In de afgelopen jaren zijn methoden voor een goede EV-isolatie steeds belangrijker geworden, waardoor hun verdere betrouwbare analyses mogelijk zijn, bijvoorbeeld in het kader van het verkrijgen van betrouwbare omica en functionele gegevens24. Op basis van eerdere onderzoekservaring lijkt het erop dat niet alleen het type isolatiemethode, maar ook andere aandoeningen tijdens deze procedure belangrijk kunnen zijn. Extracellulaire blaasjes (EV's), volgens de MISEV 2018-nomenclatuur, zijn een verzamelnaam die verschillende subtypen van door cellen uitgescheiden vliezige blaasjes beschrijft die een cruciale rol spelen in tal van fysiologische en pathologische processen 1,2. Bovendien zijn EV's veelbelovend als nieuwe biomarkers voor verschillende ziekten, evenals therapeutische middelen en voertuigen voor medicijnafgifte. Het realiseren van hun volledige potentieel is echter moeilijk omdat er nog steeds veel technische obstakels zijn met betrekking tot hun overname3. Een van die uitdagingen is de isolatie van endotoxinevrije EV's, die in veel gevallen is verwaarloosd. Een van de meest voorkomende endotoxinen is lipopolysaccharide (LPS), dat een belangrijk onderdeel is van gramnegatieve bacteriële celwanden en een acute ontstekingsreactie kan veroorzaken, als gevolg van de afgifte van een groot aantal inflammatoire cytokines door verschillende cellen 4,5. LPS induceert een respons door binding aan LPS-bindend eiwit, gevolgd door interactie met het CD14/TLR4/MD2-complex op myeloïde cellen. Deze interactie leidt tot de activering van MyD88- en TRIF-afhankelijke signaalroutes, die op hun beurt de nucleaire factor kappa B (NFkB) activeren. Translocatie van NFkB naar de kern initieert de productie van cytokines6. Monocyten en macrofagen zijn zeer gevoelig voor LPS en hun blootstelling aan LPS resulteert in een afgifte van inflammatoire cytokines en chemokines (bijv. IL-6, IL-12, CXCL8 en TNF-α)7,8. De CD14-structuur maakt de binding van verschillende LPS-soorten met vergelijkbare affiniteit mogelijk en dient als co-receptor voor andere toll-like receptoren (TLRs) (TLR1, 2, 3, 4, 6, 7 en 9)6. Het aantal studies dat wordt uitgevoerd naar de effecten van EV's op monocyten/macrofagen neemt nog steeds toemet 9,10,11. Vooral vanuit het perspectief van het bestuderen van de functies van monocyten, hun subpopulaties en andere immuuncellen, is de aanwezigheid van endotoxine en zelfs hun gemaskeerde aanwezigheid in EV's van groot belang12. De over het hoofd geziene besmetting van EV's met endotoxinen kan leiden tot misleidende conclusies en hun ware biologische activiteit verbergen. Met andere woorden, het werken met monocytische cellen vereist vertrouwen in de afwezigheid van endotoxinebesmetting13. Potentiële bronnen van endotoxinen kunnen water, commercieel verkregen media en sera, mediacomponenten en additieven, laboratoriumglaswerk en plasticwarezijn 5,14,15.
Daarom was deze studie gericht op het ontwikkelen van een protocol voor de isolatie van laag-endotoxine-bevattende EV's. Het protocol geeft eenvoudige tips over hoe endotoxinebesmetting tijdens EV-isolatie kan worden voorkomen in plaats van endotoxinen uit EV's te verwijderen. Eerder zijn veel protocollen gepresenteerd over het verwijderen van endotoxinen uit bijvoorbeeld gemanipuleerde nanodeeltjes die worden gebruikt in de nanogeneeskunde; geen van hen is echter nuttig voor biologische structuren zoals EV's. De effectieve depyrogenering van nanodeeltjes kan worden uitgevoerd door ethanol- of azijnzuurspoeling, verwarming bij 175 °C gedurende 3 uur, γ bestraling of triton X-100-behandeling; deze procedures leiden echter tot de vernietiging van EV's16,17.
Het gepresenteerde protocol is een baanbrekend onderzoek gericht op het vermijden van endotoxine onzuiverheden in EV's, in tegenstelling tot eerdere studies over het effect van EV's op monocyten9. Het toepassen van voorgestelde principes op de laboratoriumpraktijk kan helpen om betrouwbare onderzoeksresultaten te verkrijgen, wat cruciaal kan zijn bij het overwegen van het potentiële gebruik van EV's als therapeutische middelen in de kliniek12.

isolation of low endotoxin content extracellular vesicles derived from cancer cell lines

Extracellulaire blaasjes (EV's) zijn een heterogene populatie van membraanblaasjes die door cellen in vitro en in vivo worden vrijgegeven. Hun alomtegenwoordigheid en belangrijke rol als dragers van biologische informatie maken hen tot intrigerende studieobjecten, die betrouwbare en repetitieve protocollen vereisen voor hun isolatie. Het realiseren van hun volledige potentieel is echter moeilijk omdat er nog steeds veel technische obstakels zijn die verband houden met hun onderzoek (zoals de juiste acquisitie). Deze studie presenteert een protocol voor de isolatie van kleine EV's (volgens de MISEV 2018-nomenclatuur) uit het kweeksupernatant van tumorcellijnen op basis van differentiële centrifugatie. Het protocol bevat richtlijnen over hoe besmetting met endotoxinen tijdens de isolatie van EV's kan worden voorkomen en hoe deze goed kunnen worden geëvalueerd. Endotoxinebesmetting van EV's kan latere experimenten aanzienlijk belemmeren of zelfs hun ware biologische effecten maskeren. Aan de andere kant kan de over het hoofd geziene aanwezigheid van endotoxinen leiden tot onjuiste conclusies. Dit is van bijzonder belang wanneer wordt verwezen naar cellen van het immuunsysteem, waaronder monocyten, omdat monocyten een populatie vormen die bijzonder gevoelig is voor endotoxineresiduen. Daarom wordt het ten zeerste aanbevolen om EV's te screenen op endotoxinebesmetting, vooral bij het werken met endotoxinegevoelige cellen zoals monocyten, macrofagen, van myeloïden afgeleide suppressorcellen of dendritische cellen.

Een vereiste of verplichte stap voor dit protocol is de uitsluiting van mogelijke endotoxineverontreiniging uit reagentia. Alle reagentia die worden gebruikt, zoals FBS, DMEM, RPMI, PBS en zelfs ultracentrifugebuizen, moeten endotoxinevrij zijn (<0,005 EU / ml). Het handhaven van het regime van geen endotoxinebesmetting is niet eenvoudig, omdat bijvoorbeeld het reguliere/standaardserum voor celkweek de rijke bron kan zijn (0,364 EU/ml; zie tabel 1).

Dit protocol biedt richtlijnen voor het voorkomen van besmetting met endotoxine tijdens de isolatie van extracellulaire blaasjes uit celkweeksupernatanten en hoe deze op de juiste manier kunnen worden geëvalueerd. Het belangrijkste voordeel van dit protocol is dat elk laboratorium dat zich bezighoudt met EV's endotoxinebesmetting kan beperken door een aseptische procedure te volgen en eenvoudige en algemeen beschikbare apparatuur te gebruiken. Iedereen die deze techniek voor de eerste keer probeert, moet beginnen met nieuwe media, reagentia en plasticware die een laag endotoxine, niet-pyrogeen label hebben.Gebruik voor dit protocol endotoxinevrije reagentia, water, kweekmedia, gefilterde PBS, ultra-low-endotoxine FBS en ultracentrifugebuizen. Om te beginnen verzamelt u het supernatant van eerder gekweekte SW480- en SW620-cellijnen in correct gelabelde buizen van 15 milliliter. Verwijder celresten door het supernatant gedurende vijf minuten bij kamertemperatuur op 500 x g te centrifugeren.Zonder het puin te verstoren, verzamelt u het supernatant in een nieuw gelabelde buis en centrifugeert u gedurende 12 minuten bij vier graden Celsius op 3.200 x g. Breng voorzichtig 45 milliliter van het supernatant over in een verticaal geplaatste steriele buis van 50 milliliter, zonder de randen van de buis nat te maken. Wikkel de dop van de buis met een steriele, transparante film en bewaar deze verticaal bij min 80 graden Celsius voor latere isolatie van extracellulaire blaasjes.Begin met isoleren door spuitfilters, spuiten en buizen van 0,22 micrometer te bereiden die ongeveer 90 milliliter supernatant bevatten. Vul een spuit met het supernatant en bevestig het filter op de naaldadapter. Filtreer het supernatant door het filter in een buis van 50 milliliter.Pipetteer zeven milliliter van het filtraat in een voorbereide ultracentrifugebuis en centrifugeer gedurende twee uur bij 100.000 x g bij vier graden Celsius. Gebruik een steriele Pasteurpipet om het supernatant weg te gooien. Trek de pellets in een ultracentrifugebuis met behulp van een lange gefilterde pipetpunt.Spoel de extracellulaire blaasjes door zeven milliliter gefilterd endotoxinevrij PBS toe te voegen. Gooi na ultracentrifugatie het supernatant weg met een steriele Pasteurpipet en resuspensie de pellet in 200 microliter endotoxinevrije PBS. Breng de suspensie over in een steriele 1,5 milliliter eiwitarme reageerbuis.Breng vervolgens 10 microliter suspensie over naar een nieuwe buis van 1,5 milliliter voor verdere analyse. Wikkel de dop van de buis en bewaar deze bij min 80 graden Celsius. Verdun nu in een nieuwe buis een microliter suspensie met behulp van gefilterde PBS in een verhouding van 1 tot 1000 voor analyse van nanodeeltjes.Om een blottinganalyse uit te voeren, mengt u eerst 20 microgram van de monsters met een laadbuffer. Incubeer de monsters bij 70 graden Celsius gedurende 10 minuten. Laad vervolgens 20 microgram van het monster in elke put van een 10 tot 14% polyacrylamidegel met SDS en voer elektroforese uit gedurende 45 minuten op 150 volt, met lopende buffer.Plaats vervolgens de gel in een transfermachine met Towbin-buffer en voer een semi-droge overdracht van eiwitten uit op een polyvinylideendifluoridemembraan bij 25 volt gedurende een uur. Blokkeer het membraan met 1%BSA door het een uur op een rocker te laten broeden. Voeg BSA verdunde antilichamen, anti-CD9 of anti-Alix, toe aan het membraan en incuber 's nachts bij vier graden Celsius op een rocker.Na het verwijderen van de antilichamen, was het membraan driemaal met 10 milliliter TBST gedurende 10 minuten. Voeg nu geit anti-konijn of anti-muis secundair antilichaam toe en incubeer opnieuw op een rocker gedurende een uur bij kamertemperatuur. Gooi het antilichaam weg en was het membraan zoals eerder aangetoond.Giet een milliliter van de oplossing met substraat en luminol in een gelijke verhouding op het membraan. Plaats het membraan onmiddellijk in een beeldvormingssysteem en visualiseer de eiwitbanden op het scherm. In de huidige studie bevestigde western blot-analyse de aanwezigheid van twee extracellulaire blaasjesmarkers, CD9 en Alix.De gemiddelde grootte en concentraties van de blaasjes geïsoleerd uit zowel SW480 als SW620 waren vergelijkbaar. Stimulatie van monocyten met verschillende doses lipopolysaccharide, of LPS, toonde de laagste dosis LPS die de secretie van interleukine 10 mogelijk maakte en de tumornecrosefactor in monocyten was 50 picogram per milliliter. De chromogene LAL-test gaf aan dat LPS-besmetting van de extracellulaire blaasjes ongeveer 50 picogram per milliliter was voor beide cellijnen.Het belangrijkste om te onthouden is het gebruik van LPS-vrije of uitgeputte reagentia en routinematige LPS-meting bij elke stap van de procedure. Het voorkomen van endotoxinebesmetting is gemakkelijker dan het elimineren ervan. Het voorgestelde protocol beperkt de mogelijkheid van valse resultaten als gevolg van de besmetting van EV's met endotoxine en maakt het mogelijk om de EV-activiteit per cel te beoordelen.Het voorgestelde protocol is nuttig voor onderzoekers die werken met endotoxinegevoelige cellen, zoals monocyten, dendritische cellen en macrofagen.
