메티실린 내성 황색포도 상구균 (MRSA)은 전 세계 중환자실(ICU) 병동1, 심장내과 및 화상 부서에서 감염률이 가장 높은 기회 병원체 중 하나입니다. MRSA는 높은 감염률, 사망률 및 광범위한 약물 내성을 나타내어 임상 치료에 큰 어려움을 나타냅니다2. 2017년 세계보건기구(WHO)가 발표한 항생제 내성 박테리아의 글로벌 우선순위 목록에서 MRSA는 가장 중요한 범주3에 포함되었습니다. 따라서 MRSA 감염에 대한 백신이 시급히 필요합니다.
알루미늄 보조제는 오랫동안 사용되어 왔으며 보조제 보조 메커니즘은 비교적 명확하고 안전하며 효과적이며 내약성이 우수합니다4. 알루미늄 보조제는 현재 널리 사용되는 보조제 유형입니다. 일반적으로 알루미늄염 입자에 흡착된 항원은 안정성을 향상시키고 주사 부위의 항원 흡수 능력을 향상시켜 우수한 흡수와 느린 방출을 제공할 수 있다고 믿어진다5. 현재, 알루미늄 보조제의 주요 단점은 일부 백신 후보 항원에 대해 보조제 효과가 없거나 약한 보조제 효과만을 나타낸다는 것이다6. 또한, 알루미늄 보조제는 IgE 매개 과민 반응을 유도한다5. 따라서 더 강한 면역 반응을 자극하기 위해 새로운 보조제를 개발할 필요가 있습니다.
나노에멀젼 보조제는 오일, 물, 계면활성제 및 공계면활성제로 구성된 콜로이드 분산 시스템이다7. 또한, 보조제는 열역학적으로 안정하고 등방성이며, 고속 원심분리에 의해 오토클레이브 또는 안정화될 수 있고, 온화한 제조 조건 하에서 자발적으로 형성될 수 있다. 여러 에멀젼 보조제(예: MF59, NB001-002 시리즈, AS01-04 시리즈 등)가 현재 시판 중이거나 임상 연구 단계에 있지만 입자 크기는 160nm8보다 큽니다. 따라서 나노 스케일 (1-100 nm) 의약품의 장점 (즉, 큰 비 표면적, 작은 입자 크기, 표면 효과, 높은 표면 에너지, 작은 크기 효과 및 매크로 양자 터널링 효과)을 완전히 활용할 수 없습니다. 본 프로토콜에서, 직경 크기가 1-100 nm인 나노에멀젼 기술에 기초한 신규한 보조제는 양호한 보조제 활성을 나타내는 것으로 보고되었다9. 재조합 서브유닛 백신 항원 단백질 HI (α-hemolysin 돌연변이체[Hla] 및 Fe 이온 표면 결정 인자 B[IsdB] 서브유닛 N2 활성 단편 융합 단백질)를 시험하였다. 마우스 전신 감염 모델을 사용하여 물리적 특성과 안정성을 검사하고, 근육 투여 후 특정 항체 반응을 평가하고, 백신의 보호 효과를 테스트하기 위한 일련의 절차가 수립되었습니다.
