Hoewel het gebruik van een wildtype virus de werkelijke infectie simuleert, zijn lentivirale PV's een veiligere optie om de mechanismen te bestuderen die verband houden met het binnendringen en infecteren van virussen zonder de strikte veiligheidseisen die nodig zijn om met pathogene virussen te werken 4,20,21. PV's zijn samengesteld uit een replicatie-defecte virale kern omgeven door het glycoproteïne van een pathogeen virus, ...

Pseudogetypeerde virussen (PV's) zijn moleculaire hulpmiddelen die in de microbiologie worden gebruikt om interacties tussen gastheer en virus en pathogeen-pathogeen te bestuderen 1,2,3,4. PV's bestaan uit een binnenste deel, de virale kern die het virale genoom beschermt, en een buitenste deel, de envelopglycoproteïnen op het oppervlak van het virus dat het tropisme definieert5. Een pseudovirus is replicatie-incompetent in de doelcel omdat het niet alle genetische informatie bevat om nieuwe virusdeeltjes te genereren. Deze combinatie van bijzondere eigenschappen maakt PV's tot een veilig alternatief voor een wildtype virus. Wildtype virussen daarentegen zijn zeer pathogeen en kunnen niet worden gebruikt in BSL 2-laboratoria voor analyse6.
De besmettelijkheid van PV's kan worden gecontroleerd door een reporter-gen, dat meestal codeert voor een fluorescerend eiwit (GFP, RFP, YFP) of een enzym dat chemiluminescente producten produceert (luciferase). Dit zit in een van de plasmiden die worden gebruikt voor PV-productie en is opgenomen in het genoom van het pseudovirus7.
Er bestaan momenteel verschillende soorten PV-kernen, waaronder lentivirale deeltjes op basis van het HIV-1-genoom. Het grote voordeel van op HIV-1 gebaseerde PV's ten opzichte van andere platforms is hun intrinsieke integratieproces in het genoom van de doelcel8. Hoewel HIV-1 een zeer besmettelijk virus is en de veroorzaker van aids is, zijn deze lentivirale vectoren veilig te gebruiken vanwege de uitgebreide optimalisatiestappen door de jaren heen. Optimale veiligheidsomstandigheden werden bereikt met de introductie van lentivirale vectoren van de 2e generatie, waarin virale genen werden uitgeput zonder de transductiemogelijkheden te beïnvloeden9. De3e en 4egeneratie droegen bij aan de verhoogde veiligheid van de behandeling van lentivirale vectoren met de verdere splitsing van het virale genoom in afzonderlijke plasmiden10,11. De nieuwste generaties PV's worden over het algemeen gebruikt om lentivirale vectoren voor gentherapie te produceren.
PV's kunnen worden gebruikt om interacties tussen virussen en gastheercellen te bestuderen, zowel tijdens de productie- als de infectiefase. PV's worden vooral gebruikt in pseudovirusneutralisatietests (PVNA). PVNA's worden op grote schaal gevalideerd om het neutralisatiepotentieel van serum of plasma te beoordelen door zich te richten op het virale glycoproteïne op de PV-envelop12,13. Neutralisatieactiviteit, uitgedrukt als de remmende concentratie 50 (IC50), wordt gedefinieerd als de verdunning van serum/plasma die 50% van de toegang tot virale deeltjes blokkeert14. In dit protocol beschreven we de opzet van een PVNA om de antilichaamactiviteit tegen Severe Acute Respiratory Syndrome - Coronavirus 2 (SARS-CoV-2) te testen in sera die zijn verzameld voor en na ontvangst van een boostervaccindosis.

pseudotyped viruses as a molecular tool to monitor humoral immune responses against sars-cov-2 via neutralization assay

Pseudogetypeerde virussen (PV's) zijn moleculaire hulpmiddelen die kunnen worden gebruikt om interacties tussen gastheer en virus te bestuderen en om het neutraliserende vermogen van serummonsters te testen, naast hun bekendere gebruik in gentherapie voor de afgifte van een interessant gen. PV's zijn replicatiegebrekkig omdat het virale genoom is verdeeld in verschillende plasmiden die niet in de PV's zijn opgenomen. Dit veilige en veelzijdige systeem maakt het gebruik van PV's in laboratoria van bioveiligheidsniveau 2 mogelijk. Hier presenteren we een algemene methodologie om lentivirale PV's te produceren op basis van drie plasmiden zoals hier vermeld: (1) het ruggengraatplasmide dat het reporter-gen draagt dat nodig is om de infectie te monitoren; (2) het verpakkingsplasmide dat de genen bevat voor alle structurele eiwitten die nodig zijn om de PV's te genereren; (3) het glycoproteïne-expressieplasmide aan het oppervlak van het omhulsel dat het virustropisme bepaalt en het binnendringen van virussen in de gastheercel bemiddelt. In dit werk is SARS-CoV-2 Spike het envelopglycoproteïne dat wordt gebruikt voor de productie van niet-replicatieve SARS-CoV-2 pseudogetypeerde lentivirussen.
In het kort werden verpakkingscellen (HEK293T) met behulp van standaardmethoden samen met de drie verschillende plasmiden getransfecteerd. Na 48 uur werd het supernatans met de PV's geoogst, gefilterd en opgeslagen bij -80 °C. De besmettelijkheid van SARS-CoV-2 PV's werd getest door de expressie van het reporter-gen (luciferase) in een doelcellijn 48 uur na infectie te bestuderen. Hoe hoger de waarde voor relatieve luminescentie-eenheden (RLU's), hoe hoger de infectie-/transductiesnelheid. Bovendien werden de infectieuze PV's toegevoegd aan de serieel verdunde serummonsters om het neutralisatieproces van het binnendringen van pseudovirussen in doelcellen te bestuderen, gemeten als de vermindering van de RLU-intensiteit: lagere waarden die overeenkomen met een hoge neutraliserende activiteit.

Gepseudotypeerde virussen zijn moleculaire hulpmiddelen die kunnen worden gebruikt om interacties tussen gastheer en virus te bestuderen en om neutraliserende activiteit van serummonsters te testen. In het werk dat we vandaag presenteren, zal een algemene methodologie worden gepresenteerd om pseudogetypeerde virussen te produceren, gebaseerd op vrij plasmide. Voor neutralisatietests worden over het algemeen Y-type virussen gebruikt, maar ze zijn zeer pathogeen en kunnen niet worden behandeld in een BSL-2-laboratorium, waardoor dit systeem puur op grote schaal kan worden toegepast.De technologie van pseudovirussen kan een veiliger alternatief zijn in vergelijking met levend virus om de interactie tussen ziekteverwekker en gastheer mogelijk te maken en de neutraliserende activiteit van antilichamen te testen. Toekomstige experimenten zullen de rol van specifieke antilichamen onderzoeken bij het behoud van immuniteit na vaccinatie met behulp van de technologie van pseudovirussen.
