La longueur d’onde maximale d’absorption et d’émission des points HLY1 était de 740 nanomètres et 1 040 nanomètres, respectivement.De plus, la taille hydrodynamique des points HLY1 a été déterminée à 145 nanomètres par diffusion dynamique de la lumière. Chez les souris administrées avec des points HLY1, les vaisseaux cérébraux et les microvaisseaux du membre postérieur ont été clairement identifiés par imagerie NIR-II sous un filtre LP de 1 500 nanomètres. En outre, les quatre tumeurs T1 des souris porteuses de tumeurs étaient également clairement visibles par imagerie NIR-II, indiquant la perméabilité et la rétention améliorées ou l’effet EPR des points HLY1.Tous ces résultats suggèrent que les points HLY1 sont une sonde de fluorescence NIR-II brillante applicable à l’imagerie vasculaire et tumorale. La préparation de la nanosonde suspension à l’eau sous l’imagerie in vivo et par fluorescence NIR-II est la partie la plus importante de la procédure.
