ヒト脂肪由来幹細胞(hASC)を単離する従来の方法では、コラゲナーゼを用いた酵素消化とそれに続く遠心分離による洗浄ステップが行われます4。酵素による単離は、一般に、細胞収量と生存率の点で他の機械的技術よりも効率的であると考えられていますが、コラゲナーゼなど、使用される動物由来の成分は、米国食品医薬品局によって最小限に操作されていると考えられています。これは、免疫反応または疾患伝播のリスクが大幅に増加することを意味し、したがって、hASC療法の臨床現場への翻訳を制限します5,6。
トリプシンベースの消化は、ASCを単離するための別の酵素プロトコルです。トリプシン濃度、遠心分離速度、およびインキュベーション時間の点でわずかな変更を加えたさまざまな技術が説明されています。残念なことに、この方法は十分に説明されておらず、文献、特に機械的絶縁プロトコル7との比較の欠如が存在する。しかしながら、アプローチの翻訳可能性に関して、トリプシンはコラゲナーゼと同じ欠点を有する。
機械的な力に基づいて酵素を添加しないASCを得るための代替単離方法には、脂肪組織断片の高速遠心分離(800 x gまたは1,280 x g、15分)が含まれます。次いで、ペレットを赤血球溶解緩衝液(5分間)と共にインキュベートし、続いて培養培地に再懸濁する前に600 x gで別の遠心分離工程を行う。外植片法と比較して最初の数日間に単離された細胞の数が多いにもかかわらず、以前の研究では、培養2週目以降の増殖が少ないか、または存在しないことが示されました8。
それに加えて、細胞培養のための成長因子サプリメントとして使用されるウシ胎児血清(FBS)などの異種添加培地によるさらなる操作は、免疫反応性のリスクおよび宿主患者のウイルス、細菌、またはプリオン感染への曝露の増加と関連している9,10。免疫反応および蕁麻疹様発疹の発生は、FBS11で産生された間葉系幹細胞を数回投与された個体において既に説明されている。さらに、FBSはバッチ間のばらつきを受け、最終製品の品質に影響を与える可能性があります12。
適正製造基準(GMP)ガイドラインに従って、酵素組織の解離を回避し、FBSを異種を含まないサプリメントに置き換える必要があります。これらのステップは、より信頼性が高く再現性のあるプロトコルとともに、細胞療法の適用をサポートするために不可欠です3、13。
これに関連して、ヒト血小板ライセート(hPL)は、無細胞、タンパク質含有、成長因子が豊富なサプリメントであり、in vitro細胞培養および増殖のための増殖培地の添加剤として臨床グレードの細胞ベースの製品の間で以前に導入されたため、FBSの代替として提案されています14,15。hPLはヒト由来の産物であるため、臨床応用を目的としたhASCのin vitro培養中にFBSの代替として頻繁に使用され、FBSの翻訳可能性に関連する免疫反応や感染症に関する問題を軽減します15,16。
より高い生産コストにもかかわらず、FBSと比較して、hPLは多くの細胞タイプの細胞生存率をサポートし、増殖を増加させ、老化を遅らせ、ゲノム安定性を保証し、後期細胞継代でも細胞免疫表現型を保存することがすでに実証されています。これらすべての要素は、この文化サプリメント11への切り替えをサポートします。
この研究の目的は、従来のFBS培養hASCと比較して、細胞生理学とステムネスの特性を変更することなく、完全な動物を含まない方法でhASCを分離および培養するための標準化されたプロトコルを開発することでした(図1)。
