1.从仔猪肠中分离上皮隐窝
注意：准备完整的Dulbecco改良鹰培养基（DMEMc），其中DMEM补充有10%胎牛血清（FBS）和1%青霉素链霉素（P / S）。准备50mL等分试样，并在4°C下储存1个月。

	</ol></li>

</ol>2. 冷冻上皮隐窝仔猪肠道三维类器官的建立
注意：仔猪肠道3D类器官在为人类类器官生长配制的商业培养基中培养，补充有1%P / S和100μg/ mL的原代细胞抗菌剂，并在4°C下储存长达1周。肿瘤来源的细胞外基质（ECM）用于3D类器官的培养。所有商业产品的参考文献均在 材料表中列出。

</ol>3. 来自冷冻隐窝的仔猪肠道3D类器官的通过
注意：从冷冻地穴获得类器官的时间通常比从新鲜地穴开始的时间长。类器官通常在解冻后10天准备好分裂（图2B）。

	<li>源自冷冻隐窝的3D类器官的传代<ol><li>取出培养基，并在37°C下用250μL预热的PBS洗涤。</li>

		<li>在冰上每 3，000 个活细胞中用 17 μL 冷 DMEMc 重悬细胞。将体积调整到所需的孔数。</li>
		<li>每 3，000 个活细胞缓慢加入 33 μL 带冷却吸头的冷 ECM，并在冰上匀浆而不产生气泡。将体积调整到所需的孔数。 注意：将细胞重悬于含有1/3 DMEMc和2/3 ECM的溶液中。对于每个圆顶，需要 50 μL 该溶液。稀释的ECM更便宜，更容易移液。</li>
		<li>在预热的 24 孔板中用冷却的吸头每孔用 50 μL ECM 细胞悬液接种孔。</li>
		<li>在37°C，5%CO2 培养箱中孵育30分钟，用于ECM聚合。</li>
		<li>每孔加入 500 μL 培养基。在37°C，5%CO 2培养箱中孵育，每2-3 天更换培养基 注意：类器官可以直接用于i）实验，ii）维持3D类器官培养，iii）冷冻或iv）类器官细胞单层的接种（图1）。每天用显微镜检查类器官的生长情况，以选择分裂类器官的最佳时间。类器官必须具有清晰空的管腔和清晰的边缘。不应使用管腔内有黑色碎片的成熟类器官进行分裂（图3）。</li>
	</ol></li>
</ol>4. 维持类器官培养的3D效果
注意：对于传代，类器官应清晰且腔为空。分裂后约5天，管腔中出现黑色碎片，表明存在死细胞。在传代时限制死细胞的数量对于培养物的最佳维持是优选的。因此，应调整时间表，以避免达到这一成熟阶段。

	<li>肠道3D类器官的通过<ol><li>用 P1000 移液管刮擦，用 ECM 分离类器官。通过在培养基中移液小心均质，并转移到含有 5 mL 冷 DMEMc 的 15 mL 锥形管中。 注意：一个 15 mL 锥形管用于在 24 孔板的 50 μL 圆顶中培养的 12 个类器官孔的池。</li>
		<li>将收集的类器官在4°C下以500× g 离心5分钟。 注意：类器官在管底部形成白色颗粒。如果离心后类器官仍在ECM中的悬浮液中，请小心吸出上清液（不接触ECM层），用P1000移液管移液10次均质化，然后重复离心步骤。在此过程之后，ECM 层应该不可见。</li>
		<li>小心吸出上清液并将细胞沉淀重悬于补充有 10 μM Y27632 ROCK 抑制剂的预热酶解离试剂中。上下移液10次以启动类器官的解离。</li>
		<li>在37°C水浴中孵育5分钟以进行酶消化。</li>
		<li>通过使用 P1000 移液管移液 10 次来机械破坏类器官。在显微镜下检查细胞悬液。 注意：目标是获得分离的细胞或小细胞簇。如果仍然观察到大的类器官片段，则重复孵育（步骤4.2.4）和机械破碎（步骤4.2.5）。</li>
		<li>加入 4 mL 冰冷的 DMEMc。在4°C下以500× g 离心5分钟</li>
		<li>弃去上清液并将类器官细胞沉淀重悬于 1 mL DMEMc 中。</li>
		<li>如上所述（步骤3.2.7至3.2.13）进行计数细胞并将类器官细胞接种在预热的24孔板中含有3000个活细胞的50μLECM圆顶中。</li>
	</ol></li>
</ol>5. 3D类器官的冷冻
<ol><li>准备必要体积（1 mL 用于两个圆顶池）含有补充有 10% FBS、10% DMSO 和 10 μM Y27632 ROCK 抑制剂的 DMEMc 的冷冻溶液，并储存在冰上（最终 FBS 浓度为 18%）。</li>
	<li>从要冷冻的孔中取出培养基。</li>
	<li>将1mL冷冻溶液加入第一个要冷冻的孔中，通过刮擦分离ECM，并用移液管尖端匀浆。</li>
	<li>将类器官悬浮液从第一个孔转移到第二个孔中冷冻。</li>
	<li>将两个孔的池转移到冷冻管中，并将小瓶放入细胞冷冻容器中。</li>
	<li>将细胞冷冻容器在-80°C下储存24小时，然后将小瓶转移到液氮中长期储存。</li>
</ol>6. 源自3D类器官的细胞单层培养
注意：猪类器官细胞的单层在2D培养基中培养，该培养基由补充有20%FBS的用于3D类器官的培养基组成。
<ol><li>材料准备<ol><li>准备 2D 介质并将其保持在 RT。</li>
		<li>在37°C的水浴中预热补充有10μM Y27632 ROCK抑制剂的酶解离试剂。</li>
	</ol></li>
	<li>培养物插入物的涂层<ol><li>对一把镊子进行消毒并将它们转移到生物安全柜中。</li>
		<li>将细胞培养插入物（0.33cm2）放入带有镊子的24孔板中。</li>
		<li>制备含有胶原蛋白IV的涂层溶液，在冷PBS中以50μg/ mL稀释。上下移液混合</li>
		<li>向每个细胞培养插入片段中加入 150 μL 稀释的胶原蛋白 IV 溶液，相当于 22.7 μg/cm2。 注意：小心地将移液管垂直定向到渗透膜的中心，并检查胶原蛋白溶液是否覆盖了所有膜。</li>
		<li>将板置于37°C，5%CO2 培养箱中，放置过夜（或至少3小时）。</li>
	</ol></li>
	<li>将3D类器官细胞接种到细胞培养插入物中<ol><li>从3D类器官制备细胞悬液，如步骤4.2.1至4.2.7中所述。</li>
		<li>用细胞计数器在台盼蓝中以1：2的稀释度计数细胞，并计算每个培养插入片段接种2.5 x 10 5个细胞所需的体积，对应于7.6 x 105个细胞/ cm2。</li>
		<li>在4°C下以500× g 离心所需体积的细胞悬液5分钟。</li>
		<li>在离心过程中，小心地从培养物中吸出包衣溶液，并在没有盖子的罩下室温干燥5分钟。</li>
		<li>离心后，弃去上清液并将细胞沉淀重悬于补充有10μM Y27632 ROCK抑制剂的所需体积的2D培养基中。每个插入片段需要 200 μL 含有细胞的 2D 培养基体积。</li>
		<li>将 200 μL 细胞悬液（2.5 x 105 个细胞）接种到包被的透膜（顶端侧）（图 4A）。 注意：在膜的中心缓慢移液并保持尖端垂直。</li>
		<li>将 500 μL 补充有 10 μM Y27632 ROCK 抑制剂的 2D 培养基添加到下隔室（基底侧）。在37°C，5%CO2 培养箱中孵育。</li>
		<li>播种一天后，用不含Y27632 ROCK抑制剂的新鲜2D培养基替换顶端和基底培养基。</li>
		<li>每天更换 2D 介质。单层在接种后1天变得汇合，然后可用于实验。 注意：空白（无细胞插入）上方的跨上皮电阻（TEER）值确认达到汇合度（图4B）。</li>

</ol>7. 类器官细胞单层的免疫染色
<ol><li>溶液的制备 注意：根据要染色的孔数调整溶液的体积;一个孔的每个步骤都需要 200 μL 溶液。 材料表中提供了所有商业产品的参考资料。<ol><li>通过将 5 mL 的 32% PFA 添加到 35 mL 的 PBS 中，在化学罩下制备 4% 多聚甲醛 （PFA） 溶液。准备10mL等分试样并储存在-20°C。 注意：始终在化学罩下操作PFA，同时戴丁腈手套。</li>
		<li>制备 0.2% Triton X100-PBS 的溶液，在使用前立即将 2 μL Triton X100 加入 1 mL PBS 中。保持在室温。</li>

类器官的成熟阶段也是获得细胞单层的重要点。具有高级成熟的类器官（由腔中存在黑细胞碎片表示）不是接种单层的最佳选择。我们通常在传代后4天解离3D类器官以收集细胞进行2D培养。需要大约一到三个以每50μL圆顶3，000个细胞培养的3D类器官孔，以接种一个具有2.5 x 105细胞的培养插入物。细胞在 1 天内附着并形成完全融合的单层（图 4A），这由约 700 Ω·cm2 的高 TEER 证实（图 4B）。然而，在这个早期时间点，细胞边缘很难通过明场显微镜可视化，可能是由于分化水平低。TEER 在 3 天内保持高位（图 4B）。
