Las células estromales mesenquimales (MSC) son células madre pluripotentes adultas derivadas delmesodermo 1. Las MSC están presentes en casi todos los tejidos conectivos2. Desde que las MSC fueron descubiertas por primera vez en la década de 1970 y aisladas con éxito de la médula ósea en 1987 por Friedenstein et al.3,4,5, se ha utilizado una variedad de tejidos somáticos humanos (incluidos fetales y adultos) para aislar MSC como hueso, cartílago, tendón, músculo, tejido adiposo y estroma de soporte hematopoyético 1,2,6,7. Las MSC demuestran una alta capacidad proliferativa y plasticidad para diferenciarse en muchos linajes de células somáticas y podrían migrar a tejidos lesionados e inflamados 2,8,9. Estas propiedades hacen de las MSC un candidato potencial para la medicina regenerativa10. Sin embargo, las MSC derivadas de tejido somático (st-MSC) están restringidas por la donación limitada, la capacidad proliferativa celular limitada, las variaciones de calidad y la preocupación por la bioseguridad para la posible transmisión de patógenos, si los hubiera, de los donantes11,12.
Las células madre pluripotentes inducidas humanas (hiPSC) se derivan de la reprogramación de células adultas con factores de transcripción (Oct4, Sox2, Klf4 y c-Myc), que tienen funciones similares a las de las células madre embrionarias13,14. Pueden autorrenovarse y poseer el potencial de diferenciarse en cualquier tipo de células somáticas, incluidas las MSC. En comparación con las st-MSC, las iPSC-MSC tienen la ventaja de un suministro ilimitado, menor costo, mayor pureza, conveniencia en el control de calidad, fácil producción a escala y modificación genética 15,16,17.
Debido a estas ventajas de las iPSC-MSC, se han reportado una variedad de métodos que impulsan las MSC a partir de iPSC. Estos métodos de diferenciación se han centrado en dos metodologías de cultivo: (1) la formación de cuerpos embrioides (EBs) y (2) el uso de cultivos monocapa 11,18,19,20. En este trabajo se caracterizó un enfoque representativo para cada una de las dos metodologías. Además, se accedió a comparaciones entre dos enfoques representativos basados en el tiempo, el costo, la capacidad proliferativa, la expresión de biomarcadores de MSC y la capacidad de diferenciación in vitro.

Las células estromales mesenquimales (MSC) son células madre pluripotentes adultas que se han utilizado ampliamente en medicina regenerativa. Dado que las MSC derivadas de tejidos somáticos están restringidas por la donación limitada, las variaciones de calidad y la bioseguridad, en los últimos 10 años se ha visto un gran aumento en los esfuerzos para generar MSC a partir de células madre pluripotentes inducidas humanas (hiPSC). Los esfuerzos pasados y recientes en la diferenciación de las hiPSC en MSC se han centrado en dos metodologías de cultivo: (1) la formación de cuerpos embrioides (EB) y (2) el uso del cultivo monocapa. Este protocolo describe estos dos métodos representativos para derivar MSC a partir de hiPSC. Cada método presenta sus ventajas y desventajas, incluyendo el tiempo, el costo, la capacidad de proliferación celular, la expresión de marcadores MSC y su capacidad de diferenciación in vitro. Este protocolo demuestra que ambos métodos pueden derivar MSC maduras y funcionales a partir de hiPSC. El método monocapa se caracteriza por un menor costo, una operación más simple y una diferenciación osteogénica más fácil, mientras que el método EB se caracteriza por un menor consumo de tiempo.

El alcance de nuestro estudio es un mecanismo y una estrategia terapéutica de investigación de enfermedades genéticas. Nuestra investigación se basa en modelos de enfermedades multinivel, como líneas celulares, iPSC, organoides y ratones. Las iPSC tienen una clara ventaja sobre otros modelos.Las iPSC de los pacientes y cualquier célula y organoide derivado posterior comparten el mismo genoma con los pacientes, lo que proporciona un banco de células de pacientes y modelos para muchas enfermedades. Además, con los métodos de las iPSC establecidas, los pacientes están bastante maduros. Quedan muchos desafíos en la dirección de diferenciar las iPSC en células y organoides específicos, con una comprensión cada vez más profunda de los mecanismos de desarrollo en el desarrollo actual que se producirá en el campo.
