Hier wird ein einfaches und effektives Mittel zur Herstellung von ICV-Injektionen bei Mäusen beschrieben. Da diese Technik keinen stereotaktischen Rahmen benötigt, ist dieser Ansatz für die zentrale Verabreichung von Medikamenten und experimentellen Wirkstoffen für mehr Forscher zugänglich. Die Abgabe von experimentellen Wirkstoffen wie Medikamenten1, Viren2 oder Zellen3 an das Gehirn ist für die neuroendokrine Forschung oft notwendig. Wenn der Wirkstoff die Blut-Hirn-Schranke nicht ohne weiteres überwindet oder das experimentelle Ziel darin besteht, die zentralen Wirkungen des Wirkstoffs gezielt zu testen, ist es wichtig, eine zuverlässige Methode für die Verabreichung von Injektionen in das Gehirn zu haben. Darüber hinaus bietet die Injektion in den intrazerebroventrikulären Raum (ICV) die Möglichkeit, den Wirkstoff weit im Gehirn zu verteilen und bietet eine große Zielfläche, wodurch die Wahrscheinlichkeit einer erfolgreichen Injektion erhöhtwird 2. 
Eine gängige Methode zur Herstellung von ICV-Injektionen ist das Einsetzen einer permanenten Verweilkanüle. Bei diesem Ansatz ist ein stereotaktischer Rahmen notwendig, um die handelsübliche oder individuell angefertigte Kanüle zu positionieren, da die Kanüle geklebt oder zementiert wird. Oft wird nach der Genesung eine supraphysiologische Dosis Angiotensin II über die Kanüle verabreicht, und wenn das Trinkverhalten sofort beobachtet wird, gilt die Kanüle als korrekt platziert4. Dieser Ansatz hat viele Vorteile, darunter die Möglichkeit, eine Langzeitinfusion durchzuführen und dasselbe Tier mehrmals zu injizieren. Darüber hinaus kann bei Verwendung von Angiotensin II die korrekte Platzierung vor der Verabreichung von experimentellen Verbindungen bestätigt werden. Es gibt jedoch einige Einschränkungen bei der Platzierung einer permanenten Kanüle, einschließlich der Notwendigkeit einer teuren Ausrüstung (stereotaktischer Rahmen), der Möglichkeit einer Beschädigung der Kanüle nach dem Einsetzen (z. B. können Mäuse auf der Kanüle eines Käfiggefährten kauen) und der Möglichkeit von Infektionen um die permanente Kanüle herum. Einzelne ICV-Injektionen können unter Verwendung eines stereotaktischen Rahmens3 durchgeführt werden, der zwar wirksam ist, jedoch eine erhebliche Exposition gegenüber Anästhesie erfordert und daher einige akute physiologische und verhaltensbezogene Wirkungen der Behandlung verschleiern kann. Darüber hinaus erfordert die Platzierung von Mäusen in einem stereotaktischen Rahmen ein erhebliches Training, um eine stabile Platzierung zu erreichen und das Reißen der Gehörgänge zu verhindern.
Hier wird eine etablierte Methode zur Herstellung von Freihandinjektionen bei Mäusen beschrieben. Diese Methode basiert auf früheren Berichten 5,6. Die Vorteile dieser Technik sind, dass sie einfach und schnell ist und keine spezielle Ausrüstung wie einen stereotaktischen Rahmen erfordert. Wie unten beschrieben, beinhaltet dieses Verfahren die Manipulation einer Glasspritze in Bezug auf Landmarken auf dem Mauskopf, um die Injektionen durchzuführen, was schnell durchgeführt werden kann und daher nur wenige Minuten Gasanästhesie am Versuchstag erfordert.

free-hand intracerebroventricular injections in mice

Die Untersuchung neuroendokriner Systeme erfordert oft die Verabreichung von Medikamenten, Viren oder anderen experimentellen Wirkstoffen direkt in das Gehirn von Mäusen. Eine intrazerebroventrikuläre (ICV) Injektion ermöglicht die großflächige Verabreichung des experimentellen Wirkstoffs im gesamten Gehirn (insbesondere in den Strukturen in der Nähe der Ventrikel). Hier werden Methoden zur Freihand-ICV-Injektion bei erwachsenen Mäusen beschrieben. Durch visuelle und taktile Landmarken auf den Köpfen von Mäusen können Injektionen in die Seitenventrikel schnell und zuverlässig durchgeführt werden. Die Injektionen werden mit einer Glasspritze durchgeführt, die in der Hand des Versuchsleiters gehalten und in ungefährem Abstand zu den Landmarken platziert wird. Somit benötigt diese Technik keinen stereotaktischen Rahmen. Darüber hinaus erfordert diese Technik nur eine kurze Isofluran-Anästhesie, die eine anschließende Beurteilung des Verhaltens und/oder der Physiologie von Mäusen bei wachen, sich frei verhaltenden Mäusen ermöglicht. Die Freihand-ICV-Injektion ist ein leistungsfähiges Werkzeug für die effiziente Verabreichung experimenteller Wirkstoffe in das Gehirn lebender Mäuse und kann mit anderen Techniken wie häufigen Blutentnahmen, neuronalen Schaltkreismanipulationen oder In-vivo-Aufzeichnungen kombiniert werden, um neuroendokrine Prozesse zu untersuchen.

The delivery of experimental agents, such as drugs1, viruses2, or cells3, to the brain is often necessary for neuroendocrine research. If the agent does not readily cross the blood-brain barrier or the experimental objective is to specifically test the central effects of the agent, it is important to have a reliable method for delivering injections into the brain. Moreover, injection into the intracerebroventricular (ICV) space provides the opportunity to distribute the agent widely in the brain and provides a large target area, thus increasing the likelihood of successful injection2. 
A common method for making ICV injections involves the placement of a permanent indwelling cannula. In this approach, a stereotaxic frame is necessary to position the commercially available or custom-made cannula, as the cannula is glued or cemented in place. Often, upon recovery, a supraphysiological dose of angiotensin II is administered through the cannula, and if drinking behavior is immediately observed, then the cannula is considered correctly placed4. This approach has many advantages, including the ability to perform long-term infusion and the ability to inject the same animal multiple times; additionally, if angiotensin II is employed, correct placement can be confirmed prior to the administration of experimental compounds. However, there are some limitations to placing a permanent cannula, including the requirement for expensive equipment (stereotaxic frame), the possibility of damage to the cannula after placement (e.g., mice can chew on the cannula of a cage mate), and the possibility of infections around the permanent cannula. Single ICV injections can be made with the use of a stereotaxic frame3, which, although effective, requires substantial exposure to anesthesia and, thus, may obscure some acute physiological and behavioral effects of the treatment. Additionally, the placement of mice in a stereotaxic frame requires substantial training to achieve stable placement and prevent the rupturing of the ear canals.
Here, an established method for making free-hand injections in mice is described. This method is based on previous reports5,6. The advantages of this technique are that it is simple, rapid, and does not require specialized equipment such as a stereotaxic frame. As described below, this procedure involves manipulating a glass syringe relative to landmarks on the mouse head to make the injections, which can be done rapidly and, thus, requires only a few minutes of gas anesthesia on the experimental day.
