Fluorescensavbildning er det mest brukte molekylære bildebehandlingsverktøyet i grunnforskning, og brukes også ofte til å veilede kirurgisk tumorreseksjon i klinikker1. Det essensielle prinsippet om fluorescensavbildning er å bruke et kamera for å motta fluorescens som sendes ut av en laser etter bestråling av prøver (vev, organer, etc.) 2. Prosessen er fullført i løpet av få millisekunder3. Fluorescensavbildningsbølgelengdene kan deles inn i ultrafiolett (200-400 nm), synlig region (400-700 nm), nær-infrarød I (NIR-I, 700-900 nm) og nær-infrarød II (NIR-II, 1000-1700 nm) 4,5,6. Fordi de endogene molekylene som hemoglobin, melanin, deoksyhemoglobin og bilirubin i biologisk vev har sterk absorpsjon og en spredningseffekt på lyset i synlige områder, blir lysets penetrasjon og følsomhet sterkt redusert, og fluorescensavbildningen i synlige lysbølgelengder påvirkes negativt 7,8,9.
NIR-II fluorescensavbildning har lav fotonabsorpsjon og spredning, høy bildehastighet og høy bildekontrast (eller følsomhet)10,11. Etter hvert som fluorescensbølgelengden øker, reduseres absorpsjonen og spredningen av fluorescens i biologisk vev gradvis, og autofluorescensen i NIR-II-regionen er ekstremt lav12. Dermed øker NIR-II-vinduet signifikant penetrasjonsdybden til vev og oppnår en høyere oppløsning og signal-støy-forhold13,14,15. NIR-II-vinduet kan videre deles inn i NIR-IIa (1300-1400 nm) og NIR-lIb (1500-1700 nm) vinduer16. Hittil har flere milepæl NIR-II-materialer blitt rapportert, inkludert uorganisk materiale enkeltveggede karbonnanorør, sjeldne jordartnanopartikler, kvanteprikker og halvlederpolymernanopartikler av organisk materiale, småmolekylære fargestoffer, aggregeringsinduserte selvlysende materialer, etc. 1,17,18,19,20,21,22. Uorganiske nanomaterialer akkumuleres lett i leveren, milten osv., og har potensiell langvarig biotoksisitet23. Organisk småmolekylfluorofor har fordelene med rask metabolisme, lav toksisitet, enkel modifikasjon og en klar struktur, som er den mest lovende sonden for klinisk bruk24.
NIR-II optikkavbildningssystem er også en kritisk komponent i fluorescens bioimaging fordi det effektivt kan samle NIR-II fluorescenssignaler fra NIR-II-sonden, og dermed gjengi presise funksjonelle, anatomiske og molekylære bilder25,26. NIR-II bildebehandlingssystemet består hovedsakelig av kortbølge infrarøde kameraer, langpassfiltre (LP), lasere og dataprosessorer. In vivo NIR-II fluorescerende avbildning regnes som en av de mest gjennomførbare bildebehandlingsmetodene for å belyse mekanismene for sykdommer og livets natur27,28,29. NIR-II bildebehandlingsteknologi har blitt mye brukt i biomedisinske felt som kreftcelledeteksjon, dynamisk bildebehandling, in vivo målrettet sporing og målrettet terapi, spesielt innen onkologisk forskning30,31. Men med tanke på de høye tekniske kravene til NIR-II bildebehandlingsteknologi på bildeprober og instrumenter, pusler og begrenser den også den praktiske bruken av forskere på ulike felt. a bright nir-ii fluorescence probe for vascular and tumor imaging

Som en fremvoksende bildebehandlingsteknologi har nær-infrarød II (NIR-II, 1000-1700 nm) fluorescensavbildning betydelig potensial i det biomedisinske feltet på grunn av sin høye følsomhet, dype vevspenetrasjon og overlegen avbildning med romlig og tidsmessig oppløsning. Metoden for å lette implementeringen av NIR-II fluorescensavbildning for noen presserende nødvendige felt, som medisinsk vitenskap og farmasi, har imidlertid forvirret relevante forskere. Samle bildene fem minutter etter injeksjon av HLY1-prikker i mus og behandle dataene ved hjelp av ImageJ-programvare.Fluorescensintensiteten til HLY1 i 90 % THF-oppløsningen inneholdende 90 % vann var fem ganger den i THF-oppløsningen, noe som indikerer en fremtredende aggregasjonsindusert utslipp eller AIE-funksjon ved HLY1. HLY1-punktene sendte ut sterke fluorescerende signaler under et 1.500 nanometer lavpass eller LP-filter som fastslo dets egnethet for NIR-IIb-avbildning. Maksimal absorpsjon og utslippsbølgelengde av HLY1-punkter var henholdsvis 740 nanometer og 1.040 nanometer.I tillegg ble den hydrodynamiske størrelsen på HLY1-punkter bestemt til å være 145 nanometer ved dynamisk lysspredning. Hos mus administrert med HLY1-prikker ble hjernekarene og mikrokarene i bakbenet identifisert tydelig ved NIR-II-avbildning under et 1.500 nanometer LP-filter. Videre var de fire T1-svulstene i de tumorbærende musene også tydelig synlige ved NIR-II-avbildning, noe som indikerer den forbedrede permeabiliteten og retensjonen eller EPR-effekten av HLY1-prikker.Alle disse resultatene antydet at HLY1-prikker er en lys NIR-II fluorescenssonde som gjelder for vaskulær og tumoravbildning. Forberedelse av vannsuspenderbar nanoprobe under in vivo og NIR-II fluorescensavbildning er den viktigste delen av prosedyren.
