Een continustroomprotocol van de Balz-Schiemann-reactie is met succes uitgevoerd door een combinatie van een microkanaalstroomreactor en een dynamisch gemengde stroomreactor. Deze strategie heeft verschillende voordelen ten opzichte van het batchproces: (i) het is veiliger met gecontroleerde diazoniumzoutvorming; ii) het is beter vatbaar voor een hogere reactietemperatuur, 10 °C versus -20 °C; en (iii) het is efficiënter zonder isolatie van het diazonium intermediair, twee stappen in één continu proces. De Balz-Schiemann-reactie is een klassieke methode om de diazoniumgroep te vervangen door fluor door ArN2+BF4− te verhitten zonder oplosmiddel 1,2. De reactie kan worden toegepast op een breed scala aan arylaminesubstraten, waardoor het een algemeen toepasbare benadering is om arylaminen te synthetiseren, die vaak worden gebruikt voor geavanceerde tussenproducten in de farmaceutische of fijnchemische industrie 2,3. Helaas worden harde reactieomstandigheden vaak gebruikt in de Balz-Schiemann-reactie en genereert de reactie potentieel explosieve aryldiazoniumzouten 4,5,6,7,8. Andere uitdagingen in verband met de Balz-Schiemann-reactie zijn de vorming van bijproducten tijdens het thermische ontledingsproces en de bescheiden opbrengst ervan. Om de vorming van het bijproduct te minimaliseren, kan thermische dediazotisatie worden uitgevoerd in niet-polaire oplosmiddelen of met behulp van nette diazoniumzouten 9,10, wat betekent dat de aryldizaniumzouten moeten worden geïsoleerd. De diazotisatie van aromatische amines is echter over het algemeen exotherm en snel, wat een risico is dat gepaard gaat met de isolatie van het explosieve diazoniumzout, vooral bij grootschalige productie.
In de afgelopen jaren hebben continue stroomsynthesetechnologieën bijgedragen aan het overwinnen van de veiligheidsproblemen in verband met de Balz-Schiemann-reacties11,12. Hoewel er enkele voorbeelden zijn van diazotisatie van aromatische amines met behulp van continue microreactoren voor deaminering op posities para tot arylchloriden, 5-azodyes en chloorsulfonylering, werden deze bijdragen alleen gerapporteerd op laboratoriumschaal 13,14,15,16,17. Yu en collega's ontwikkelden een continu proces op kiloschaal voor de synthese van arylfluoride18. Ze hebben aangetoond dat de verbeterde warmte- en massaoverdracht van een stromingssysteem zowel het diazotisatieproces als het fluorisatieproces ten goede zou komen. Ze gebruikten echter twee afzonderlijke continue stroomreactoren; daarom werden de diazotisatie- en thermische ontledingsprocessen afzonderlijk onderzocht. Een andere bijdrage werd gepubliceerd door Buchwald en collega's19, waar zij een hypothese presenteerden dat als de productvorming door het SN2Ar- of SN1-mechanisme ging, de opbrengst kan worden verbeterd door de concentratie van de fluoridebron te verhogen. Ze ontwikkelden een flow-to-continuous stirred tank reactor (CSTR) hybride proces waarbij de diazoniumzouten op een continue en gecontroleerde manier werden gegenereerd en geconsumeerd. De warmte- en massaoverdrachtsefficiëntie van een CSTR is echter niet goed genoeg als buisstroomreactor en van een grote CSTR kan niet worden verwacht dat deze wordt gebruikt met explosieve diazoniumzouten in grootschalige productie. Vervolgens ontwikkelden Naber en collega's een volcontinu stroomproces om 2-fluoroadenine te synthetiseren uit 2,6-diaminopurine20. Ze ontdekten dat de exotherme Balz-Schiemann-reactie gemakkelijker te regelen was op een continue stroommanier en dat de buisafmetingen van de stroomreactor de warmteoverdracht en temperatuurregelingsaspecten zouden beïnvloeden - een buisreactor met grote afmetingen vertoont een positieve verbetering. Het opgeschaalde effect van de buisreactor zal echter opmerkelijk zijn en de slechte oplosbaarheid van het polaire aryl diazoniumzout in organische oplosmiddelen is lastig voor statische buisreactoren, die een verstoppingsrisico lopen. Hoewel er opmerkelijke vooruitgang is geboekt, zijn er nog steeds enkele problemen in verband met grootschalige Balz-Schiemann-reacties. De ontwikkeling van een verbeterd protocol dat snelle en schaalbare toegang tot arylfluoride zou bieden, is dus nog steeds belangrijk.
De uitdagingen in verband met grootschalige Balz-Schiemann-reactieverwerking omvatten het volgende:(i)de thermische instabiliteit van een geaccumuleerd diazoniumtussenproduct gedurende een korte periode21; ii) de lange verwerkingstijden; en iii) de niet-uniforme verwarming of de aanwezigheid van water in het diazoniumfluooboraat, wat leidt tot oncontroleerbare thermische ontleding en verhoogde vorming van bijproducten22,23. Bovendien (iv) is in sommige stroomverwerkingsmodi nog steeds een isolatie van het diazoniumtussenproduct vereist vanwege de lage oplosbaarheid14, die vervolgens wordt ingevoerd in een ongecontroleerde snelheidsafbraakreactie. Het risico van het hanteren van een grote hoeveelheid in-line diazoniumzout kan niet worden vermeden. Er is dus een aanzienlijk voordeel bij het ontwikkelen van een continue stroomstrategie om de bovengenoemde problemen op te lossen en zowel de accumulatie als de isolatie van de onstabiele diazoniumsoort te voorkomen.
Om een inherent veiligere productie van chemicaliën in farmaceutische producten tot stand te brengen, heeft onze groep zich gericht op multi-step continuous flow-technologie. In dit werk passen we deze technologie toe op de Balz-Schiemann-synthese op kilogramschaal op een manier die de isolatie van aryl diazoniumzouten elimineert en tegelijkertijd efficiënte fluorinatie mogelijk maakt.

De vraag naar aromatische fluoriden neemt gestaag toe in de farmaceutische en fijnchemische industrie. De Balz-Schiemann-reactie is een eenvoudige strategie voor het bereiden van arylfluoride uit arylaminen, via de bereiding en omzetting van diazoniumtetrafluorboraattussenproducten. Er bestaan echter aanzienlijke veiligheidsrisico's bij het hanteren van de aryl diazoniumzouten bij het opschalen. Om het gevaar te minimaliseren, presenteren we een continu stroomprotocol dat met succes is uitgevoerd op een kilogramschaal dat de isolatie van aryl diazoniumzouten elimineert en tegelijkertijd efficiënte fluorinatie vergemakkelijkt. Het diazotisatieproces werd uitgevoerd bij 10 °C met een verblijftijd van 10 min, gevolgd door een fluoreringsproces bij 60 °C met een verblijftijd van 5,4 s met ongeveer 70% opbrengst. De reactietijd is drastisch verminderd door de introductie van dit meerstaps continue stroomsysteem.

De modelreactie is weergegeven in figuur 2. 2-Methylpyridin-3-amine (verbinding 1 in figuur 2) werd gekozen als uitgangsmateriaal voor de bereiding van 2-methylpyridine-3-fluoride (verbinding 3 in figuur 2) via de Balz-Schiemann-reactie. Lees eerst alle stappen en zoek de details van het werken uit voordat u deze techniek probeert. Om de Balz-Schiemann-reactie in een continustroomsysteem uit te voeren, bereidt u twee modules van de microstroomreactor voor met een intern reactievolume van negen milliliter en één dynamische mengbuisreactor met een intern reactievolume van 500 milliliter.Bereid ook een pomp met constante stroom voor met een PTFE-pompkop en drie pompen met constante stroom met de roestvrijstalen pompkop van 316 L. Monteer de apparatuur zoals beschreven in het manuscript. Controleer de mechanische integriteit van alle verbindingen tussen pompen, pijpleidingen en stroomreactoren voor gebruik.Stel de debieten van de pompen in. Stel vervolgens de jacket outlet, temperatuur van de premixing en diazonium zoutvormingszone in op min vijf graden Celsius en de jacket outlet temperatuur van de thermische ontledingszone op 60 graden Celsius om de temperatuurregeling te handhaven. Om de veiligheid en lekkage van apparatuur te controleren, plaatst u eerst de doseerpijplijn van pompen A, B, C en D in de feed E-fles.En het lozen van een pijpleiding in de afvalinzamelfles. Start vervolgens de pompen en regel langzaam de tegendruk tot drie bar. Observeer de stabiliteit van elke pomp en controleer alle verbindingen, pijpleidingen en reactoren op eventuele oplosmiddellekkage.Controleer ook of de inlaat- en uitlaattemperatuur van elke zone en de huidige inlaatdruk van elke pomp binnen het doelbereik liggen. Stop vervolgens de pompen na 10 minuten steady state evenwicht. Om het reactieproces te starten, plaatst u de A, B, C en D van de doseerleiding in de bijbehorende feeds en plaatst u de afvoerpijplijn in de afvalinzamelfles.Start pompen A en B tegelijkertijd en noteer de tijd. Start vervolgens pomp C na 30 seconden en pomp D na 8 minuten. Plaats na 10 minuten steady state evenwicht de ontladingspijplijn in het productverzamelingsvat.Observeer en registreer de temperatuur van elke zone en de druk van elke pomp. Zodra het pompen van voer B is voltooid, plaatst u de doseerleiding B in voer E.Plaats vervolgens de afvoerpijp in een afvalinzamelfles. En het doseren van pijplijnen, A, C en D in de feed E fles.Schakel de pompen A, B, C en D uit na 10 minuten wassen. Een continue stroomreactor werd met succes toegepast op de Balz-Schiemann-reactie op een modelsubstraat. De reactietemperatuur van de stroomexperimenten werd met succes gecontroleerd op 10 graden Celsius met 70% zuiverheid van het ruwe product, wat hoger was dan wat werd verkregen in batchverwerking.Bijna 98% van de conversie werd bereikt in 600 seconden van de tijd van de bewoner. Wanneer het debiet 50 milliliter per minuut was in de diazotisatiezone en 5,4 seconden in de thermische ontledingszone, bij een stroomsnelheid van 100 milliliter per minuut. Het verhogen van het debiet laat een grote hoeveelheid uitgangsmateriaal achter in de reactie en het verlagen van het debiet kan het uitgangsmateriaal volledig verbruiken, maar de productie-efficiëntie beperken.Het ruwe product werd verder gezuiverd en geanalyseerd met behulp van massaspectroscopie, proton-NMR en fluor-NMR. Het is belangrijk om aan te tonen dat de reactoren pompen zijn op basis van de chemische eigenschap en opneembaarheid van stoffen, vooral voor degenen die sterk of voor die reacties met vaste stofvorming zijn. Reacties die een vloeibaar effect en een snellere reactiesnelheid hebben, kunnen met behulp van een continustroomsysteem effect sorteren.Bovendien kan het een reactie veroorzaken die al dan niet kan resulteren in de vorming van een vaste stof.
