プロトコルで使用される材料の一部は、ポリプロピレンではなくガラスフライバイアルを使用するなど、代替することができます。ただし、使用される材料は、試薬間の不要な化学反応やガラス製品の洗浄に起因する可能性のある化学物質への暴露を回避するために、不活性で使い捨ての両方になるように選択されました。
液体食品の使用は、食品配達のための車両を必要とする。セルロースアセテート濾紙は、その柔軟性および不活性な性質のためにこの目的のために選択された28。他の研究者も同様のプロトコルを使用しましたが、繊細なタスクワイプやグラスファイバーフィルターなどの他の車両を使用しました29,30。セルロースアセテート濾紙は、紙とバイアルの壁の間に大きな隙間なくフライバイアルの底に収まる理想的な形状に切断できる不活性ビヒクルであり、ハエが媒体や車両自体に詰まることによる死亡を防ぐため、これらのニーズに適合しました。
このシステムの重要な制限は、化学物質の最大試験可能濃度が化学物質の溶解度に結びついていることです。非水溶性化合物は追加の溶媒を必要とし、これは目的の化学物質とのさらなるまたは相乗効果をもたらし得る。これはまた、全ての生物において所望のエンドポイントを達成するのに十分濃縮されたストック溶液を調製することができない状況を作り出すことができ、したがって、結果として得られるデータ31の分析を制限する。これに対処するために、水溶性の低い化学物質は、食品溶液に最大0.5%のジメチルスルホキシドを添加することによってテストできます。他の溶媒も使用できますが、生物への溶媒の影響を最小限に抑えながら溶解度を最大化するために、溶液内の最大許容溶媒濃度を決定するには、関心のある各溶媒について追加の研究が必要です。
ショウジョウバエの嗅覚応答の広範な特徴付けは、ハエが有毒な化合物40,41の消費をどのように回避し、処理された培地への摂食の減少につながるかを説明しています。青色色素アッセイは、研究者が実験化学物質42,43,44の各濃度を与えられたハエの摂食行動を効率的にスクリーニングできるようにすることで、この現象に対処します。ハエの胃腸管の青の有無は、ハエが毒物含有培地を食べていたかどうかを示します。ハエの摂食行動を評価するためのより洗練された方法、例えば、フライ液体食品相互作用カウンター45が存在するが、この定性的方法は、よりハイスループットのスクリーニングにより適している。
このプロトコルの注目すべき側面は、ハエを移したり、露光バイアルに液体を追加したりすることなく、48時間の曝露期間に最適化されていることです。湿度チャンバーを使用し、チャンバーを高湿度に保たれたインキュベーターに置くことで、この期間中に供給媒体を含むろ紙が乾燥するのを防ぎました。プロトコルはより長い暴露時間に適応させることができますが、ろ紙が乾燥して溶液濃度の大幅な変化や乾燥による致死性を引き起こさないように方法を調整する必要があります。

ショウジョウバエ32,33,34,35,36における亜ヒ酸ナトリウムの毒性に関するさまざまな先行研究のため、オレゴンRハエはこの化合物で処理され、システムの有効性が実証されました。オスのハエは0.65 mMのLD50を示し、メスは0.90 mMのLD50を示しました。これは、亜ヒ酸ナトリウムで処理された成体ショウジョウバエの以前の研究と一致しています。例えば、GoldsteinとBabich37は、ハエ(男女混合)の50%が0.5mM NaAsO2に7日間曝露した後に死亡したことを発見した。これは現在観察されているよりもわずかに低い線量ですが、それらの方法とこの方法の違い(固体曝露媒体の使用、より長い時間スケール、および男女混合を含む)がこの違いを説明している可能性があります。重要なことに、両方の方法が全体的に類似したLD50値をもたらした。

人間は、空気1、食品2、水3,4、医薬品5、洗浄剤6、パーソナルケア製品7、工業用化学物質7、建築材料7など、さまざまなソースからの化学物質に絶えずさらされています。さらに、毎年何千もの新しい化学物質が導入されており8、その多くは健康と環境の安全性について適切に審査されていません。この適切な化学的安全性試験の欠如は、マウスやラットなどの哺乳類モデルへの過度の依存に一部起因しています。このようなげっ歯類モデルは有益であるが、これらのシステムにおける化学的安全性試験は、高価で時間がかかり、しばしば試験動物に許容できないレベルの苦痛を引き起こす9。

ハエは長い間、亜ヒ酸ナトリウム(NaAsO2)の毒性を測定するための研究のモデルとして役立ってきました35,36,37,38。プロトコルの有効性を実証するために、これらの結果を以前の研究と比較することを目的として、オスおよびメスのハエをNaAsO2に曝露した。 上記の方法論を用いて、成人オレゴン-R(BDSCストック#2057)の雄および雌を、ある範囲のNaAsO2濃度(0、0.01、0.02、0.1、0.2、1、および2mM)に曝露し、曝露開始後48時間で致死率をスコア化した(図1A、B)。
この初期分析の目的は、NaAsO2毒性のより正確な特徴付けを可能にする濃度のおおよその範囲を特定することであった。その後の実験において、NaAsO 2用量反応曲線をより正確に定義する濃度(0、0.2、0.5、1、1.5、2、2.5、3、3.5、および5mM)を選択した(図1C、D)。結果として得られた分析は、100%の致死性を誘発するいくつかの濃度を調べたことに注意してください。データは、環境保護庁の公開されているベンチマーク線量ソフトウェアバージョン3.2.0.125を使用して分析されました。データは「二分」としてモデル化され、二分ヒルモデルはその後の分析に使用されました。このモデルに基づいて、NaAsO2を給餌したオスのハエの最終的なLD10、LD25、およびLD50は、それぞれ0.30 mM、0.50 mM、および0.65 mMでした。メスのハエの場合、これらの値はわずかに高く、LD10は0.30 mM、LD25は0.65 mM、LD50は0.90 mMでした。全体として、この方法を使用して得られた値は、ショウジョウバエメラノガスター35,36,37,38におけるヒ素毒性について以前に報告されたものと同様であり、したがって方法論を検証します。
線量反応曲線の計算に用いた6回の反復に加えて、オスとメスのハエには、光学顕微鏡を使用して消化管で容易に見える1%FD&Cブルーを含むNaAsO2曝露溶液も与えられました。NaAsO 2を給餌したハエの腸内の青色色素の存在に基づいて、液体培地中に存在するNaAsO 2濃度に関係なく、オスとメスの両方のハエが化学物質曝露の開始後24時間摂食し続けました(図2)。 しかし、摂取した食物は、女性で0.2 mM、男性で0.5 mMを超える用量で時折逆流することが観察されました(図2)。これらの知見は、逆流がショウジョウバエのヒ素中毒に対する反応に重要な役割を果たす可能性があることを示唆している。
