La imagen de fluorescencia es la herramienta de imagen molecular comúnmente utilizada en la investigación básica, y también se usa a menudo para guiar la resección quirúrgica del tumor en las clínicas1. El principio esencial de las imágenes de fluorescencia es emplear una cámara para recibir la fluorescencia emitida por un láser después de la irradiación de muestras (tejidos, órganos, etc.) 2. El proceso se completa en unos pocos milisegundos3. Las longitudes de onda de las imágenes de fluorescencia se pueden dividir en ultravioleta (200-400 nm), región visible (400-700 nm), infrarrojo cercano I (NIR-I, 700-900 nm) e infrarrojo cercano II (NIR-II, 1000-1700 nm)4,5,6. Debido a que las moléculas endógenas como la hemoglobina, la melanina, la desoxihemoglobina y la bilirrubina en los tejidos biológicos tienen una fuerte absorción y un efecto de dispersión sobre la luz en las regiones visibles, la penetración y la sensibilidad de la luz se reducen considerablemente, y la imagen de fluorescencia en longitudes de onda de luz visible se ve afectada negativamente 7,8,9.
Las imágenes de fluorescencia NIR-II tienen baja absorción y dispersión de fotones, alta velocidad de imagen y alto contraste (o sensibilidad) de imagen10,11. A medida que aumenta la longitud de onda de la fluorescencia, la absorción y dispersión de la fluorescencia en los tejidos biológicos disminuye gradualmente, y la autofluorescencia en la región NIR-II es extremadamente baja12. Así, la ventana NIR-II aumenta significativamente la profundidad de penetración de los tejidos y obtiene una mayor resolución y relación señal-ruido13,14,15. La ventana NIR-II se puede subdividir en las ventanas NIR-IIa (1300-1400 nm) y NIR-lIb (1500-1700 nm)16. Hasta la fecha, se han reportado varios materiales NIR-II importantes, incluidos nanotubos de carbono de pared simple de material inorgánico, nanopartículas de tierras raras, puntos cuánticos y nanopartículas de polímeros semiconductores de material orgánico, colorantes de moléculas pequeñas, materiales luminiscentes inducidos por agregación, etc. 1,17,18,19,20,21,22. El sistema de imágenes ópticas NIR-II también es un componente crítico de la bioimagen de fluorescencia porque puede recolectar eficazmente señales de fluorescencia NIR-II de la sonda NIR-II, lo que genera imágenes funcionales, anatómicas y moleculares precisas25,26. El sistema de imágenes NIR-II comprende principalmente cámaras infrarrojas de onda corta, filtros de paso largo (LP), láseres y procesadores de computadora. La tecnología de imagen NIR-II ha sido ampliamente utilizada en campos biomédicos como la detección de células cancerosas, imágenes dinámicas, rastreo dirigido in vivo y terapia dirigida, especialmente en investigación oncológica30,31. a bright nir-ii fluorescence probe for vascular and tumor imaging

Como tecnología de imagen emergente, las imágenes de fluorescencia de infrarrojo cercano II (NIR-II, 1000-1700 nm) tienen un potencial significativo en el campo biomédico, debido a su alta sensibilidad, penetración de tejido profundo e imágenes superiores con resolución espacial y temporal. Sin embargo, el método para facilitar la implementación de imágenes de fluorescencia NIR-II para algunos campos urgentemente necesarios, como la ciencia médica y la farmacia, ha desconcertado a los investigadores relevantes. Este protocolo describe en detalle la construcción y las aplicaciones de bioimagen de una sonda molecular de fluorescencia NIR-II, HLY1, con un esqueleto D-A-D (donante-aceptor-donante). HLY1 mostró buenas propiedades ópticas y biocompatibilidad. Además, las imágenes vasculares y tumorales NIR-II en ratones se realizaron utilizando un dispositivo de imágenes ópticas NIR-II. Se adquirieron imágenes de fluorescencia NIR-II de alta resolución en tiempo real para guiar la detección de tumores y enfermedades vasculares. Recopile las imágenes cinco minutos después de la inyección de puntos HLY1 en ratones y procese los datos utilizando el software ImageJ.La intensidad de fluorescencia de HLY1 en la solución de THF al 90% que contenía 90% de agua fue cinco veces mayor que en la solución de THF, lo que indica una emisión prominente inducida por agregación o característica AIE de HLY1. Los puntos HLY1 emitieron fuertes señales fluorescentes bajo un filtro de paso bajo o LP de 1.500 nanómetros estableciendo su idoneidad para imágenes NIR-IIb. La absorción máxima y la longitud de onda de emisión de los puntos HLY1 fueron de 740 nanómetros y 1.040 nanómetros, respectivamente.Además, se determinó que el tamaño hidrodinámico de los puntos HLY1 era de 145 nanómetros por dispersión dinámica de luz. En ratones administrados con puntos HLY1, los vasos cerebrales y los microvasos en la extremidad posterior se identificaron claramente mediante imágenes NIR-II bajo un filtro LP de 1.500 nanómetros. Además, los cuatro tumores T1 de los ratones portadores de tumores también fueron claramente visibles mediante imágenes NIR-II, lo que indica la permeabilidad mejorada y el efecto de retención o EPR de los puntos HLY1.Todos estos resultados sugirieron que los puntos HLY1 son una sonda de fluorescencia NIR-II brillante aplicable para imágenes vasculares y tumorales. La preparación de la nanosonda suspendible en agua bajo la imagen de fluorescencia in vivo y NIR-II es la parte más importante del procedimiento.
