伝統的な漢方薬(TCM)の製造において、TCM抽出中間体および関連する液体製剤の安定性は常に検査の焦点でした1。医薬品、特にポリフェノールを主要な有効成分とする医薬品の臨床効果は、重大な安定性の問題のために損なわれています2,3。サナジョン内服液とヌオディカン内服液は、この問題4の典型的な例です。したがって、効率的なツールを使用して、TCM製造プロセスにおける液体中間体の安定性を迅速かつ正確に評価および最適化する方法を学ぶことが重要です。東南アジアで広く普及している薬用植物であるフィランサスエンブリカL.(PE)は、優れた抗酸化作用5、抗炎症作用6、抗菌作用7、抗腫瘍作用8を持っていると考えられています。熱抽出手順中に、PEのタンニンは激しく変換されます9。高温の触媒作用下では、これらのタンニンは急速に加水分解して没食子酸やエラグ酸などの分子を生成し、溶解性が低いために不安定性または沈殿を引き起こします1。加速試験や遠心分離など、TCMの安定性を評価するための現在の方法は、通常、面倒であり4、関連する調製プロセスのさらなる開発を制限しています。
多重光散乱(MLS)の原理に基づき、PEF抽出物の高速安定性評価法を確立し、不安定性メカニズムを解析した。MLSは、近赤外光源のスキャンに基づく測定方法です。ソリューションシステムを変更すると、光の強度が変化します。入射光は、サンプルの粒子に吸収または透過されると散乱されます。システムは、サンプルを通過するときに透過光信号を記録します。サンプルの光透過率が低い場合、システムは後方散乱光信号を記録します。目視観察と比較して、これは多くの時間を節約でき 1、不安定現象を迅速かつ正確に詳細に分析できるため、抽出プロセスの最適化を導くためのより有用な情報を提供します。

伝統的な漢方薬の抽出中間体は、準備プロセスの重要な中間体であり、その安定性は最終製品の有効性と品質に重要な影響を及ぼします。しかし、既存の安定性評価法は、長時間の観察や複雑な装置(高速液体クロマトグラフィーなど)の操作が必要で、時間のかかることが多く、システムの不安定性に関するより多くの物理的情報を得ることは困難である。したがって、伝統的な中国医学のための迅速かつ正確な安定性分析技術を確立することが急務です。多重光散乱は、サンプルの性質や状態を変えたり、有機試薬を使用したりすることなく、環境にやさしい方法で伝統的な漢方薬の安定性を正確かつ迅速に評価できる最先端の分析方法です。

安定性評価方法は、多くの場合、時間と労力がかかります。この手法により、約200倍の時間を節約し、フィランサスエンブリカL.抽出物の多くの安定性パラメータを収集できます。この手法は、不安定現象を迅速かつ正確に詳細に分析できるため、抽出プロセスの楽観主義を導くためのより有用な情報を提供します。まず、適量のフィランサスエンブリカL.を正確に計量し、逆流抽出のために植物の重量の10倍の脱イオン水を追加します。秤量後、還流抽出用に5つのサンプルをセットし、0時間でE1、0.5時間でE2、1時間でE3、1.5時間でE4、2時間でE5をセットしました。ピペットを使用して20ミリリットルのサンプル溶液をサンプルボトルに追加し、毎回追加する溶液が同じ高さになるようにします。複数の光散乱(MLS)検出装置の電源を入れ、30分間ウォームアップします。トップメニューの[ファイルの作成]ボタンをクリックして新しいテストファイルを作成し、[タービスカンラボ温度の表示]ボタンをクリックして、機器の目標温度を摂氏25度に設定します。[プログラムスキャン]をクリックしてセットアップ分析プログラムに入り、プログラムをリストに追加します。バランス時間を20分に設定します。タスク バーで、分析シーケンスに [48 時間のスキャン] を追加し、サイクルとして 5 分間のスキャンを追加します。後続のすべての測定に対してこの分析プログラムを選択します。準備したサンプルボトルをMLS検出システムに入れ、[開始]をクリックして測定を開始します。データ収集後、計算パラメータリストをクリックします。分散相屈折率を1.36、連続相屈折率を1.33に設定して安定性指数、粒径、粒子移動速度を計算し、体積分率を1に設定し、E1〜E5サンプルの連続位相光透過強度を99.99%MLSスペクトルにこの図に示します。スペクトルデータは、E2サンプルの変動が少なく、サンプルの安定性が高いことを示していますが、E1は透過光の全体的な減少により濁りがあった可能性があります。E3からE5のサンプルは非常に不安定であり、異なる高さのサンプルのスペクトルデータは異なり、層別化が後期に発生したことを示しています。T値は時間とともに増加し、サンプルをより不安定にします。E3とE4では、デルタTレベルは最終的に前の段階に戻り、これらの抽出物で凝集と沈殿が起こったことを示しています。E5のデルタTは濁り後も低いままであり、E5が大量の沈降を起こした可能性があることが示唆された。光子自由行程の傾向は、試料の透過光の変化を反映することができる。さまざまな抽出物の安定性は、時間の経過とともに継続的に変動しました。粒子サイズの動的変化により、すべてのサンプルの粒子サイズが8〜20時間以内に大幅に変化し、E3とE5の粒子サイズが測定範囲を超えていることが明らかになりました。異なる抽出方法で得られたフィランサス・エンブリカL.抽出物の不安定性をここに示す。各結果の上部にある色度バンドは、異なる色に対応する光強度値を表し、青色の部分は透過を表し、茶色の部分は後方散乱強度を表します。測定に適した分析プログラムを選択し、安定性指数、粒子サイズ、粒子移動速度を計算するためのパラメータリストを選択することは、これらの2つのステップが結果の最新性に直接影響するため、この粒子にとって非常に重要です。層の厚さなどの重要な情報も取得でき、乳化または沈殿の形成規則を分析できます。この技術は、広範な投薬不安定性データに基づく安定性予測モデルの開発を容易にします。さらに、この手法を他の検出方法と組み合わせて強化し、研究の可能性をさらに広げることができます。
