IsdB, een bacterieel celwandverankerd en ijzergereguleerd oppervlakte-eiwit, speelt een belangrijke rol in het proces van het verkrijgen van heemijzer15. Methicilline-resistente Staphylococcus aureus (MRSA) is een opportunistische ziekteverwekker met een van de hoogste infectiepercentages op een intensive care unit (ICU) afdeling1, cardiologieafdelingen en brandwondenafdelingen wereldwijd. MRSA vertoont hoge infectiepercentages, mortaliteit en brede resistentie tegen geneesmiddelen, wat grote problemen oplevert bij klinische behandeling2. In de wereldwijde prioriteitenlijst van antibioticaresistente bacteriën die in 2017 door de Wereldgezondheidsorganisatie (WHO) werd vrijgegeven, werd MRSA opgenomen in de meest kritieke categorie3. Een vaccin tegen MRSA-infectie is daarom dringend nodig.
Aluminiumadjuvans wordt al lange tijd gebruikt en het hulpmechanisme van het adjuvans is relatief duidelijk, veilig, effectief en goed verdragen4. Aluminium adjuvantia zijn momenteel een veel gebruikt type adjuvans. Over het algemeen wordt aangenomen dat antigenen geadsorbeerd aan aluminiumzoutdeeltjes de stabiliteit kunnen verbeteren en het vermogen van de injectieplaats om antigenen op te nemen kunnen verbeteren, wat zorgt voor een goede absorptie en langzame afgifte5. Momenteel is het grootste nadeel van aluminiumadjuvantia dat ze geen adjuvans effect hebben of slechts een zwak adjuvans effect vertonen op sommige kandidaat-vaccinantigenen6. Bovendien induceren aluminiumadjuvantia IgE-gemedieerde overgevoeligheidsreacties5. Daarom is het noodzakelijk om nieuwe adjuvantia te ontwikkelen om een sterkere immuunrespons te stimuleren.
Nano-emulsie-adjuvantia zijn colloïdale dispersiesystemen die bestaan uit olie, water, oppervlakteactieve stoffen en cosurfactanten7. Bovendien zijn de adjuvantia thermodynamisch stabiel en isotroop, kunnen ze worden geautoclaveerd of gestabiliseerd door centrifugatie met hoge snelheid en kunnen ze spontaan worden gevormd onder milde bereidingsomstandigheden. Verschillende emulsie-adjuvantia (zoals MF59, NB001-002-serie, AS01-04-serie, enz.) zijn momenteel op de markt of in de klinische onderzoeksfase, maar hun deeltjesgroottes zijn groter dan 160 nm8. Daarom kunnen de voordelen van medicinale preparaten op nanoschaal (1-100 nm) (d.w.z. groot specifiek oppervlak, kleine deeltjesgrootte, oppervlakte-effect, hoge oppervlakte-energie, klein effect en macro-kwantumtunneleffect) niet volledig worden benut. In het huidige protocol is gemeld dat een nieuw adjuvans op basis van nano-emulsietechnologie met een diametergrootte van 1-100 nm een goede adjuvante activiteit vertoont9. We testten het recombinatiesubunitvaccinantigeeneiwit HI (α-hemolysinemutant [Hla] en Fe-ionenoppervlakbepalende factor B [IsdB] subunit N2 actief fragmentfusie-eiwit); Een reeks procedures werd vastgesteld om de fysieke eigenschappen en stabiliteit te onderzoeken, de specifieke antilichaamrespons na intramusculaire toediening te evalueren en het beschermende effect van het vaccin te testen met behulp van een systemisch infectiemodel voor muizen.

evaluating the immune response of a nanoemulsion adjuvant vaccine against methicillin-resistant staphylococcus aureus (mrsa) infection

Nano-emulsie-adjuvante vaccins hebben uitgebreide aandacht getrokken vanwege hun kleine deeltjesgrootte, hoge thermische stabiliteit en het vermogen om geldig immuunresponsen te induceren. Het vaststellen van een reeks uitgebreide protocollen om de immuunrespons van een nieuw nano-emulsie-adjuvansvaccin te evalueren, is echter van vitaal belang. Daarom bevat dit artikel een rigoureuze procedure om de fysisch-chemische kenmerken van een vaccin te bepalen (door transmissie-elektronenmicroscopie [TEM], atoomkrachtmicroscopie [AFM] en dynamische lichtverstrooiing [DLS]), de stabiliteit van het vaccinantigeen en -systeem (door een hogesnelheidscentrifugetest, een thermodynamische stabiliteitstest, SDS-PAGE en western blot) en de specifieke immuunrespons (IgG1, IgG2a en IgG2b). Met behulp van deze aanpak kunnen onderzoekers het beschermende effect van een nieuw nano-emulsieadjuvansvaccin in een dodelijk MRSA252 muismodel nauwkeurig evalueren. Met deze protocollen kan het meest veelbelovende nano-emulsievaccinadjuvans in termen van effectief adjuvanspotentieel worden geïdentificeerd. Bovendien kunnen de methoden helpen bij het optimaliseren van nieuwe vaccins voor toekomstige ontwikkeling.

Het protocol voor de bereiding van het nano-emulsieadjuvansvaccin en in vitro en in vivo tests van dit vaccin werd geëvalueerd. Evaluating the Immune Response of a Nanoemulsion Adjuvant Vaccine Against Methicillin-Resistant Staphylococcus aureus MRSA Infection

Het gedemonstreerde protocol onderstreepte de directe en effectieve experimentele aanpak en de gedetailleerde stappen naar immuunresponseffecten van nieuwe virale adjuvantia. Dit protocol biedt het onderzoek niet alleen een in principe immuunevaluatiemethode voor adjuvante vaccins, maar kan ook hun massastabiliteit bepalen. Neem om te beginnen de 10 milligram per milliliter werkoplossing van het MRSA HI-antigeeneiwit en voeg er achtereenvolgens de drie hulpstoffen aan toe, isopropylmyristaat, polyoxyethyleenricinusolie en propyleenglycol, met behoud van een massaverhouding van één is tot zes is tot drie.Roer het mengsel goed en voeg vervolgens geleidelijk gesteriliseerd zuiver water toe om een systeemvolume van 10 milliliter te bereiken. Om het nano-emulsievaccin te bereiden met behulp van een emulgeringsmethode met lage energie, roert u het gedurende ongeveer 20 minuten met 500 rotaties per minuut en 25 graden Celsius. Vervolgens wordt het nano-emulsievaccin met één milligram per milliliter eiwit verkregen als een heldere transparante vloeistof.Voor antigeencoating van de enzymgebonden immunosorbenttest of ELISA-plaat, verdun HI-eiwitantigeen tot 10 microgram per milliliter met coatingoplossing en voeg 100 microliter van het verdunde antigeen per put toe aan de ELISA-plaat. Incubeer de plaat 's nachts bij vier graden Celsius. Was na incubatie de plaat met 300 microliter PBST per put.Sluit de putten af met 300 microliter van de afdichtingsoplossing per put gedurende twee uur bij 37 graden Celsius, verdun vervolgens drie microliter van het muizenserum van elke experimentele groep 100 keer door 297 microliter PBST toe te voegen en vortex het verdunde serum. Voeg 100 microliter verdund serum per put toe aan de gecoate ELISA-plaat voor elk serummonster. Verdun op dezelfde manier de positieve en negatieve controleserums 100 keer met PBST door vier microliter positief of negatief controleserum toe te voegen aan 396 microliter PBST, gevolgd door vortexmenging.Om verdunning met dubbele verhouding uit te voeren, voegt u 200 microliter van het eerder verdunde experimentele serum toe aan de eerste rij van de gecoate ELISA-plaat en voegt u vervolgens 100 microliter PBST toe aan alle putten, behalve die in rij één en 12. Stel vervolgens het pipetpistool in op 100 microliter en breng 100 microliter serum over van de eerste rij naar de tweede rij met PBST. Meng de inhoud in de tweede rij 10 keer met behulp van het pipetpistool.Op dezelfde manier voert u één is uit tot twee opeenvolgende verdunningen van het serum tot rij 11. Als u klaar bent, gooit u 100 microliter vloeistof uit rij 11 weg. Voeg 100 microliter verdund negatief en positief serum toe aan elke overeenkomstige put in rij 12 volgens de monstersjabloon.Sluit de ELISA-plaat af met plasticfolie en incubeer gedurende een uur bij 37 graden Celsius. Verdun ondertussen secundaire antilichaam anti-muis IgG HRP 10.000 keer door PBST toe te voegen. Na de incubatie van een uur wast u de ELISA-plaat met 300 microliter PBST per put om de ongebonden antilichamen te reinigen, voegt u vervolgens 100 microliter van het verdunde secundaire antilichaam toe aan elke put en incubeert u de plaat gedurende 40 minuten bij 37 graden Celsius.Was de plaat met 300 microliter PBST per put en voeg vervolgens 100 microliter TMB-kleurontwikkelingsoplossing toe aan elke put. Plaats de platen op 37 graden Celsius en uit de buurt van licht gedurende 10 minuten. Beëindig daarna de reactie onmiddellijk met 50 microliter van twee molaire zwavelzuur.Detecteer de optische dichtheid of OD-waarden op 415 nanometer met behulp van een enzymmarker binnen 15 minuten na beëindiging. De fysische kenmerken van het nieuwe nano-emulsievaccin werden geanalyseerd met behulp van transmissie-elektronenmicroscopie en atoomkrachtmicroscopie. Dynamische lichtverstrooiing werd gebruikt om de deeltjesgrootteverdeling en de zetapotentiaal van het monster te bepalen.SDS-PAGE en western blotting toonden aan dat de hoeveelheid antigeen in het neerslag en supernatant niet significant uitstelde na centrifugatie, wat aangeeft dat het vaccin structureel intact, specifiek en immunogeen was. Het nano-emulsieadjuvansvaccin verhoogde de totale IgG-, IgG1- en IgG2a-antilichaamtiters van de muizen aanzienlijk. De immunogeniciteit van het eiwitvaccin was significant verbeterd.De serum IgG2b OD-waarden bij 450 nanometer waren het hoogst wanneer PBS werd gebruikt bij een verdunning van één tot 5.000. Het dodelijke model van MRSA 252-muizen weerspiegelde dat het nano-emulsievaccin een goed beschermend effect vertoonde en bacteriële infectie met MRS A 252 effectief remde, waardoor de overlevingskans van geïnfecteerde muizen verbeterde. De bereiding van nano-emulsie en de toevoeging van serum aan de plant zijn de belangrijkste.Ook bij detectie van de antilichaamtiter moet men letten op het corrigeren van een bredere en op vloeistof gebaseerde uit. Naast adjuvantia kan deze methode ook worden gebruikt voor de immuunevolutie van andere wiskundige arrays, zoals kunststoffen of toedieningssystemen.
