Vi utvikler en vevs-GI-modell av blod-hjernebarrieren for å forstå virkningen av sepsis på hjernen og bidraget fra blodbårne faktorer. Bruk av menneskeskapte pluripotente stamceller. Vi planlegger å evaluere bidraget fra genetiske faktorer til følsomheten til visse pasientpopulasjoner.
Nåværende eksperimentelle utfordringer innen vevschip-samfunnet er reproduserbarheten av resultater og adopsjon av teknikker av eksterne laboratorier. Unike vevsbrikkedesign, små volumer i vevsbrikker og mangel på standard funksjonelle analyser bidrar alle til disse utfordringene og begrenser ofte brukere til fluorescerende baserte analyser som deres primære utgang. Micro simm-enheter tilbyr modularitet for allsidig in vitro-modellering, en glassoverflate som er ideell for livsstilsbilder med høy oppløsning og ultra syn 100 nanometer, høyt produsert nanomembran, noe som sikrer sømløs løselig faktor crosstalk og presise permeabilitetsanalyser.
Med micro simm har vi sett at spesifikke celletyper har en tendens til å produsere flere av matriksproteinene som gir et støttestillas rundt hjernens blodkar. Nå kan vi spørre om betennelse får de samme cellene til å styrke eller bryte ned matrisene sine, og dermed endre barriereintegriteten. I fremtiden vil vi legge til et annet rom i modellen med grønne blodlegemer som astrocytter og mikroglia for å studere hvordan disse cellene samhandler med blod-hjernebarrieren i tilfeller av systemisk betennelse som sepsis.
Vi planlegger også å undersøke flere årsaker til systemisk betennelse som postoperativt delirium, lagt oppå demens.
