Vi utvecklar en vävnads-GI-modell av blod-hjärnbarriären för att förstå effekterna av sepsis på hjärnan och bidraget från blodburna faktorer. Användning av humaninducerade pluripotenta stamceller. Vi planerar att utvärdera genetiska faktorers bidrag till mottagligheten hos vissa patientpopulationer.
Aktuella experimentella utmaningar inom vävnadschipgemenskapen är reproducerbarheten av resultat och antagandet av tekniker av externa laboratorier. Unika vävnadschipdesigner, små volymer inom vävnadschip och brist på standardiserade funktionella analyser bidrar alla till dessa utmaningar och begränsar ofta användarna till fluorescensbaserade analyser som sin primära produktion. Micro simm-enheter erbjuder modularitet för mångsidig in vitro-modellering, en ljusyta av glas som är idealisk för högupplöst livsstilsavbildning och ultra syn 100 nanometer, högproducerat nanomembran, vilket säkerställer sömlös överhörning av lösliga faktorer och exakta permeabilitetsanalyser.
Med mikrosimmen har vi sett att specifika celltyper tenderar att producera mer av de matrixproteiner som utgör en stödställning runt hjärnans blodkärl. Nu kan vi fråga oss om inflammation får samma celler att stärka eller bryta ner sina matriser och därmed förändra barriärens integritet. I framtiden kommer vi att lägga till ytterligare ett fack i modellen med gröna celler som astrocyter och mikroglia för att studera hur dessa celler interagerar med blod-hjärnbarriären vid systemisk inflammation som sepsis.
Vi planerar också att undersöka ytterligare orsaker till systemisk inflammation som postoperativt delirium, överlagrat på demens.
