蛍光イメージングは、基礎研究で一般的に使用される分子イメージングツールであり、クリニックでの外科的腫瘍切除のガイドにもよく使用されます1。蛍光イメージングの本質的な原理は、サンプル(組織、臓器など)の照射後にレーザーによって放出された蛍光を受信するためにカメラを使用することです。2.プロセスは数ミリ秒以内に完了します 3.蛍光イメージング波長は、紫外(200-400 nm)、可視領域(400-700 nm)、近赤外I(NIR-I、700-900 nm)、近赤外II(NIR-II、1000-1700 nm)4,5,6に分けることができます。生体組織内のヘモグロビン、メラニン、デオキシヘモグロビン、ビリルビンなどの内因性分子は、可視領域の光に強い吸収と散乱効果があるため、光の透過と感度が大幅に低下し、可視光波長での蛍光イメージングに悪影響を及ぼします7,8,9。
NIR-II蛍光イメージングは、光子の吸収と散乱が低く、イメージング速度が速く、画像のコントラスト(または感度)が高い10,11。蛍光波長が増加するにつれて、生体組織における蛍光の吸収および散乱は徐々に減少し、NIR-II領域の自己蛍光は極めて低い12。したがって、NIR-IIウィンドウは、組織の浸透深さを大幅に増加させ、より高い解像度と信号対雑音比13,14,15を取得します。NIR-IIウィンドウは、NIR-IIa(1300-1400nm)およびNIR-lIb(1500-1700nm)ウィンドウ16にさらに細分することができる。これまでに、無機材料単層カーボンナノチューブ、希土類ナノ粒子、量子ドット、有機材料半導体ポリマーナノ粒子、低分子色素、凝集誘起発光材料など、いくつかの画期的なNIR-II材料が報告されています。 1,17,18,19,20,21,22。無機ナノ材料は肝臓や脾臓などに蓄積しやすく、長期的な生体毒性の可能性があります23。有機低分子蛍光色素は、代謝が速く、毒性が低く、修飾が容易で、構造が明確であるという利点があり、臨床使用のための最も有望なプローブです24。
NIR-II光学イメージングシステムは、NIR-IIプローブからNIR-II蛍光シグナルを効果的に収集し、正確な機能的、解剖学的、および分子画像をレンダリングできるため、蛍光バイオイメージングの重要なコンポーネントでもあります25,26。NIR-IIイメージングシステムは、主に短波赤外線カメラ、ロングパス(LP)フィルター、レーザー、およびコンピュータープロセッサで構成されています。 生体内NIR-II蛍光イメージングは、疾患のメカニズムと生命の性質を解明するための最も実行可能なイメージングアプローチの1つと考えられています27,28,29。NIR-IIイメージング技術は、癌細胞検出、ダイナミックイメージング、in vivoターゲットトレーシング、標的治療などの生物医学分野で、特に腫瘍学研究で広く使用されています30,31。しかし、イメージングプローブや機器におけるNIR-IIイメージング技術の高い技術的要件を考慮すると、さまざまな分野の研究者の実用化を困惑させ、制限することもできません。そこで、NIR-IIイメージングプローブの作製とNIR-IIイメージングの応用について、本稿で詳しく紹介します。

新しいイメージング技術として、近赤外II(NIR-II、1000-1700 nm)蛍光イメージングは、その高感度、深部組織の浸透、および空間的および時間的解像度による優れたイメージングにより、生物医学分野で大きな可能性を秘めています。しかし、医学や薬学など、緊急に必要な分野でNIR-II蛍光イメージングの実装を容易にする方法は、関連する研究者を困惑させています。このプロトコルでは、D-A-D(ドナー-アクセプター-ドナー)骨格を持つNIR-II蛍光分子プローブHLY1の構築とバイオイメージングアプリケーションについて詳しく説明します。HLY1は良好な光学特性と生体適合性を示した。さらに、マウスにおけるNIR-II血管および腫瘍のイメージングは、NIR-II光学イメージングデバイスを用いて行った。リアルタイムの高解像度NIR-II蛍光画像を取得して、腫瘍や血管疾患の検出をガイドしました。プローブの準備からデータ取得まで、イメージング品質が大幅に向上し、生体内イメージングにおけるデータ記録用のNIR-II分子プローブの信頼性が保証されます。

近赤外から蛍光ナノプローブによって高解像度の血管および腫瘍イメージングが実現され、血管疾患および癌を指示するための正確で効果的な方法を提供しました。NIR-IIウィンドウでの生体内イメージングは、生体を深く見ることを可能にし、生物医学研究と臨床応用の機会を生み出します。撮像装置のキャリアの中央に市販の黒い段ボールを置き、近赤外IIまたはNIR-IIの撮像の準備をする。次に、サンプルを黒い段ボールの上に置き、キャリアの中央にくるようにします。適切なフィルターを選択したら、プラットフォームの高さ調整を実行します。キャリアコンソールコントロールエリアのタッチスクリーンインターフェイスでプラットフォームのオプションを長押しして、上に移動します。そして、プラットフォームを長押しして下に移動します。NIR-II色素HLY1を合成した後、DSPE-PEG-2Kをカプセル化マトリックスとして用いたナノ沈降法によりHLY1ドットを作製した。これを行うには、摂氏25度で超音波処理するために、DSPE-PEG-2Kの10ミリグラムの溶液を含むビーカーを9ミリリットルの水に入れます。次に、1ミリグラムのHLY1を1ミリリットルのテトラヒドロフランまたはTHFに溶解します。そして、超音波処理を受けているDSPE-PEG-2K水溶液に溶液をゆっくりと加えます。続いて、透析によって混合物からTHFを除去する。上記の溶液を7, 100 Gで10分間限外ろ過しながら遠心的に濃縮した後、将来の使用のために摂氏4度の冷蔵庫に入れます。in vivoイメージングのために、HLY1ドットを麻酔したマウスに静脈内注射する。そして3分後、光学NIR-IIイメージングシステムを用いてマウスの全身の血管のNIR-II蛍光イメージングを行う。マウスの頭にさらに焦点を合わせて、脳血管イメージングを収集します。光学NIR-IIイメージングシステムの機器パラメータを90ミリワット/平方センチメートルと808ナノメートルレーザーに設定します。マウスにHLY1ドットを注入してから5分後に画像を収集し、ImageJソフトウェアを使用してデータを処理します。90%水を含む90%THF溶液中のHLY1の蛍光強度は、THF溶液中の5倍であり、HLY1の顕著な凝集誘発発光またはAIEの特徴を示した。HLY1ドットは、1, 500ナノメートルのローパスまたはLPフィルターの下で強い蛍光シグナルを発し、NIR-IIbイメージングへの適合性を確立しました。HLY1ドットの最大吸収波長と発光波長はそれぞれ740ナノメートルと1,040ナノメートルでした。さらに、HLY1ドットの流体力学的サイズは、動的光散乱によって145ナノメートルであると決定された。HLY1ドットを投与したマウスでは、後肢の大脳血管および微小血管を1,500ナノメートルLPフィルター下でのNIR-IIイメージングによって明確に同定した。また、担癌マウスの4つのT1腫瘍は、NIR-IIイメージングによっても明瞭に視認され、HLY1ドットの透過性および保持またはEPR効果の増強を示した。以上の結果から、HLY1ドットは血管や腫瘍のイメージングに適用可能な明るいNIR-II蛍光プローブであることが示唆されました。in vivoおよびNIR-II蛍光イメージング下での水懸濁可能なナノプローブの調製は、手順の最も重要な部分です。
