アルミニウムアジュバントは長い間使用されており、アジュバント補助メカニズムは比較的明確で、安全で、効果的で、忍容性が良好です4。アルミニウムアジュバントは、現在広く使用されているタイプのアジュバントである。一般に、アルミニウム塩粒子に吸着された抗原は、安定性を改善し、注射部位が抗原を取り込む能力を高め、良好な吸収と徐放を提供すると考えられています5。現在、アルミニウムアジュバントの主な欠点は、アジュバント効果を欠くか、またはいくつかのワクチン候補抗原に対して弱いアジュバント効果しか示さないことである6。さらに、アルミニウムアジュバントはIgEを介した過敏反応を誘導します5。したがって、より強力な免疫応答を刺激するための新規アジュバントを開発する必要があります。

実証されたプロトコルは、直接的かつ効果的な実験アプローチと、新規ウイルスアジュバントの免疫応答効果に対する詳細なステップを強調しました。このプロトコルは、アジュバントワクチンの基本的な免疫評価法を研究に提供するだけでなく、それらの質量安定性を決定することもできます。はじめに、MRSA HI抗原タンパク質の1ミリリットル当たり10ミリグラムの作業溶液を取り、それに、3つの賦形剤、ミリスチン酸イソプロピル、ポリオキシエチレンヒマシ油、及びプロピレングリコールを順次添加し、質量比を1〜6〜3に維持する。混合物をよくかき混ぜてから、滅菌した純水を徐々に加えて、システム容量が10ミリリットルになるまでします。低エネルギー乳化法によってナノエマルジョンワクチンを調製するには、毎分500回転、摂氏25度で約20分間攪拌する。続いて、1ミリリットル当たり1ミリグラムのタンパク質を含有するナノエマルジョンワクチンが透明な透明な液体として得られる。酵素結合免疫吸着アッセイまたはELISAプレートの抗原コーティングのために、コーティング溶液でHIタンパク質抗原を1ミリリットルあたり10マイクログラムに希釈し、ELISAプレートでウェルあたり100マイクロリットルの希釈抗原を追加します。プレートを摂氏4度で一晩インキュベートします。インキュベーション後、ウェルあたり300マイクロリットルのPBSTでプレートを洗浄します。ウェルを1ウェルあたり300マイクロリットルのシーリング溶液で37°Cで2時間密封し、次に297マイクロリットルのPBSTを加えて各実験群のマウス血清3マイクロリットルを100倍に希釈し、希釈した血清をボルテックスします。各血清サンプルについて、コーティングされたELISAプレートにウェルあたり100マイクロリットルの希釈血清を追加します。同様に、396マイクロリットルのPBSTに4マイクロリットルのポジティブまたはネガティブコントロール血清を添加し、続いてボルテックス混合することにより、ポジティブコントロールおよびネガティブコントロール血清をPBSTで100倍に希釈します。ダブルレシオ希釈を行うには、コーティングされたELISAプレートの最初の列に200マイクロリットルの以前に希釈した実験血清を追加し、次に1列目と12列目を除くすべてのウェルに100マイクロリットルのPBSTを追加します。次に、ピペットガンを100マイクロリットルに調整し、PBSTを含む1列目から2列目に100マイクロリットルの血清を移します。ピペットガンを使用して2列目の内容物を10回混合します。同様に、行11まで血清の希釈を1回〜2回連続して行う。完了したら、行11から100マイクロリットルの液体を廃棄します。100マイクロリットルの希釈陰性および陽性血清を、サンプルテンプレートに続く行12の対応する各ウェルに追加します。ELISAプレートをラップで密封し、摂氏37度で1時間インキュベートします。一方、二次抗体抗マウスIgG HRPをPBSTを加えて10, 000倍に希釈する。1時間のインキュベーション後、ELISAプレートをウェルあたり300マイクロリットルのPBSTで洗浄して未結合抗体を洗浄し、次に100マイクロリットルの希釈二次抗体を各ウェルに加え、プレートを摂氏37度で40分間インキュベートします。ウェルあたり300マイクロリットルのPBSTでプレートを洗浄し、次に各ウェルに100マイクロリットルのTMB発色溶液を追加します。プレートを摂氏37度に置き、光から10分間離します。その後、50マイクロリットルの2モル硫酸で直ちに反応を終了する。酵素マーカーを使用して415ナノメートルの光学濃度またはOD値を、終了後15分以内に検出します。新規ナノエマルジョンワクチンの物理的特性を透過型電子顕微鏡と原子間力顕微鏡を用いて解析した。動的光散乱を用いて、試料の粒度分布およびゼータ電位を決定した。SDS-PAGEおよびウェスタンブロッティングは、沈殿物および上清中の抗原量が遠心分離後に有意に遅延しないことを示し、ワクチンが構造的に無傷で特異的で免疫原性であることを示しています。ナノエマルジョンアジュバントワクチンは、マウスの総IgG、IgG1、およびIgG2a抗体価を有意に増加させた。タンパク質ワクチンの免疫原性は有意に改善された。450ナノメートルでの血清IgG2b OD値は、PBSを1〜5, 000希釈で使用した場合に最も高かった。MRSA 252マウスの致死モデルは、ナノエマルジョンワクチンが良好な防御効果を示し、MRS A 252による細菌感染を効果的に阻害し、感染したマウスの生存率を改善することを反映した。ナノ乳化の調製および植物への血清の添加が最も重要である。また、抗体価を検出するところは、より広く且つ液体ベースに補正するように注意すべきである。アジュバント以外にも、この方法は、プラスチックやデリバリーシステムなどの他の数学アレイの免疫進化にも使用できます。
