L. paraplantarum FT-259 is een Gram-positieve, staafvormige bacterie, is een producent van bacteriocines met antilisteriële activiteit en heeft een hoog probiotisch potentieel20. Son et al.24 toonden eerder de immunostimulerende en antioxiderende capaciteit van L. paraplantarum-stammen aan. Bovendien hebben ze een groot probiotisch potentieel, met eigenschappen zoals stabiliteit onder kunstmatige maag- en galomstandigheden, gevoeligheid voor antibiotica e...

Om als probiotica te worden gedefinieerd, moeten micro-organismen die aan voedingsmiddelen (of supplementen) worden toegevoegd, levend worden geconsumeerd, kunnen overleven tijdens passage in het maagdarmkanaal van de gastheer en de plaats van actie in voldoende hoeveelheden bereiken om gunstige effecten uit te oefenen 1,2,7.
De groeiende interesse in probiotica is te wijten aan de verschillende voordelen voor de menselijke gezondheid die ze bieden, zoals de stimulatie van het immuunsysteem, de verlaging van het serumcholesterolgehalte en de verbetering van de darmbarrièrefunctie door op te treden tegen schadelijke microben, evenals hun gunstige effecten bij de behandeling van het prikkelbare darmsyndroom, onder andere 2,3. Bovendien hebben verschillende studies aangetoond dat probiotica een positieve invloed kunnen hebben op andere delen van het menselijk lichaam waar onevenwichtige microbiële gemeenschappen infectieziekten kunnen veroorzaken 3,4,5.
Om probiotica therapeutisch effectief te laten zijn, moet het product tussen 10 6-107 CFU / g bacteriën bevatten op het moment van consumptie6. Aan de andere kant heeft het Italiaanse ministerie van Volksgezondheid en Volksgezondheid Canada vastgesteld dat het minimumniveau van probiotica in voedsel 109 CFU / g levensvatbare cellen per dag of per portie moet zijn, respectievelijk7. Aangezien hoge belastingen probiotica nodig zijn om te garanderen dat ze gunstige effecten zullen hebben, is het essentieel om hun overleving te garanderen tijdens verwerking, schapopslag en passage door het maagdarmkanaal (GI). Verschillende studies hebben aangetoond dat micro-inkapseling een effectieve methode is om de algehele levensvatbaarheid van probioticate verbeteren 8,9,10,11.
In deze context zijn verschillende methoden ontwikkeld voor de micro-inkapseling van probiotica, zoals sproeidrogen, vriesdrogen, sproeikoelen, emulsie, extrusie, coacervatie en, meer recent, gefluïdiseerde bedden11,12,13,14. Micro-inkapseling door sproeidrogen (SD) wordt veel gebruikt in de voedingsmiddelenindustrie omdat het een eenvoudig, snel en reproduceerbaar proces is. Het is eenvoudig op te schalen en heeft een hoge productieopbrengst bij lage energiebehoefte11,12,13,14. Niettemin kan de blootstelling aan hoge temperaturen en een laag vochtgehalte de overleving en levensvatbaarheid van de probiotische cellen beïnvloeden15. Beide parameters kunnen voor een bepaalde stam worden verbeterd door de effecten van de kweekleeftijd en -omstandigheden te bepalen om de cultuur vooraf aan te passen en de sproeidroogomstandigheden (inlaat- en uitlaattemperaturen, vernevelingsproces) en de inkapselingssamenstelling 8,14,16,17,18 te optimaliseren.
De samenstelling van de inkapselende oplossing is ook een belangrijke factor tijdens SD, omdat het het beschermingsniveau tegen ongunstige omgevingsomstandigheden kan bepalen. Inuline, Arabische gom, maltodextrines en magere melk worden veel gebruikt als inkapselingsmiddelen voor probiotische droging 5,17,18,19. Inuline is een fructooligosaccharide die een sterke prebiotische activiteit vertoont en de darmgezondheid bevordert19. Magere melk is zeer effectief in het handhaven van de levensvatbaarheid van gedroogde bacteriële cellen en genereert een poeder met goede reconstitutie-eigenschappen17.
Lactiplantibacillus paraplantarum FT-259 is een melkzuurbacterie die bacteriocine produceert en antilisteriale activiteit vertoont, naast probiotische eigenschappen20,21. Het is een facultatieve heterofermentatieve staafvormige Gram-positieve bacterie die groeit van 15 °C tot 37 °C20 en compatibel is met de homeostatische lichaamstemperatuur. Deze studie was bedoeld om alle stappen te presenteren die betrokken zijn bij de productie en fysisch-chemische karakterisering van een gesproeidroogd probioticum (L. paraplantarum FT-259) en de invloed van de beschermstoffen en droogtemperaturen te evalueren.

Probiotica en prebiotica zijn van groot belang voor de voedings- en farmaceutische industrie vanwege hun gezondheidsvoordelen. Probiotica zijn levende bacteriën die gunstige effecten kunnen hebben op het welzijn van mens en dier, terwijl prebiotica soorten voedingsstoffen zijn die de gunstige darmbacteriën voeden. Poeder probiotica hebben aan populariteit gewonnen vanwege het gemak en de bruikbaarheid van hun inname en opname in het dieet als voedingssupplement. Het droogproces interfereert echter met de levensvatbaarheid van cellen, omdat hoge temperaturen probiotische bacteriën inactiveren. In deze context was deze studie gericht op het presenteren van alle stappen die betrokken zijn bij de productie en fysisch-chemische karakterisering van een gesproeidroogd probioticum en het evalueren van de invloed van de beschermende stoffen (gesimuleerde magere melk en inuline: maltodextrine associatie) en droogtemperaturen bij het verhogen van de poederopbrengst en de levensvatbaarheid van de cel. De resultaten toonden aan dat de gesimuleerde magere melk een hogere probiotische levensvatbaarheid bevorderde bij 80 °C. Met dit beschermmiddel verminderen de probiotische levensvatbaarheid, het vochtgehalte en de wateractiviteit (Aw) zolang de inlaattemperatuur toeneemt. De levensvatbaarheid van de probiotica neemt omgekeerd af met de droogtemperatuur. Bij temperaturen dicht bij 120 °C vertoonde het gedroogde probioticum levensvatbaarheid rond 90%, een vochtgehalte van 4,6% w / w en een Aw van 0,26; waarden die toereikend zijn om de stabiliteit van het product te garanderen. In dit verband zijn sproeidroogtemperaturen van meer dan 120 °C vereist om de levensvatbaarheid en houdbaarheid van de microbiële cellen in de poedervormige bereiding en overleving tijdens de verwerking en opslag van levensmiddelen te waarborgen.

In ons laboratorium doen we onderzoek naar nuttige microben, waaronder probiotica, die levende micro-organismen zijn die aan voedsel kunnen worden toegevoegd om de gezondheid van consumenten te verbeteren, meestal door te concurreren met pathogene bacteriën in de darm om ze uit te sluiten en symptomen te verlichten, zoals diarree of andere gastro-intestinale stoornissen. Tegenwoordig zijn er veel technieken toegepast om te garanderen dat de probiotica tijdens de houdbaarheid in voedsel zullen overleven. En ook moesten ze de barre omstandigheden van het maagdarmkanaal tolereren om hun gunstige effecten in de darm uit te oefenen.We hebben veel technologieën bestudeerd die de overleving van probiotica kunnen verbeteren, hetzij in voedsel of in de menselijke gastheer. Onder de technieken is micro-inkapseling door sproeidrogen van groot belang omdat het eenvoudig, snel en reproduceerbaar is en hoge opbrengsten biedt bij lage energiebehoeften. Het is echter een uitdaging om de juiste omstandigheden te bepalen om gedroogde probiotica te verkrijgen.En daarvoor moeten geschikte beschermmiddelen worden gekozen. Magere melk, maltodextrine, of andere soortgelijke probiotische inuline zijn gebruikt met verschillende probiotische stammen. Het gebied van micro-inkapseling is een gelukkig evoluerend onderzoeksgebied dat het potentieel heeft om de voedingsindustrie te beïnvloeden door nieuwe producten te leveren die als functionele ingrediënten kunnen worden gebruikt.In deze studie laten we zien hoe een micro-ingekapseld preparaat van een potentiële probiotische cultuur kan worden verkregen met behulp van de sproeidroogtechniek. We laten ook zien hoe de belangrijkste parameters te bepalen om het poeder te karakteriseren, te verkrijgen en hoe de overlevingskans van het probioticum te evalueren.
