Vi udvikler en vævs-GI-model af blod-hjerne-barrieren for at forstå virkningen af sepsis på hjernen og bidraget fra blodbårne faktorer. Brug af menneskeskabte pluripotente stamceller. Vi planlægger at evaluere genetiske faktorers bidrag til visse patientpopulationers modtagelighed.
Nuværende eksperimentelle udfordringer inden for vævschipsamfundet er reproducerbarheden af resultater og vedtagelsen af teknikker af eksterne laboratorier. Unikke vævschipdesign, små mængder inden for vævschips og mangel på standard funktionelle assays bidrager alle til disse udfordringer og begrænser ofte brugerne til fluorescerende baserede assays som deres primære output. Micro simm-enheder tilbyder modularitet til alsidig in vitro-modellering, en glaslysoverflade, der er ideel til livsstilsbilleddannelse med høj opløsning og ultrasyn 100 nanometer, højt produceret nanomembran, der sikrer sømløs opløselig faktorkrydstale og præcise permeabilitetsassays.
Med mikrosimmen har vi set, at specifikke celletyper har tendens til at producere flere af matrixproteinerne, der giver et støttende stillads omkring hjernens blodkar. Nu kan vi spørge, om inflammation får de samme celler til at styrke eller nedbryde deres matricer og derved ændre barriereintegriteten. I fremtiden vil vi tilføje endnu et rum til modellen med grønne celler som astrocytter og mikroglia for at studere, hvordan disse celler interagerer med blod-hjerne-barrieren i tilfælde af systemisk inflammation som sepsis.
Vi planlægger også at undersøge yderligere årsager til systemisk inflammation som postoperativt delirium, overlejret demens.
