체외 세포 배양은 생물학의 기초 지식을 확립하는 데 필요하고 매우 귀중한 것으로 입증되었습니다. 특히 생물학과 암에 대한 과학적 이해의 대부분은 단층에서 자라는 세포인 2D 배양 시스템에서 비롯되었습니다. 2D 배양이 지배적인 세포 배양 시스템이었지만 잠재적으로 더 이상의 생물학적 진보를 저해할 수 있는 많은 단점이 있습니다. 예를 들어, 2D 배양은 세포 신호 전달 및 증식에 중요한 세포 간 상호 작용이 부족합니다1. 현재까지 3D 배양 시스템은 분화, 약물 반응, 종양 침습 및 생물학 2,3,4,5를 더 잘 모델링하는 것으로 나타났습니다. 악성 암의 3D 모델링은 고령화 인구와 암 사망률의 증가로 인해 특히 중요합니다. 간세포암(Hepatocellular carcinoma, HCC)은 전 세계적으로 암 관련 사망률의 주요 원인 중 하나이며, 예후가 좋지 않은 경우가 많다6. 간세포암은 치료율이 낮고, 약물 반응이 좋지 않으며, 재발률이 높은 것으로 알려져있다 6,7,8. 정상 간 및 간세포암에 대한 여러 3D 모델이 생체 내 정상 및 악성 간 조직 9,10의 생리학을 모방하여 개발되었습니다.
현재 3D 시스템에는 액체 오버레이, 바이오리액터, 하이드로겔, 스캐폴드 및 3D 프린팅 구조물이 포함됩니다. 생물반응기에서 생성된 스페로이드는 영양소 노출, 가스 교환 및 세포 증식/정지의 종양 분포를 모방하기 때문에 특히 고유한 이점을 제공합니다11. 바이오리액터는 사용 편의성, 큰 확장성, 영양소 확산 및 접근성으로 인해 암 모델에 특히 적합하다11. 또한 바이오리액터는 높은 처리량의 실험, 더 큰 재현성 및 인적 오류 감소를 허용할 수 있습니다. 이 연구에 사용된 바이오리액터는 일반적인 바이오리액터에 적용되는 파괴적 전단력을 최소화하여 더 나은 재현성을 허용하는 감소된 중력 시스템을 시뮬레이션하기 때문에 독특합니다12. 전방위 중력과 전단력의 감소는 세포가 보다 생리적인 방식으로 발달할 수 있도록 합니다. 증거로, 이 방법론에 따라 성장한 HepG2/C3A 세포는 생체 내 ATP, 아데닐레이트 키나아제, 요소 및 콜레스테롤13,14을 생성하는 구형 소기관을 발달시킵니다. 또한 이 3D 시스템의 약물 치료는 2D 배양에 비해 더 발전되고 자동화되어 있습니다. 2D 배양에서 약물 치료는 트립신화하고 세포 건강을 유지해야 하기 때문에 짧은 시간 경과를 거쳐야 하는 경우가 많습니다. 그러나 우리의 경우 세포의 구조와 생리를 방해할 필요 없이 스페로이드의 장기 약물 치료를 수행할 수 있습니다. 따라서 생체 내 생물학적 현상을 더 잘 모델링하고 과학적 발전을 이루기 위해 2D에서 3D 배양으로의 전환이 필요합니다.

semi-automated phenotypic analysis of functional 3d spheroid cell cultures

THLE-3 및 HepG2/C3A 세포주의 3D 배양을 위해 프로토콜을 최적화했습니다. 여기에 설명된 프로토콜은 사용이 간편하며 바이오리액터당 > 100개의 스페로이드를 재현성 있고 비용 효율적으로 배양할 수 있습니다. 생물반응기에 들어가면 스페로이드는 2D 배양에서 유지되는 세포와 유사하게 처리됩니다. 최적의 성장 조건은 일주일에 2-3회 배지를 교환하고(그림 2B) 스페로이드의 성장과 크기에 따라 회전 속도를 조정함으로써 얻을 수 있습니다(그림 2C). 회전하는 바이오리액터에서 3D 스페로이드를 배양하는 이 시스템(그림 2E,F)은 스페로이드를 동일하고 매우 낮은 전단력에 노출시켜 3D 구조를 위한 최적의 성장 환경을 제공합니다.
우리는 이전에 스페로이드가 염색질 변형16의 분석에 어떻게 사용될 수 있는지 보여주었습니다. 여기에서는 간 스페로이드를 얻는 방법과 전체 단백질체 분석을 위해 단백질체학 실험을 수행하는 방법을 자세히 보여줍니다(그림 1). 간단히 말해서, 프로토콜은 배양이 80% 밀도에 도달할 때까지 THLE-3 또는 HepG2/C3A 편평 세포를 사용하여 시작되었습니다. 세포를 스페로이드로 배양하기 위해 약 2,000개의 세포를 마이크로웰이 포함된 초저 부착판에 도금하여 자가 응집할 수 있도록 한 다음 형성된 스페로이드를 바이오리액터로 옮겼습니다(그림 1A). 스페로이드는 배양 후 3주 후에 기능적으로 활성화되지만, 앞서17번 설명한 바와 같이 이 프로토콜에 대해 배양 36일 후 수집된 스페로이드의 결과를 보여줍니다. 채취 후, 스페로이드를 회전시키고, 분석을 위해 펠릿과 상층액을 모두 저장하였다. 세포 생존율은 앞서 설명한 바와 같이, 손상된 세포에 의해 방출되는 아데닐레이트 키나아제의 정량화를 위해 상층액으로부터 평가하였다17. 세포 펠릿에서 세포 단백질을 추출하고, 전체 단백질체를 고분해능 질량분석법으로 분석했습니다(그림 1B).
이 프로토콜은 또한 공개 이미지 처리 프로그램인 FIJI(Fiji Is Just ImageJ)18를 사용하여 스페로이드 계수를 위한 반자동 방법(그림 3A)을 보여줍니다. 분석을 위해 스페로이드의 좋은 품질의 사진을 찍어야 하며 섹션 5에서 언급한 대로 일부 매개변수를 고려해야 합니다. 그런 다음 분석을 위해 그림을 준비한 후 매크로 스크립트(보충 파일 1)를 사용하여 스페로이드를 계산합니다. 매크로는 먼저 스페로이드 그림이 있는 폴더 안에 FIJI Spheroids counting이라는 폴더를 만들어 작동합니다. 이 폴더에는 분석의 모든 정보가 저장됩니다. 여기에는 각 스페로이드의 ID 번호와 함께 계산된 스페로이드의 사진이 포함됩니다. 여기에는 스페로이드 카운팅이라는 Excel 파일도 포함되어 있습니다. 이 파일에는 계산된 각 회전 타원체의 픽셀 영역과 ID 번호가 포함되어 있습니다. 분석된 사진 하나에 해당하는 데이터가 파일의 각 탭에 표시됩니다. 탭은 분석된 그림의 이름에 따라 레이블이 지정됩니다. 스페로이드 크기는 혈관 내 구조물의 수와 약물 처리를 포함한 여러 요인에 의해 손상될 수 있으므로 표면적을 모니터링하는 것도 중요합니다(planimetry). 여기에 제시된 매크로 스크립트(보충 파일 2)는 그림의 스페로이드에 해당하는 검은색 영역을 측정하여 작동합니다(그림 3B). 출력은 각 회전 타원체의 측정된 면적, 둘레 및 직경을 포함하는 planimetry.xlsx 파일에 수집됩니다. 직경을 계산하는 데 사용되는 Feret이라는 측정도 있습니다. Feret은 가능한 가장 긴 직경이고 minFeret은 가장 짧은 직경입니다. 직경은 이 둘의 평균입니다. 출력 폴더 안에는 planimetry.xlsx 파일 외에도 측정된 스페로이드의 그림도 있습니다.
단백질체 분석을 진행하기 전에 배양 시간 동안 스페로이드의 생존력을 평가했습니다. AK의 수준은 17일째까지 증가하여 세포 사멸의 약 7%에 도달한 다음 사멸은 5% 미만 수준으로 감소하며(그림 4A), 이는 이전에 발표된 연구17에 따릅니다. 이 프로토콜은 또한 세포 표현형을 모니터링하기 위한 전체 단백질체 분석을 보여줍니다. 먼저, THLE-3 및 HepG2/C3A 편평 세포와 스페로이드의 단백질체를 비교했습니다. 첫 번째 주성분(PC1)을 분석하면 편평 세포 배양에서 스페로이드 샘플이 엄격하게 분리되어 있음이 분명하며 세포 유형(THLE-3 및 HepG2/C3A)의 상관관계는 관련이 없는 것으로 보입니다(그림 4B). THLE-3 및 HepG2/C3A 스페로이드는 함께 뭉치지 않지만 간 기능과 일치하는 유사한 프로필을 공유합니다. 우리는 이 프로토콜에서 간에서 수행되는 금속 해독에 중요한 역할을 하는 메탈로티오네인의 예를 보여줍니다. 단백질체학 분석에서 편평 세포(MT1E 및 MT1X)와 비교하여 스페로이드에서 과발현되는 2개의 동형을 확인했습니다(그림 4C). 또한 스페로이드로 성장한 두 세포주의 유전자 온톨로지(GO) 농축을 보여줍니다. 트리카르복실산 회로(TCA 회로), 전자 전달 사슬(세포 호흡) 및 피루브산 대사로 구성된 탄수화물 대사 과정은 자주 사용되는 용어이며 HepG2/C3A 및 THLE-3 스페로이드가 풍부합니다(그림 4D,E). 세포 해독, 지방산 및 콜레스테롤 대사는 두 스페로이드에 풍부하게 함유된 다른 기능입니다. 이러한 기능은 간 기능에 매우 중요한 것으로 알려져 있습니다.
