Methicillin-resistenter Staphylococcus aureus (MRSA) ist ein opportunistischer Erreger mit einer der höchsten Infektionsraten auf einer Intensivstation (ICU) Station1, kardiologischen Abteilungen und Verbrennungsabteilungen weltweit. MRSA weist hohe Infektionsraten, Mortalität und breite Arzneimittelresistenzen auf, was eine große klinische Behandlung erschwert2. In der 2017 von der Weltgesundheitsorganisation (WHO) veröffentlichten globalen Prioritätenliste antibiotikaresistenter Bakterien wurde MRSA in die kritischste Kategorie3 eingestuft. Ein Impfstoff gegen eine MRSA-Infektion wird daher dringend benötigt.
Aluminium-Adjuvans wird seit langem verwendet, und der Hilfsmechanismus des Adjuvans ist relativ klar, sicher, wirksam und gut verträglich4. Aluminium-Adjuvantien sind derzeit eine weit verbreitete Art von Adjuvans. Es wird allgemein angenommen, dass Antigene, die an Aluminiumsalzpartikeln adsorbiert werden, die Stabilität und die Fähigkeit der Injektionsstelle, Antigene aufzunehmen, verbessern können, was zu einer guten Absorption und langsamen Freisetzung führt5. Derzeit besteht der Hauptnachteil von Aluminium-Adjuvantien darin, dass ihnen eine adjuvante Wirkung fehlt oder sie nur eine schwache adjuvante Wirkung auf einige Antigene von Impfstoffkandidaten aufweisen6. Darüber hinaus induzieren Aluminium-Adjuvantien IgE-vermittelte Überempfindlichkeitsreaktionen5. Daher ist es notwendig, neuartige Adjuvantien zu entwickeln, um eine stärkere Immunantwort zu stimulieren.
Nanoemulsions-Adjuvantien sind kolloidale Dispersionssysteme, die aus Öl, Wasser, Tensiden und Cotensiden bestehen7. Darüber hinaus sind die Adjuvantien thermodynamisch stabil und isotrop, können autoklaviert oder durch Hochgeschwindigkeitszentrifugation stabilisiert werden und können unter milden Präparationsbedingungen spontan gebildet werden. Mehrere Emulsionshilfsstoffe (z. B. MF59, NB001-002-Serie, AS01-04-Serie usw.) sind derzeit auf dem Markt oder befinden sich in der klinischen Forschung, aber ihre Partikelgrößen sind größer als 160 nm8. Daher können die Vorteile nanoskaliger (1-100 nm) medizinischer Zubereitungen (d. h. große spezifische Oberfläche, kleine Partikelgröße, Oberflächeneffekt, hohe Oberflächenenergie, kleiner Größeneffekt und Makro-Quantentunneleffekt) nicht voll ausgeschöpft werden. Im vorliegenden Protokoll wurde berichtet, dass ein neuartiges Adjuvans auf Basis der Nanoemulsionstechnologie mit einem Durchmesser von 1-100 nm eine gute Adjuvansaktivität aufweist9. Wir testeten die Rekombinationsuntereinheit Impfstoff-Antigen-Protein HI (α-Hämolysin-Mutante [Hla] und Fe-Ionen-Oberflächen-bestimmender Faktor B [IsdB] Untereinheit N2 aktives Fragment-Fusionsprotein); Es wurde eine Reihe von Verfahren etabliert, um die physikalischen Eigenschaften und die Stabilität zu untersuchen, die spezifische Antikörperantwort nach intramuskulärer Verabreichung zu bewerten und die Schutzwirkung des Impfstoffs anhand eines systemischen Infektionsmodells der Maus zu testen.

Nanoemulsions-Adjuvans-Impfstoffe haben aufgrund ihrer geringen Partikelgröße, ihrer hohen thermischen Stabilität und ihrer Fähigkeit, gültige Immunreaktionen zu induzieren, große Aufmerksamkeit erregt. Die Etablierung einer Reihe umfassender Protokolle zur Bewertung der Immunantwort eines neuartigen Nanoemulsions-Adjuvans-Impfstoffs ist jedoch von entscheidender Bedeutung. Daher wird in diesem Artikel ein strenges Verfahren zur Bestimmung der physikalisch-chemischen Eigenschaften eines Impfstoffs (mittels Transmissionselektronenmikroskopie [TEM], Rasterkraftmikroskopie [AFM] und dynamischer Lichtstreuung [DLS]), der Stabilität des Impfstoffantigens und -systems (durch einen Hochgeschwindigkeitszentrifugentest, einem thermodynamischen Stabilitätstest, SDS-PAGE und Western Blot) und der spezifischen Immunantwort (IgG1, IgG2a und IgG2b). Mit diesem Ansatz können Forscher die Schutzwirkung eines neuartigen Nanoemulsions-Adjuvans-Impfstoffs in einem tödlichen MRSA252-Mausmodell genau bewerten. Mit diesen Protokollen kann das vielversprechendste Nanoemulsions-Impfstoff-Adjuvans in Bezug auf das effektive Adjuvanspotenzial identifiziert werden. Darüber hinaus können die Methoden dazu beitragen, neuartige Impfstoffe für die zukünftige Entwicklung zu optimieren.

Das demonstrierte Protokoll unterstrich den direkten und effektiven experimentellen Ansatz und die detaillierten Schritte zur Immunantwort neuartiger viraler Adjuvantien. Dieses Protokoll bietet der Forschung nicht nur eine grundsätzlich immunologische Bewertungsmethode für adjuvante Impfstoffe, sondern kann auch deren Massenstabilität bestimmen. Nehmen Sie zunächst die 10 Milligramm pro Milliliter Arbeitslösung des MRSA-HI-Antigenproteins und fügen Sie nacheinander die drei Hilfsstoffe Isopropylmyristat, Polyoxyethylen-Rizinusöl und Propylenglykol hinzu, wobei Sie ein Massenverhältnis von eins zu sechs zu drei beibehalten.Rühren Sie die Mischung gut um und fügen Sie dann nach und nach sterilisiertes reines Wasser hinzu, um ein Systemvolumen von 10 Millilitern zu erreichen. Um den Nanoemulsionsimpfstoff mit der Niedrigenergie-Emulgiermethode herzustellen, rühren Sie ihn etwa 20 Minuten lang bei 500 Umdrehungen pro Minute und 25 Grad Celsius um. Anschließend wird der Nanoemulsionsimpfstoff, der ein Milligramm pro Milliliter Protein enthält, als klare, transparente Flüssigkeit erhalten.Für die Antigenbeschichtung des enzymgebundenen Immunosorbent-Assays oder der ELISA-Platte wird das HI-Proteinantigen mit einer Überzugslösung auf 10 Mikrogramm pro Milliliter verdünnt und 100 Mikroliter des verdünnten Antigens pro Vertiefung in die ELISA-Platte gegeben. Inkubieren Sie die Platte über Nacht bei vier Grad Celsius. Waschen Sie die Platte nach der Inkubation mit 300 Mikrolitern PBST pro Well.Versiegeln Sie die Vertiefungen mit 300 Mikrolitern der Versiegelungslösung pro Vertiefung für zwei Stunden bei 37 Grad Celsius, verdünnen Sie dann drei Mikroliter des Mäuseserums aus jeder Versuchsgruppe 100 Mal durch Zugabe von 297 Mikrolitern PBST und wirbeln Sie das verdünnte Serum vor. Für jede Serumprobe werden 100 Mikroliter verdünntes Serum pro Well in die beschichtete ELISA-Platte gegeben. In ähnlicher Weise verdünnen Sie die Positiv- und Negativkontrollseren 100 Mal mit PBST, indem Sie 396 Mikroliter PBST vier Mikroliter Positiv- oder Negativkontrollserum hinzufügen, gefolgt von einer Vortexmischung.Um eine Verdünnung mit doppeltem Verhältnis durchzuführen, geben Sie 200 Mikroliter des zuvor verdünnten Versuchsserums in die erste Reihe der beschichteten ELISA-Platte und geben Sie dann 100 Mikroliter PBST in alle Vertiefungen mit Ausnahme der Vertiefungen in den Reihen eins und 12. Stellen Sie dann die Pipettenpistole auf 100 Mikroliter ein und übertragen Sie 100 Mikroliter Serum aus der ersten Reihe in die zweite Reihe mit PBST. Mischen Sie den Inhalt in der zweiten Reihe 10 Mal mit der Pipettenpistole.In ähnlicher Weise führen Sie eine bis zwei aufeinanderfolgende Verdünnungen des Serums bis Zeile 11 durch. Wenn Sie fertig sind, entsorgen Sie 100 Mikroliter Flüssigkeit aus Reihe 11. Geben Sie 100 Mikroliter verdünntes negatives und positives Serum in jede entsprechende Vertiefung in Zeile 12 nach der Probenvorlage.Die ELISA-Platte mit Frischhaltefolie verschließen und eine Stunde lang bei 37 Grad Celsius inkubieren. In der Zwischenzeit wird der sekundäre Antikörper Anti-Maus-IgG HRP 10.000 Mal durch Zugabe von PBST verdünnt. Waschen Sie nach der einstündigen Inkubation die ELISA-Platte mit 300 Mikrolitern PBST pro Well, um die ungebundenen Antikörper zu reinigen, geben Sie dann 100 Mikroliter des verdünnten Sekundärantikörpers in jede Vertiefung und inkubieren Sie die Platte 40 Minuten lang bei 37 Grad Celsius.Waschen Sie die Platte mit 300 Mikrolitern PBST pro Vertiefung und geben Sie dann 100 Mikroliter TMB-Farbentwicklungslösung in jede Vertiefung. Stellen Sie die Teller bei 37 Grad Celsius und 10 Minuten lang vor Licht geschützt. Beenden Sie danach die Reaktion sofort mit 50 Mikrolitern zweimolarer Schwefelsäure.Bestimmen Sie die optische Dichte oder die OD-Werte bei 415 Nanometern mit einem Enzymmarker innerhalb von 15 Minuten nach der Terminierung. Die physikalischen Eigenschaften des neuartigen Nanoemulsionsimpfstoffs wurden mittels Transmissionselektronenmikroskopie und Rasterkraftmikroskopie analysiert. Mittels dynamischer Lichtstreuung wurden die Partikelgrößenverteilung und das Zetapotenzial der Probe bestimmt.SDS-PAGE und Western Blot zeigten, dass die Menge des Antigens im Niederschlag und Überstand nach der Zentrifugation nicht signifikant abnahm, was darauf hindeutet, dass der Impfstoff strukturell intakt, spezifisch und immunogen war. Der Nanoemulsions-Adjuvans-Impfstoff erhöhte signifikant die Gesamttiter der IgG-, IgG1- und IgG2a-Antikörper der Mäuse. Die Immunogenität des Proteinimpfstoffs wurde signifikant verbessert.Die Serum-IgG2b-OD-Werte bei 450 Nanometern waren am höchsten, wenn PBS mit einer Verdünnung von eins zu 5.000 verwendet wurde. Das letale Modell von MRSA-252-Mäusen zeigte, dass der Nanoemulsionsimpfstoff eine gute Schutzwirkung zeigte und die bakterielle Infektion mit MRS A 252 wirksam hemmte, wodurch die Überlebensrate infizierter Mäuse verbessert wurde. Die Herstellung der Nanoemulsion und die Zugabe von Serum zur Pflanze sind die wichtigsten.Auch beim Nachweis des Antikörpertiters sollte auf eine breitere und flüssigkeitsbasierte Ausscheidung geachtet werden. Neben Adjuvantien kann diese Methode auch für die Immunevolution anderer mathematischer Arrays, wie z.B. Kunststoffe oder Verabreichungssysteme, eingesetzt werden.
