Die Fluoreszenzbildgebung ist das am häufigsten verwendete molekulare Bildgebungswerkzeug in der Grundlagenforschung und wird auch häufig zur Steuerung der chirurgischen Tumorresektion in Klinikeneingesetzt 1. Das wesentliche Prinzip der Fluoreszenzbildgebung besteht darin, eine Kamera zu verwenden, um die von einem Laser emittierte Fluoreszenz nach der Bestrahlung von Proben (Gewebe, Organe usw.) zu empfangen. 2. Der Vorgang ist innerhalb weniger Millisekunden abgeschlossen3. Die Wellenlängen der Fluoreszenzbildgebung können in ultraviolette (200-400 nm), sichtbare Bereiche (400-700 nm), Nahinfrarot I (NIR-I, 700-900 nm) und Nahinfrarot II (NIR-II, 1000-1700 nm) unterteilt werden4,5,6. Da die endogenen Moleküle wie Hämoglobin, Melanin, Desoxyhämoglobin und Bilirubin in biologischen Geweben eine starke Absorption und einen Streueffekt auf das Licht in sichtbaren Bereichen haben, werden das Eindringen und die Empfindlichkeit des Lichts stark reduziert, und die Fluoreszenzbildgebung in Wellenlängen des sichtbaren Lichts wird nachteilig beeinflusst 7,8,9.
Die NIR-II-Fluoreszenzbildgebung weist eine geringe Photonenabsorption und -streuung, eine hohe Abbildungsgeschwindigkeit und einen hohen Bildkontrast (oder eine hohe Empfindlichkeit) auf10,11. Mit zunehmender Fluoreszenzwellenlänge nimmt die Absorption und Streuung von Fluoreszenz in biologischen Geweben allmählich ab, und die Autofluoreszenz in der NIR-II-Region ist extrem niedrig12. Somit erhöht das NIR-II-Fenster die Eindringtiefe von Geweben signifikant und erhält eine höhere Auflösung und ein höheres Signal-Rausch-Verhältnis13,14,15. Das NIR-II-Fenster kann weiter unterteilt werden in die Fenster NIR-IIa (1300-1400 nm) und NIR-lIb (1500-1700 nm)16. Bisher wurden mehrere Meilensteine von NIR-II-Materialien berichtet, darunter einwandige Kohlenstoffnanoröhren aus anorganischem Material, Seltenerd-Nanopartikel, Quantenpunkte und Halbleiterpolymer-Nanopartikel aus organischem Material, niedermolekulare Farbstoffe, aggregationsinduzierte Lumineszenzmaterialien usw. 1,17,18,19,20,21,22. Anorganische Nanomaterialien reichern sich leicht in Leber, Milz usw. an und haben eine potenzielle langfristige Biotoxizität23. Organisches niedermolekulares Fluorophor hat die Vorteile eines schnellen Stoffwechsels, einer geringen Toxizität, einer einfachen Modifikation und einer klaren Struktur, die die vielversprechendste Sonde für den klinischen Einsatz ist24.
Das optische Bildgebungssystem NIR-II ist auch eine wichtige Komponente der Fluoreszenz-Biobildgebung, da es NIR-II-Fluoreszenzsignale von der NIR-II-Sonde effizient erfassen und so präzise funktionelle, anatomische und molekulare Bilder liefernkann 25,26. Das NIR-II-Bildgebungssystem besteht hauptsächlich aus Kurzwellen-Infrarotkameras, Langpassfiltern, Lasern und Computerprozessoren. Im lebenden Organismus Die NIR-II-Fluoreszenzbildgebung gilt als einer der praktikabelsten bildgebenden Ansätze zur Aufklärung der Mechanismen von Krankheiten und der Natur des Lebens27,28,29. Die NIR-II-Bildgebungstechnologie ist in biomedizinischen Bereichen wie der Erkennung von Krebszellen, der dynamischen Bildgebung, der gezielten In-vivo-Verfolgung und der gezielten Therapie, insbesondere in der onkologischen Forschung, weit verbreitet30,31. Angesichts der hohen technischen Anforderungen der NIR-II-Bildgebungstechnologie an bildgebende Sonden und Instrumente verwirrt und schränkt sie jedoch auch den praktischen Einsatz von Forschern in verschiedenen Bereichen ein. Als aufstrebende Bildgebungstechnologie hat die Nahinfrarot-II-FLUORESZENZBILDGEBUNG (NIR-II, 1000-1700 nm) aufgrund ihrer hohen Empfindlichkeit, ihrer tiefen Gewebedurchdringung und ihrer überlegenen Bildgebung mit räumlicher und zeitlicher Auflösung ein erhebliches Potenzial im biomedizinischen Bereich. Die Methode zur Erleichterung der Implementierung der NIR-II-Fluoreszenzbildgebung für einige dringend benötigte Bereiche wie Medizin und Pharmazie hat jedoch relevante Forscher verwirrt. Dieses Protokoll beschreibt detailliert den Aufbau und die Bioimaging-Anwendungen einer molekularen NIR-II-Fluoreszenzsonde, HLY1, mit einem D-A-D-Skelett (Donor-Akzeptor-Donor). HLY1 zeigte gute optische Eigenschaften und Biokompatibilität. Darüber hinaus wurde die NIR-II-Gefäß- und Tumorbildgebung bei Mäusen mit einem NIR-II-Optik-Bildgebungsgerät durchgeführt. Es wurden hochauflösende NIR-II-Fluoreszenzbilder in Echtzeit aufgenommen, um die Erkennung von Tumoren und Gefäßerkrankungen zu steuern. Von der Sondenvorbereitung bis zur Datenerfassung wird die Bildqualität erheblich verbessert und die Authentizität der molekularen NIR-II-Sonden für die Datenaufzeichnung in der intravitalen Bildgebung sichergestellt.

Die Fluoreszenzintensität und Helligkeit von wasserabhängigen HLY1-Punkten wurde mit einem NIR-II-Bildgebungsgerät bestimmt. Die hochauflösende Gefäß- und Tumorbildgebung wurde mit dem Nahinfrarot- bis Fluoreszenz-Nanoprob realisiert und bietet eine genaue und effektive Methode zur Steuerung von Gefäßerkrankungen und Krebs. Die In-vivo-Bildgebung im NIR-II-Fenster, die es uns ermöglicht, tief in lebende Probanden zu blicken, bietet Möglichkeiten für die biomedizinische Forschung und unsere klinischen Anwendungen. Bereiten Sie sich auf die Nahinfrarot-II- oder NIR-II-Bildgebung vor, indem Sie handelsüblichen schwarzen Karton in die Mitte des Trägers des Bildgebungsgeräts legen.Legen Sie dann die Probe auf den schwarzen Karton, so dass sie sich in der Mitte des Trägers befindet. Stellen Sie die Geräteparameter des optischen NIR-II-Bildgebungssystems auf 90 Milliwatt pro Quadratzentimeter und 808 Nanometer Laser ein. Sammeln Sie die Bilder fünf Minuten nach der Injektion von HLY1-Punkten in Mäuse und verarbeiten Sie die Daten mit der ImageJ-Software.Die Fluoreszenzintensität von HLY1 in der 90%igen THF-Lösung, die 90% Wasser enthielt, war fünfmal so hoch wie in der THF-Lösung, was auf eine prominente aggregationsinduzierte Emission oder AIE-Eigenschaft von HLY1 hinweist. Die HLY1-Punkte emittierten starke Fluoreszenzsignale unter einem 1.500-Nanometer-Tiefpass- oder LP-Filter, was ihre Eignung für die NIR-IIb-Bildgebung bestätigte. Die maximale Absorptions- und Emissionswellenlänge von HLY1-Punkten betrug 740 Nanometer bzw. 1.040 Nanometer.Zusätzlich wurde die hydrodynamische Größe von HLY1-Punkten durch dynamische Lichtstreuung auf 145 Nanometer bestimmt. Bei Mäusen, denen HLY1-Punkte verabreicht wurden, wurden die Hirngefäße und die Mikrogefäße in den Hinterbeinen durch NIR-II-Bildgebung unter einem 1.500-Nanometer-LP-Filter eindeutig identifiziert. Darüber hinaus waren die vier T1-Tumoren der tumortragenden Mäuse auch durch NIR-II-Bildgebung deutlich sichtbar, was auf die verbesserte Permeabilität und Retention oder den EPR-Effekt von HLY1-Punkten hinweist.Alle diese Ergebnisse deuten darauf hin, dass HLY1-Punkte eine helle NIR-II-Fluoreszenzsonde sind, die für die Gefäß- und Tumorbildgebung geeignet ist. Die Herstellung der wassersuspendierbaren Nanosonde unter der In-vivo- und NIR-II-Fluoreszenzbildgebung ist der wichtigste Teil des Verfahrens.
