Macroautofagie (gewoonlijk autofagie genoemd) is een gespecialiseerd katabolisch proces dat selectief cytoplasmatische componenten afbreekt, waaronder eiwitten en beschadigde organellen 1,2. Autofagie stelt cellen in staat om fysiologisch te reageren op stressprikkels en zo cellulaire homeostase3 te behouden. Tijdens autofagie wordt een dubbel membraanblaasje gevormd: het autofagosoom. Het autofagosoom bevat de ladingmoleculen en drijft ze naar het lysosoom voor afbraak 1,4.
Autofagosomen worden versierd door het autofagische eiwit microtubule-geassocieerde eiwit lichtketen 3 (LC3)5. Wanneer autofagie niet wordt geïnduceerd, wordt LC3 diffuus in het cytoplasma en de kern in de LC3-I-conformatie. Aan de andere kant, wanneer autofagie wordt geïnduceerd, wordt LC3 geconjugeerd met een fosfatidylethanolamine in het membraan van de autofagische structuren6. Deze nieuwe LC3-conformatie staat bekend als LC3-II1. De LC3-conformatieverschuiving veroorzaakt veranderingen in de cellulaire lokalisatie en de migratie van dodecylnatriumsulfaat-polyacrylamidegelelektroforese (SDS-PAGE), die kan worden gedetecteerd door technieken zoals immunofluorescentie en western blot 5,7. Op deze manier is LC3-conjugatie een belangrijke gebeurtenis in het autofagische proces die kan worden gebruikt om autofagische activiteit te meten.
De pancreasacinaire cel is een sterk gedifferentieerde cel die, onder gezonde omstandigheden, een lage mate van autofagie heeft. In verschillende fysiologische omstandigheden of onder farmacologische stimulatie kunnen ze echter autofagie activeren. Daarom is de bepaling van autofagische niveaus in deze cellijn nuttig voor het bestuderen van de potentiële directe of indirecte effecten van verschillende farmacologische of biologische agentia op autofagie 8,9.
Ductaal adenocarcinoom van de alvleesklier is een van de dodelijkste vormen van kanker, gezien de late diagnose en de hoge chemotherapieresistentie10. Alvleesklierkankercellen hebben een hoge autofagie-activiteit als gevolg van de transcriptionele upregulatie en posttranslationele activering van autofagie-gerelateerde eiwitten11. Alvleesklierkankercellen kunnen hun autofagieniveaus aanpassen als reactie op ongunstige omstandigheden zoals hypoxie, tekort aan voedingsstoffen of door chemotherapie veroorzaakte schade11. Daarom kan het analyseren van de autofagieniveaus in alvleesklierkankercellen helpen begrijpen hoe ze zich aanpassen aan verschillende omgevingen en de effectiviteit van autofagiemodulerende behandelingen evalueren.
Deze studie toont een methode om LC3-immunofluorescentie uit te voeren in twee verschillende pancreascellulaire modellen. Het eerste model, PANC-1-cellen, diende als model voor ductaal adenocarcinoom van de alvleesklier. Deze cellen werden behandeld met gemcitabine, een chemotherapeuticum waarvan eerder is aangetoond dat het autofagie induceert, met name in alvleesklierkankercellen die het oncogene Kirsten rat sarcoma virus-gen (KRAS)12,13 dragen. Het tweede model, AR42J-cellen, diende als een meer fysiologisch model van exocriene pancreascellen. Deze cellen werden gedifferentieerd met dexamethason om meer op acinaire pancreascellen te gaan lijken14. In deze cellen werd autofagie farmacologisch geïnduceerd door het gebruik van PP242, een krachtige mTOR-remmer15. In deze studie tonen we de toepasbaarheid aan van het protocol beschreven met twee verschillende pancreasmodellen en het vermogen om onderscheid te maken tussen toestanden van lage en hoge autofagie.

Autofagie is een gespecialiseerd katabolisch proces dat selectief cytoplasmatische componenten afbreekt, waaronder eiwitten en beschadigde organellen. Autofagie stelt cellen in staat om fysiologisch te reageren op stressprikkels en zo cellulaire homeostase te behouden. Kankercellen kunnen hun autofagieniveaus moduleren om zich aan te passen aan ongunstige omstandigheden zoals hypoxie, tekort aan voedingsstoffen of schade veroorzaakt door chemotherapie. Ductaal adenocarcinoom van de alvleesklier is een van de dodelijkste vormen van kanker. Alvleesklierkankercellen hebben een hoge autofagie-activiteit als gevolg van de transcriptionele upregulatie en posttranslationele activering van autofagie-eiwitten.
Hier werd de PANC-1-cellijn gebruikt als een model van menselijke kankercellen van de alvleesklier en de AR42J pancreas-acinaire cellijn werd gebruikt als een fysiologisch model van sterk gedifferentieerde zoogdiercellen. Deze studie gebruikte de immunofluorescentie van microtubule-geassocieerde eiwit lichtketen 3 (LC3) als een indicator van de status van autofagie-activering. LC3 is een autofagie-eiwit dat in basale omstandigheden een diffuus distributiepatroon vertoont in het cytoplasma (bekend als LC3-I in deze aandoening). Autofagie-inductie activeert de conjugatie van LC3 tot fosfatidylethanolamine op het oppervlak van nieuw gevormde autofagosomen om LC3-II te vormen, een membraangebonden eiwit dat helpt bij de vorming en uitbreiding van autofagosomen. Om het aantal gelabelde autofagische structuren te kwantificeren, werd de open-source software FIJI gebruikt met behulp van de "3D Objects Counter" -tool.
De meting van de autofagische niveaus, zowel in fysiologische omstandigheden als in kankercellen, stelt ons in staat om de modulatie van autofagie te bestuderen onder verschillende omstandigheden zoals hypoxie, chemotherapiebehandeling of de knockdown van bepaalde eiwitten.

Dit protocol voert immunofluorescentie van LC3 uit in pancreascellijnen om de autofagieniveaus in verschillende omstandigheden te bepalen. Het resultaat van dit experiment was de obtentie van cellulaire beelden van de rode en blauwe kanalen, overeenkomend met LC3 en DAPI. De LC3-beelden geven de cellulaire verdeling van dit eiwit aan, terwijl de DAPI de nucleaire lokalisatie toont. Evaluating Autophagy Levels in Two Different Pancreatic Cell Models Using LC3 Immunofluorescence

Het doel van dit protocol is om autofagische niveaus in alvleesklierkanker en pancreasacinaire cellen te bepalen door middel van LC3-immunofluorescentie en LC3-puntkwantificering.

De LC3-immunofluorescentie maakt de bepaling van autofagieniveaus in pancreascellen mogelijk. Het maakt de studie mogelijk van de potentiële effecten van verschillende middelen op pancreascelautofagie. Immunofluorescentie is een techniek die wordt gebruikt om endogene LC3-eiwitten te detecteren, die problemen vermijdt die worden veroorzaakt door overexpressie van LC3, zoals het vormen van eiwitaggregaten die geen verband houden met autofagie.Felipe Renna, promovendus, en Malena Herrera Lopez, promovendus uit het laboratorium van Maria Ines Vaccaro, zullen de procedure demonstreren. Om de celvoorbereiding te starten, weekt u 12 millimeter ronde afdekslips in absolute ethanol. Nadat u de hoes hebt verwijderd, plaatst u de doorweekte afdekplaat verticaal in de 24-putplaat.Stel de multi-well plaat gedurende 15 minuten bloot aan ultraviolette straling. Plaats vervolgens de afdekslip horizontaal en was deze met DMEM. Zie nu het lage doorgangsaantal pancreascellen, opnieuw gesuspendeerd in DMEM, met 10% FBS, penicilline en streptomycine, en incuber de cellen gedurende twee dagen.Bereid twee dagen na het zaaien van de cel gemcitabine-oplossing in DMEM met één microgram per microliterconcentratie. Zuig vervolgens de helft van het volume van elke put in een aparte buis en voeg de juiste hoeveelheid gemcitabine-oplossing toe. Breng het halve volume dat overeenkomt met elke put terug in de behandelde putten en incuber de cellen gedurende 24 uur.Voeg voor het fixeren en permeabiliseren van de cellen koude methanol toe aan een 24-putplaat met cellen. Maak ook een bord met zes putten met koude PBS. Neem met behulp van een spuitnaald en een pincet elke dekstrook en was deze twee keer in PBS.Incubeer vervolgens gedurende zes minuten in methanol. Na twee keer wassen met PBS, incubeer je de afdekslip gedurende een uur in een blokkerende oplossing. Voeg een anti LC3 toe in de blokkerende oplossing in een verhouding van één tot 1000 en onderhoud op ijs.Plaats een stuk laboratoriumafdichtingsfolie over het deksel met meerdere putten en voeg 25 microliter anti-LC3-oplossing per afdeklaag over de afdichtingsfolie toe. Plaats met behulp van een spuitnaald en een pincet elke afdekslip over de primaire antilichaamdruppel en zorg ervoor dat de celzijde in contact is met de oplossing. Plaats de multi-well plaat in de vochtigheidskamer.Bedek het met folie en incubeer een nacht in de koelkast. Verwijder de volgende dag de multi-well plaat uit de vochtigheidskamer. Plaats de afdekplaten terug op de multi-well plaat en was ze drie keer met PBS.Verdun fluorescerend gelabeld anti-konijn in een blokkerende oplossing in een verhouding van één tot 800 en blijf op ijs in het donker. Voeg na het afdichten van het meerputtige deksel 25 microliter anti-konijnoplossing per afdeklaag toe aan de afdichtingsfolie. En behandel de dekslip met een secundair antilichaam zoals eerder aangetoond.Incubeer de plaat vervolgens gedurende twee uur in de vochtigheidskamer bij kamertemperatuur, beschermd tegen licht. Aan het einde van de incubatie met antilichaam, was de afdekslip op de multi-well plaat met PBS drie keer. Incubeer elke afdekplaat met de voorbereide DAPI-oplossing gedurende 10 minuten.Eenmaal gewassen met PBS, onderhoudt u de multi-well plaat in het donker. Bereid voor celmontage twee bekers voor met water en een stuk papier. Voeg 10 microliter PVA-DABCO-oplossing per afdeklaag toe aan een dia.Was de afdeklaag in water en droog deze vervolgens op papier. Plaats het ten slotte over de PVA-DABCO-druppel. En 's nachts droog in het donker.De volgende dag visualiseert u met behulp van een omgekeerde confocale microscoop de gelabelde cellen en legt u de beelden vast. Nadat u afbeeldingen hebt vastgelegd, sleept u elk afbeeldingsbestand met de vastgelegde kanalen in het Fiji-scherm. Klik in het dialoogvenster op OK om de afbeelding te openen.Sluit vervolgens het geopende consolevenster. Selecteer op het tabblad Afbeelding de optie Kleur en vervolgens Kanalen splitsen. Sluit de afbeelding die overeenkomt met de andere kanalen dan de LC3-afbeelding.Selecteer vervolgens op het tabblad Afbeelding de optie Aanpassen, gevolgd door Kleurbalans. Verplaats de maximale schuifregelaar naar links totdat de afbeelding verzadigd is om de celcontouren te visualiseren. Gebruik het selectiegereedschap uit de vrije hand om de celomtrek te tekenen en klik op Opnieuw instellen om de kleuren aan te passen.Als u het geselecteerde item van het tabblad Bewerken wilt knippen, selecteert u Knippen en sluit u de afbeelding zonder deze op te slaan. En selecteer op het tabblad Bewerken de optie plakken. Kies in het menu Analyseren het gereedschap 3D-objectenteller.Stel de drempel in op 2000 en het groottefilter op 50 tot 500. Zorg ervoor dat de objecten en samenvattingsvakken zijn gemarkeerd en klik op OK.In het logboekvenster, het aantal stippen wordt beschreven als gedetecteerde objecten. De immunofluorescentie gaf de cellulaire verdeling van LC3-eiwit aan, terwijl de DAPI nucleaire lokalisatie in pancreascellen vertoonde.De LC3-stippen namen significant toe onder behandeling met gemcitabine, wat wijst op autofagische activiteit. Onder een overmatige samenvloeiing hebben de exocriene alvleeskliercellen de neiging zich op te stapelen en op elkaar te groeien. De paraformaldehydecelfixatiemethode was niet effectief voor het behoud van LC3-eiwitten, omdat deze geen nauwkeurige verdeling weerspiegelde.Wanneer fixatie of behandeling werd uitgevoerd zonder twee dagen na het zaaien te wachten, hadden de PANC1-cellen een ronde vorm. Deze morfologie verminderde de relatie tussen het cytoplasma en de kern, waardoor het moeilijk werd om de intracellulaire verdeling van LC3 te begrijpen. Onvolledige methanolfixatie kan de juiste LC3-immunolabeling verstoren, wat leidt tot onduidelijke beelden en suboptimale kwantificering.Het belangrijkste om te onthouden tijdens deze procedure is dat de cellen gedurende zes minuten in koude methanol moeten worden gefixeerd, in plaats van paraformaldehyde. Volgens deze procedure kan colokalisatie tussen LC3 en andere eiwitten worden geanalyseerd, waardoor de studie van verschillende moleculaire routes gerelateerd aan LC3-functies mogelijk is.
