초파리 멜라노가스터(Drosophila melanogaster)는 유전자 변형(gene modification)1, 약물 치료(drug treatment)2, 노화(sagingence)3에 의해 생성된 뉴런 질환 모델에서 세포 및 분자 기능을 조사하기 위한 우수한 모델 유기체를 제공한다. 인간과 초파리 사이의 생물학적 경로, 물리적 특성 및 질병 관련 상동체 유전자의 높은 보존은 초파리를 분자에서 행동 수준까지 이상적인 모방체로 만듭니다4. 많은 질병 모델에서, 행동 결핍은 다양한 인간 신경병증에 대한 유용한 모델을 제공하는 중요한 지표이다 5,6. 초파리는 현재 파킨슨병 및 근위축성 측삭 경화증과 같은 여러 인간 질병, 신경 발달 및 신경퇴행성 질환을 연구하는 데 사용됩니다 7,8. 질병 모델의 운동 능력을 감지하는 것은 병원성 진행을 이해하는 데 중요하며 질병 과정의 기초가 되는 분자 메커니즘에 대한 표현형 상관관계를 제공할 수 있습니다.

FlyGrAM11 및 Drosophila island assay10과 같은 성체 초파리를 테스트하기 위한 동물 추적 그룹 기반 전략은 사전 정의된 영역에서 사회적 모집 및 개별 추적을 구현합니다. 그럼에도 불구하고, 반항적인 지역에서의 사회적 개별적 제한은 파리의 충돌 또는 겹침으로 인해 이미지의 식별에 부정적인 영향을 미칠 수 있습니다. 현재 개발된 방법은 피지(ImageJ) 영상 처리 프로그램과 호환되는 AnimalTracker 응용 프로그래밍 인터페이스(API)를 활용하여 고화질(HD) 비디오에서 추적 행동을 분석하여 성충 파리와 유충 파리의 운동 활동을 체계적으로 평가합니다. Fiji는 강력한 소프트웨어 라이브러리와 수많은 스크립팅 언어를 결합할 수 있는 오픈 소스 소프트웨어 ImageJ 배포판으로, 이미지 처리 알고리즘의 신속한 프로토타이핑을 가능하게 하여 이미지 분석 기능으로 생물학자들 사이에서 인기가 있습니다(20). 현재 접근 방식에서 피지의 AnimalTracker API에 대한 통합은 개인화된 알고리즘 삽입을 통해 고유한 초파리 행동 분석을 개발하는 데 활용되며, 운동 행동의 강력한 분석 기능을 지원하기 위한 자세한 문서 및 자습서를 위한 유용한 단계를 제공합니다(그림 1). 파리의 충돌 또는 겹침으로 인한 이미지에서 객관적인 식별의 복잡성을 피하기 위해 각 경기장은 단 하나의 파리만 호스팅하도록 제한됩니다. 접근법의 추적 정밀도를 평가한 결과, 파킨슨병의 동물 모델에 일반적으로 사용되는 독성 약물 로테논과 함께 투여된 초파리 의 운동 운동을 추적하고 정량화하여 궁극적으로 약물 치료에서 운동 장애를 발견하기 위해 구현되었습니다21. 오픈 소스 및 자유 소프트웨어를 사용하는 이 방법론은 고비용의 계측을 필요로 하지 않으며 초파리 행동 운동을 정확하고 재현 가능하게 분석할 수 있습니다.
