TBC er en viktig tibetansk medisin med effekten av å fjerne kulde og lindre smerte. Det har vært mest brukt til å behandle traumatisk skade og revmatisk artralgi i Kina i tusenvis av år24,25,26. Diterpenoid alkaloider er både aktive og giftige ingredienser av TBC27,28,29. De viktigste toksiske effektene av aconitumalkaloider av TBC...

Den tørkede roten av Aconitum pendel Busch og A. flavum Hand.-Mazz., en av de mest kjente tibetanske medisinene, kalles Tiebangchui (TBC) på kinesisk 1,2. De tørkede røttene til TBC er nyttige for å fjerne kulde og vind, redusere smerte og beroligende sjokk. Det ble registrert i første volum av "Drug Standards (tibetansk medisin) av Helsedepartementet i Folkerepublikken Kina", som sier at de tørkede røttene til TBC ofte brukes til å behandle revmatoid artritt, blåmerker og andre forkjølelsessykdommer3. Imidlertid er den kliniske terapeutiske dosen av TBC lik den toksiske dosen, og tilfeller av forgiftning eller død har blitt hyppig rapportert på grunn av feil bruk4. Derfor har reduksjon av toksisiteten og bevaring av effekten av TBC blitt et forskningspunkt gjennom årene.
I tibetansk medisin er behandling en av de mest effektive metodene for å dempe toksisiteten av TBC. I denne artikkelen ble et monoester-diterpenoid alkaloid (MDA) (benzoylakonitin) og to diester-diterpenoidalkaloider (DDA) (akonitin, 3-deoksyakonitin) brukt som evalueringsindekser. Den tørkede roten av Aconitum pendel Busch., kalt Tiebangchui (TBC) på kinesisk, er en av de mest berømte tibetanske medisinene. Det er en mye brukt urt i Nordvest-Kina. Imidlertid har mange tilfeller av forgiftning oppstått på grunn av TBCs intense toksisitet og fordi dets terapeutiske og giftige doser er like. Derfor er det en presserende oppgave å finne en sikker og effektiv metode for å redusere toksisiteten. Et søk gjennom de tibetanske medisinklassikerne viser at behandlingsmetoden for TBC-stekt med Zanba ble registrert i "Processing specification of Tibetan medicine of Qinghai Province (2010)". Imidlertid er de spesifikke behandlingsparametrene ennå ikke klare. Dermed har denne studien som mål å optimalisere og standardisere prosesseringsteknologien til Zanba-stir-fried TBC.
Først ble det utført et enkeltfaktoreksperiment på fire faktorer: skivetykkelsen på TBC, mengden Zanba, prosesseringstemperatur og tid. Med monoester og diesteralkaloidinnhold i Zanba-stir-fried TBC som indekser, ble CRITIC kombinert med Box-Behnken-responsoverflatemetoden brukt til å optimalisere prosesseringsteknologien til Zanba-stir-fried TBC. De optimaliserte behandlingsforholdene for Zanba-wokede TBC var en TBC-skivetykkelse på 2 cm, tre ganger mer Zanba enn TBC, en behandlingstemperatur på 125 °C og 60 minutters steking. Denne studien bestemte de optimaliserte og standard behandlingsbetingelsene for bruk av Zanba-stir-fried TBC, og ga dermed et eksperimentelt grunnlag for sikker klinisk bruk og industriell produksjon av Zanba-stir-fried TBC.

Omfanget av denne protokollen ligger i fortsettelsen av de tibetanske medisinene. Dette personalet har som mål å optimalisere og standardisere prosesseringsteknologien til Zanba-stir-fried Tiebangchui. Vårt laboratorium vil være mer oppmerksom på cytokjemisk undersøkelse for å oppnå målrettet mengdekontroll.
