CNS er et komplekst netværk, der spænder fra hjernen ned til rygmarven og består af mange celletyper, overvejende neuroner, oligodendrocytter, astrocytter og microglia1. Da hver celle har en vigtig rolle i at opretholde homeostase og generere passende svar på udfordringer i CNS 9,10,11, er et dyrkningssystem, der indeholder alle disse celletyper, et nyttigt og alsidigt værktøj til at undersøge, hvordan hjerne...

Forbedring af vores forståelse af centralnervesystemet (CNS) er afgørende for at forbedre terapeutiske muligheder for mange neuroinflammatoriske og neurodegenerative sygdomme. CNS, et komplekst netværk af sammenkoblede celler i hjernen, rygmarven og synsnerverne, omfatter neuroner, oligodendrocytter, astrocytter og deres medfødte immunceller, microglia1. En in vitro-tilgang kan ofte drastisk reducere antallet af mus, der kræves for at udføre meningsfuld forskning; CNS's komplekse karakter gør det imidlertid umuligt at rekapitulere in vivo-situationen ved hjælp af cellelinjer. Blandede neurale cellekulturer giver et ekstremt værdifuldt forskningsværktøj til at undersøge neuro(immun)logiske spørgsmål i en relevant model i overensstemmelse med principperne om udskiftning, reduktion og forfining (3R) 2,3.
Thomson et al. beskrev en cellekulturmetode ved hjælp af prænatale rygmarvsceller, der differentierer sig til alle de førnævnte vigtigste CNS-celletyper4. Dette system har også synapsedannelse, myelinerede axoner og knudepunkter af Ranvier. Hovedbegrænsningen ved denne dyrkningsmetode er, at den ikke med fordel modellerer hjernen, da den er rygmarv, og celleudbyttet fra embryonal dag 13 (E13) rygmarv trækker sig sammen. Dette begrænser således antallet af eksperimentelle forhold, der kan undersøges. Derfor havde denne undersøgelse til formål at udvikle et nyt cellekultursystem, der rekapitulerer hjernens egenskaber med øget celleudbytte for at reducere kravene til dyr.
Med udgangspunkt i Thomson et al. udviklede vi en cellekulturmodel, der udelukkende stammer fra prænatale musehjerner. Disse kulturer har de samme cellepopulationer, sammenkobling og behandlingsmuligheder som rygmarvskulturerne, bortset fra at der er mindre myelinering ved sammenligning. Imidlertid er det mere effektivt at have en CNS in vitro-model med et cirka tre gange højere celleudbytte, hvilket kræver færre mus og mindre tid til behandling af embryoner. Vi optimerede dette unikke dyrkningssystem til flere downstream-applikationer og vægte, herunder brug af glasdæksler til mikroskopianalyse og forskellige størrelser af plastbrøndplader, herunder 96-brøndsplader til forskning med høj kapacitet.

Modeller af centralnervesystemet (CNS) skal rekapitulere det komplekse netværk af sammenkoblede celler, der findes in vivo. CNS består primært af neuroner, astrocytter, oligodendrocytter og mikroglia. På grund af den stigende indsats for at erstatte og reducere brugen af dyr er der udviklet en række in vitro-cellekultursystemer for at udforske medfødte celleegenskaber, som muliggør udvikling af lægemidler til CNS-infektioner og patologier. Mens visse forskningsspørgsmål kan løses af menneskebaserede cellekultursystemer, såsom (inducerede) pluripotente stamceller, har arbejdet med humane celler sine egne begrænsninger med hensyn til tilgængelighed, omkostninger og etik. Her beskriver vi en unik protokol til isolering og dyrkning af celler fra embryonale musehjerner. De resulterende blandede neurale cellekulturer efterligner flere cellepopulationer og interaktioner, der findes i hjernen in vivo. Sammenlignet med de nuværende ækvivalente metoder efterligner denne protokol hjernens egenskaber og samler også flere celler, hvilket gør det muligt at undersøge flere eksperimentelle tilstande fra en gravid mus. Desuden er protokollen relativt let og meget reproducerbar. Disse kulturer er optimeret til brug i forskellige skalaer, herunder 96-brøndbaserede skærme med høj kapacitet, 24-brønds mikroskopianalyse og 6-brøndkulturer til flowcytometri og revers transkriptionskvantitativ polymerasekædereaktion (RT-qPCR) analyse. Denne kulturmetode er et kraftfuldt værktøj til at undersøge infektion og immunitet inden for rammerne af nogle af kompleksiteten af CNS med bekvemmeligheden ved in vitro-metoder .

Det ultimative forskningsspørgsmål i mit projekt er at finde ud af, hvordan vi kan manipulere de medfødte immunresponser i centralnervesystemet for at beskytte mod virusinfektioner. I øjeblikket undersøger vi deres reaktion på det dybt antivirale protein interferon beta. En eksperimentel udfordring er at generere nok celler til at muliggøre detaljeret undersøgelse.Hvert resultat skal valideres ved hjælp af flere teknikker, og det kan desværre resultere i, at mange dyr aflives. På grund af det store antal celler, der genereres af denne nye kulturteknik, kan mange forskellige eksperimentelle forhold undersøges fra en gravid mus. Derudover reducerer brugen af in vitro-alternativer, hvor det er muligt, også i høj grad dyrenes lidelser.Denne metode reducerer og forfiner dyrebrug og følger nøje principperne i de tre R'er.Denne kulturmetode kan bruges til at løse mange forskellige forskningsspørgsmål ved hjælp af en række udlæsningsmetoder såsom ELISA, qPCR, mikroskopi og flowcytometri. Denne teknik kan hjælpe andre forskere med at reducere og forfine deres dyrebrug. Vi håber at forbedre vores forståelse af det medfødte antivirale respons ved at modulere det for at beskytte mod opportunistiske vira såsom humant polyomavirus 2 eller John Cunningham-virus, som det er almindeligt kendt, det primære middel til PML, som kan være en alvorlig bivirkning af immunsuppressive lægemidler, der anvendes til behandling af MS og andre sygdomme.
